Comprehensive analysis of lncRNAs and mRNAs in skeletal muscle of rainbow trout (Oncorhynchus mykiss) exposed to estradiol

Estradiol (E2) is a steroid hormone that negatively affects muscle growth in rainbow trout (Oncorhynchus mykiss), but the mechanisms directing with this response are not fully understood. To better characterize the effects of E2 in muscle, we identified differentially regulated mRNAs and lncRNAs in juvenile rainbow trout exposed to E2. Here, we performed next-generation RNA sequencing and comprehensive bioinformatics analyses to characterize the transcriptome profiles, including mRNAs and long noncoding RNAs (lncRNAs), in skeletal muscle of rainbow trout injected with E2. A total of 226 lncRNAs and 253 mRNAs were identified as differentially regulated. We identified crucial pathways, including several signal transduction pathways, hormone response, oxidative response and protein, carbon and fatty acid metabolism pathways. Subsequently, a functional lncRNA-mRNA co-expression network was constructed, which consisted of 681 co-expression relationships between 164 lncRNAs and 201 mRNAs. Moreover, a lncRNA-pathway network was constructed. A total of 65 key lncRNAs were identified that regulate 20 significantly enriched pathways. Overall, our analysis provides insights into mRNA and lncRNA networks in rainbow trout skeletal muscle and their regulation by E2 while understanding the molecular mechanism of lncRNAs.

Whole-genome sequencing identifies I-SceI-mediated transgene integration sites in Xenopus tropicalis snai2:eGFP line

Summary: This study investigated the transgenic process mediated by I-SceI nuclease and identified the transgene integration sites through whole-genome sequencing. It was found that the transgene integration loci were located in a noncoding region of chromosome 1, possibly mediated by sequence homology. This is the first genome-wide sequencing study on a transgenic organism generated with I-SceI, providing insights into the mechanisms of this transgenic method.

Efficient induction and sustenance of pluripotent stem cells from bovine somatic cells

Summary: Despite the lack of success in deriving naive bovine embryonic stem cells, the generation of bovine induced pluripotent stem cells (biPSCs) has been reported. Enhancing reprogramming efficacy and inhibiting TGF beta signaling have been found to support robust self-renewal of biPSCs. The findings contribute to advancing biPSC-based biotechnologies and understanding bovine pluripotency.

4-Methylcyclohexane methanol (MCHM) affects viability, development, and movement of Xenopus embryos

Summary: This study analyzed the toxicological effects of the coal cleaning chemical, MCHM, on HEK293 T human cell line cultures and Xenopus tropicalis embryos following a spill in 2014. Antioxidants were found to alleviate the toxicity of MCHM, and the embryos showed developmental defects and reversible non-responsiveness. Continued follow-up studies on the impact of MCHM on these cell lines and embryos are important.
