Lipidomic dysregulation within the lung parenchyma following whole-thorax lung irradiation: Markers of injury, inflammation and fibrosis detected by MALDI-MSI

Radiation-induced lung injury (RILI) is a delayed effect of acute radiation exposure that can limit curative cancer treatment therapies and cause lethality following high-dose whole-thorax lung irradiation (WTLI). To date, the exact mechanisms of injury development following insult remain ill-defined and there are no FDA approved pharmaceutical agents or medical countermeasures. Traditionally, RILI development is considered as three phases, the clinically latent period, the intermediate acute pneumonitis phase and the later fibrotic stage. Utilizing matrix-assisted laser desorption ionization mass spectrometry imaging, we identified a number of lipids that were reflective of disease state or injury. Lipids play central roles in metabolism and cell signaling, and thus reflect the phenotype of the tissue environment, making these molecules pivotal biomarkers in many disease processes. We detected decreases in specific surfactant lipids irrespective of the different pathologies that presented within each sample at 180 days post whole-thorax lung irradiation. We also detected regional increases in ether-linked phospholipids that are the precursors of PAF, and global decreases in lipids that were reflective of severe fibrosis. Taken together our results provide panels of lipids that can differentiate between naive and irradiated samples, as well as providing potential markers of inflammation and fibrosis.

Summary: Gaucher disease is caused by mutations in the GBA1 gene, leading to the accumulation of GlcSph which activates an mTORC1-dependent pathogenic mechanism in neurons. Inhibition of acid ceramidase can prevent mTOR hyperactivity and lysosomal dysfunction, suggesting a potential therapeutic target for GBA1-associated neurodegeneration. Incubation with exogenous GlcSph in wild-type neurons can recapitulate the abnormalities observed in mutant neurons, highlighting the role of GlcSph in the pathogenesis of Gaucher disease.

Rapid Detection of Viral Envelope Lipids Using Lithium Adducts and AP-MALDI High-Resolution Mass Spectrometry

Summary: An unmet need exists for analytical strategies that can characterize the lipid composition of viral envelopes and do so on a high-throughput scale. The use of atmospheric pressure MALDI with lithium adduct consolidation allows for rapid and confident profiling of total lipid extracts from enveloped viruses, specifically geared towards sphingolipid detection. This approach provides a robust and high-throughput lipid detection platform for structural characterization of lipid extracts from viruses.

Cardiac retinoic acid levels decline in heart failure

Summary: The study showed that there is a decline in cardiac ATRA in heart failure, despite sufficient reserves of cardiac vitamin A. Strategies to normalize ATRA metabolism, such as CYP26 inhibition, may have therapeutic potential in preventing cardiac remodeling and functional decline. This suggests that low cardiac ATRA attenuates critical gene expression programs in heart failure.

Structure-specific, accurate quantitation of plasmalogen glycerophosphoethanolamine

Structural Fingerprinting of Short Interfering RNA Therapeutics by Solution Nuclear Magnetic Resonance Spectroscopy

Summary: Nucleic acids are a popular platform for developing biotherapeutics. There are currently 14 short oligonucleotide therapeutics and 2 mRNA vaccines approved by the FDA. However, there is a lack of high-resolution structural techniques for characterizing these molecules. In this study, nuclear magnetic resonance (NMR) methods were used to structurally fingerprint short oligonucleotide therapeutics, demonstrating their utility and broad applicability.

Altered RBP1 Gene Expression Impacts Epithelial Cell Retinoic Acid, Proliferation, and Microenvironment

Summary: The study investigated the relationship between RBP1 expression and atRA, finding that RBP1 and atRA are reduced in mammary tumor tissue and tumorigenic epithelial cell lines. Conditions in tumor microenvironments reduced RBP1 expression and atRA, while treatments with specific agonists or agents could restore RBP1 expression and atRA levels. This work highlights the potential therapeutic benefit of restoring RBP1 expression and endogenous atRA in diseases with reduced RBP1.

Hepatic CYP2B10 is highly induced by binge ethanol and contributes to acute-on-chronic alcohol-induced liver injury

Rapid identification of short oligonucleotide impurities using lithium adduct consolidated MALDI-TOF mass spectrometry
