Intranasally administered in situ gelling nanocomposite system of dimenhydrinate: preparation, characterization and pharmacodynamic applicability in chemotherapy induced emesis model

The aim of the current manuscript was to test the applicability of a nanocomposite system of penetration enhancer vesicles (PEVs) within polymeric in situ forming gel network composed of poloxamer and hyaluronic acid for the intranasal delivery of the antiemetic dimenhydrinate (DMH). PEVs were prepared using phospholipids and labrasol/transcutol/PEG 400 as penetration enhancers, and characterized for entrapment efficiency (EE%), particle size, zeta potential and morphology. The nanocomposite in situ forming gel system was characterized for its sol-gel temperature, viscosity and mucoadhesiveness, and was pharmacodynamically tested on a cisplatin induced emesis model in rats in terms of food, water, kaolin intake and stomach weight content. The selected PEVs formula displayed EE% of 83% for DMH, particle size of 121 nm and a surface charge of 0.83 mV. The selected nanocomposite in situ gelling formula showed a viscosity of 2.13 Pa.S, mucoadhesive force of 0.62 N and DMH controlled release over 6 hours. The pharmacodynamic study showed the superiority of the nanocomposite in situ gelling formula; being administered at a lower dose than the oral marketed formula. The described nanocomposite system proved to be successful for the intranasal delivery of DMH, thus presenting a promising delivery modality for similar antiemetics.

Curcumin nanoemulsion ameliorates brain injury in diabetic rats

Summary: The aim of this study was to investigate the therapeutic effects of curcumin nanoemulsion (NC) on cerebral damage in diabetic rats. The results showed that NC treatment reduced neurological deficit and redox imbalance, decreased inflammation and apoptosis, and restored glucose transporter 1 expression in the brain.

Towards more efficient inhalable chemotherapy: Fabrication of nanodiamonds-releasing microspheres

Summary: In this study, dried powder form of nanodiamonds with controlled particle size was developed for pulmonary delivery of chemotherapeutic agents. Addition of mannitol in the nanospray-drying process prevented the aggregation of nanodiamonds, and the resulting microparticles exhibited optimal aerodynamic size for pulmonary delivery. In vitro and in vivo studies showed that nanodiamonds embedded in mannitol microspheres could efficiently deliver drugs to lung cancer cells with limited systemic circulation.

Curcumin nanoemulsion counteracts hepatic and cardiac complications associated with high-fat/high-fructose diet in rats

Summary: The study found that CUR nanoemulsion can improve insulin resistance and hyperlipidemia caused by a high-fat high-fructose diet, and have beneficial effects on electrocardiogram, serum enzyme levels, and other biological indicators of rats. It also has a protective effect on hepatic and cardiac oxidative stress and DNA damage.

Intranasally administered melatonin core-shell polymeric nanocapsules: A promising treatment modality for cerebral ischemia

Summary: This study successfully developed MEL-loaded polymeric nanocapsules (MEL-PNCs) and investigated their potential in neuroprotection in a rat model of cerebral ischemia reperfusion injury. The MEL-PNCs showed good drug release and permeation properties, and demonstrated a significant reduction in oxidative stress and inflammation caused by cerebral ischemia, leading to the restoration of hippocampal neurons.

Curcumin-resveratrol nano-formulation counteracting hyperammonemia in rats

Summary: Malnutrition and low dietary protein intake can lead to peripheral and central hyperammonemia, particularly in pediatrics. Curcumin and resveratrol have shown effectiveness against liver and brain injuries, but their bioavailability is low. This study investigated the use of a nanoemulsion formulation containing both curcumin and resveratrol in ameliorating protein-deficient diet-induced hyperammonemia in rats. The results demonstrated that the nanoemulsion effectively reduced ammonia levels, oxidative damage, and improved cellular energy performance in the liver and brain.

Intranasal ondansetron microemulsion counteracting the adverse effects of cisplatin: animal study

Summary: This study demonstrated that ondansetron microemulsion, administered via the nasal or oral route, can alleviate the adverse effects of cisplatin chemotherapy. The nasal administration of ondansetron microemulsion showed superior effects compared to oral administration.
