Summary: This study utilized two-photon intravital imaging and single cell transcriptome analysis to evaluate the in vivo behavior of engrafted multipotent MSCs in an acute IC/BPS animal model. The results showed that MSCs gradually integrated into a perivascular-like structure and modulated key genes such as FOS and CDK1, influencing their engraftment, migration, and anti-inflammatory functions, thereby determining their therapeutic potency in vivo.

Summary: The aim of this study was to investigate the expression levels of sensory receptors, inflammatory proteins, and pro-apoptotic proteins in the urothelium of non-Hunner's interstitial cystitis (NHIC) bladders of patients with different clinical and cystoscopic phenotypes. The study concluded that IC/BPS patients with frequency and bladder pain complaints have higher levels of urothelial sensory receptors, and inflammatory and pro-apoptotic proteins. The expression levels of these sensory receptors, inflammatory proteins, and pro-apoptotic proteins are not significantly different among IC/BPS bladders with different conditions.

Summary: This study used 16S rRNA sequencing and untargeted metabolomics techniques to analyze the urinary microbial and metabolite profiles of IC/BPS patients and healthy controls. The results showed differential genera and metabolites associated with the inflammatory response of IC/BPS, including Lactobacillus as a protective bacterium and Sphingomonas as a pathogenic factor. The metabolite theophylline may also play a role in downregulating the inflammatory response of IC/BPS.

Summary: Interstitial cystitis/bladder pain syndrome (IC/BPS) is a chronic bladder disorder characterized by suprapubic pain and discomfort during bladder filling, accompanied by lower urinary tract symptoms. The diagnosis of IC/BPS is still not standardized, but cystoscopy and bladder biopsy can determine the presence of ulcers. The pathophysiology of IC/BPS may involve chronic inflammation, autoimmune disorders, and other factors. Biomarkers in urine and serum specimens could be useful for the diagnosis of IC/BPS.
