Repeated mild traumatic brain injury (rmTBI), the most common type of traumatic brain injuries, can result in neurological dysfunction and cognitive deficits. However, the molecular mechanisms and the long-term consequence of rmTBI remain elusive. In this study, we developed a modified rmTBI mouse model and found that rmTBI-induced transient neurological deficits and persistent impairments of spatial memory function. Furthermore, rmTBI mice had long-lasting detrimental effect on cognitive function, exhibiting memory deficits even 12 weeks after rmTBI. Microarray analysis of whole genome gene expression showed that rmTBI significantly altered the expression level of 87 genes which are involved in apoptosis, stress response, metabolism, and synaptic plasticity. The results indicate the potential mechanism underlying rmTBI-induced acute neurological deficits and its chronic effect on memory impairments. This study suggests that long-term monitoring and interventions for rmTBI individuals are essential for memory function recovery and reducing the risk of developing neurodegenerative diseases.

Regulator of calcineurin 1 is a novel RNA-binding protein to regulate neuronal apoptosis

Yan Yun, Yun Zhang, Chen Zhang, Liyan Huang, Shichuan Tan, Pin Wang, Carles Vilarino-Guell, Weihong Song, Xiulian Sun

Summary: The study reveals that RCAN1 is a novel RNA-binding protein, and its RNA aptamer can block RCAN1-induced inhibition of NFAT and NF-kappa B signaling pathways, reducing neuronal apoptosis and playing a neuroprotective role.

Summary: This study suggests that the use of antidepressants may be a risk factor for the new onset of T2DM, particularly tricyclic antidepressants. The risk of T2DM increases in a linear trend with the duration of antidepressant use. Routine T2DM screening is recommended for antidepressant users.

Associations between feelings/behaviors during COVID-19 pandemic lockdown and depression/anxiety after lockdown in a sample of Chinese children and adolescents

Summary: This study found that specific factors such as suicidal ideation, quarreling with parents, insomnia, difficulty in concentrating during online learning, and anxious and depressed mood during lockdown were positively associated with depression and anxiety among children and adolescents after COVID-19 pandemic lockdown. Missing teachers was negatively associated with both depression and anxiety. Living in urban areas and not living with parents were positively associated with depression.

Brain-derived neurotrophic factor in Alzheimer's disease and its pharmaceutical potential

Lina Gao, Yun Zhang, Keenan Sterling, Weihong Song

Summary: Synaptic abnormalities are a key aspect of Alzheimer's disease, connecting pathological changes to cognitive impairment. Brain-derived neurotrophic factors are crucial in synaptic plasticity for learning and memory, and have potential as diagnostic biomarkers and therapeutic molecules for AD. The exact mechanisms underlying the effects of impaired BDNF signaling on AD are still unclear.

Summary: Alzheimer's disease is a neurodegenerative disorder that severely impairs memory and cognition. It is characterized by Aβ deposition, tau tangles, synaptic abnormalities, and dysfunctions. Targeting the synapse and early treatment windows hold promise for finding effective treatments for AD patients.

Summary: The use of clinical psychoactive drugs can pose unpredictable threats to fetal development. Opicapone, a newly effective peripheral COMT inhibitor, is used for the treatment of Parkinson's disease and other dopamine-related disorders. However, the impact of opicapone on embryonic development is still unknown.

Distinct effects of SDC3 and FGFRL1 on selective neurodegeneration in AD and PD

Juelu Wang, Peiye Chen, Bolang Hu, Fang Cai, Qin Xu, Sipei Pan, Yili Wu, Weihong Song

Summary: Alzheimer's disease (AD) and Parkinson's disease (PD) are age-related neurodegenerative disorders that cause specific neuronal loss in the cholinergic and dopaminergic systems, respectively. The mutations in APP(SWE) and SNCA(A53T) have been implicated in the pathology of AD and PD, promoting the generation of A beta and the aggregation of alpha-synuclein, respectively. We found that SDC3 and FGFRL1 genes were differentially expressed in cells carrying these mutations and their knockdown attenuated cell death induced by oxidative stress.

Targeting NLRP3 inflammasome for neurodegenerative disorders

Jing Yao, Zhe Wang, Weihong Song, Yun Zhang

Summary: Neuroinflammation is a crucial pathological feature in neurological diseases, including Alzheimer's disease (AD). NLRP3, a member of the NLRs family, is primarily expressed in microglia and plays a significant role in inflammation by activating the NLRP3 inflammasome to release proinflammatory cytokines and cause pyroptosis. Recent studies have also highlighted the importance of neuronal NLRP3 in the pathogenesis of neurological disorders.

Application of a Digital Mental Health Clinic in Secondary Schools: Functionality and Effectiveness Evaluation

Yi Xu, Hongshen Yang, Zhou Jin, Jian Xiang, Haiyun Xu, Yili Huang Pokay, Haibo Mao, Xugong Cai, Yili Wu, Deborah Baofeng Wang

Summary: This study evaluated the functionality and effectiveness of a digital mental health clinic (DMHC) in secondary schools in China. The DMHC provided accessible and free mental health services to students, addressing issues such as learning difficulties, interpersonal relationships, and emotional distress. The mini course videos on emotional assistance, personal growth, and family member relationships were highly popular among students. The DMHC also served as a crisis prevention and intervention tool, helping students at high risk for suicide and self-injury.

Amyloid β-based therapy for Alzheimer's disease: challenges, successes and future

Yun Zhang, Huaqiu Chen, Ran Li, Keenan Sterling, Weihong Song

Summary: Amyloid & beta; protein (A & beta;) is the main component of neuritic plaques in Alzheimer's disease. It has been considered as the molecular driver of Alzheimer's pathogenesis. The failures of A & beta;-targeted clinical trials have raised doubts about the amyloid cascade hypothesis, but recent successes have alleviated those doubts. This review discusses the evolution of the hypothesis, its application in Alzheimer's diagnosis and treatment, and the prospects for future research and development of A & beta;-targeted approaches.

Summary: GSDME plays a crucial role in the pathogenesis of Alzheimer's disease by regulating the switch from apoptosis to pyroptosis and participating in neuroinflammatory response. Knockdown of GSDME has been shown to improve cognitive impairments, indicating that GSDME could be a therapeutic target for Alzheimer's disease.
