Activated platelet-rich plasma improves adipose-derived stem cell transplantation efficiency in injured articular cartilage

Introduction: Adipose-derived stem cells (ADSCs) have been isolated, expanded, and applied in the treatment of many diseases. ADSCs have also been used to treat injured articular cartilage. However, there is controversy regarding the treatment efficiency. We considered that ADSC transplantation with activated platelet-rich plasma (PRP) may improve injured articular cartilage compared with that of ADSC transplantation alone. In this study, we determined the role of PRP in ADSC transplantation to improve the treatment efficiency. Methods: ADSCs were isolated and expanded from human adipose tissue. PRP was collected and activated from human peripheral blood. The effects of PRP were evaluated in vitro and in ADSC transplantation in vivo. In vitro, the effects of PRP on ADSC proliferation, differentiation into chondrogenic cells, and inhibition of angiogenic factors were investigated at three concentrations of PRP (10%, 15% and 20%). In vivo, ADSCs pretreated with or without PRP were transplanted into murine models of injured articular cartilage. Results: PRP promoted ADSC proliferation and differentiation into chondrogenic cells that strongly expressed collagen II, Sox9 and aggrecan. Moreover, PRP inhibited expression of the angiogenic factor vascular endothelial growth factor. As a result, PRP-pretreated ADSCs improved healing of injured articular cartilage in murine models compared with that of untreated ADSCs. Conclusion: Pretreatment of ADSCs with PRP is a simple method to efficiently apply ADSCs in cartilage regeneration. This study provides an important step toward the use of autologous ADSCs in the treatment of injured articular cartilage.

The N-methyl, C-4 methoxy, and C-5 hydroxy groups in the acridone skeleton were suggested to contribute to its cytotoxic activity.

Optimization of the isolation procedure and culturing conditions for hepatic stellate cells obtained from mouse

Summary: This study investigated the optimal conditions for HSC isolation and culture to maintain the undifferentiated phenotype. Nycodenz at 9.6% was found to yield the best purity and quantity of HSCs. Serum-free DMEM supplemented with glucose, GLN, vitA, and insulin allowed for the perpetuation of HSCs in culture while maintaining their undifferentiated state.

Transplantation of mesenchymal stem cell sheet to treat acute hind-limb ischemia: A preclinical study

Summary: The transplantation of Fi-MSC sheets positively impacted acute limb ischemia mouse models by improving SpO2 levels and enhancing blood reperfusion. Compared to the control group, the treatment group displayed better blood reperfusion, neovascularization, and muscle tissue repair, suggesting the potential efficacy of Fi-MSC sheet transplantation in treating acute limb ischemia.

Breast cancer treatment by transplantations of dendritic cells and cytokine-induced killer cells: An update on clinical trials

Comparison of cytotoxic potency between freshly cultured and freshly thawed cytokine-induced killer cells from human umbilical cord blood

Roles of cancer stem cells in cancer immune surveillance

Summary: The interaction between cancer cells and the immune system has been extensively studied for over 130 years, but how to strengthen the immune system and suppress cancer cells remains unclear. Cancer stem cells, a subpopulation of cancer cells, have been found to play important roles in the initiation, progression, metastasis, and recurrence of various types of cancer. Despite the discovery of mechanisms by which cancer cells can escape immune surveillance, the specific roles of cancer stem cells in these mechanisms are still unknown. This review summarizes the mechanisms involving immune-related molecules in cancer cells and the roles of cancer stem cells in immune surveillance.

Brown adipocyte and browning thermogenesis: metabolic crosstalk beyond mitochondrial limits and physiological impacts

Summary: Brown adipocytes have the potential for therapeutic applications in reversing metabolic conditions such as obesity and diabetes. They can produce heat through thermogenesis, which promotes balanced energy expenditure and improves metabolic disorders. The regulation of thermogenesis involves not only thermogenin UCPs within the mitochondria, but also signaling molecules that affect nearby and distant tissues. This review highlights the importance of understanding the crosstalk between different organelles within brown adipocytes and the interorgan axes in regulating whole body metabolism, and suggests considering the individual characteristics of brown adipocytes in future clinical applications.

Angiogenic potential of hypoxia preconditioned human adipose and umbilical cord-derived mesenchymal stem cells: a comparative study

Summary: The study found that preconditioning with hypoxia significantly increased the secretion of bFGF, VEGF, and HGF by both AT-MSCs and UC-MSCs, along with a significant up-regulation of related gene expression. In a quail embryo model, normoxic UC-MSCs showed better angiogenic promotion than normoxic AT-MSCs, but there was no significant difference in the angiogenic potency of hypoxic AT-MSCs and UC-MSCs.
