Serum Metabolomic Profiles for Breast Cancer Diagnosis, Grading and Staging by Gas Chromatography-Mass Spectrometry

Detection of metabolic signature for breast cancer (BC) has the potential to improve patient prognosis. This study identified potentially significant metabolites differentiating between breast cancer patients and healthy controls to help in diagnosis, grading, staging and determination of neoadjuvant status. Serum was collected from 152 pre-operative breast cancer (BC) patients and 155 healthy controls in this case-controlled study. Gas chromatography-mass spectrometry (GC-MS) was used to obtain metabolic profiles followed by chemometric analysis with the identification of significantly differentiated metabolites including 7 for diagnosis, 18 for grading, 23 for staging, 15 for stage III subcategory and 10 for neoadjuvant status (p-value < 0.05). Partial Least Square Discriminant Analysis (PLS-DA) model revealed a distinct separation between healthy controls and BC patients with a sensitivity of 96% and specificity of 100% on external validation. Models for grading, staging and neoadjuvant status were built with Decision Tree Algorithm with predictive accuracy of 71.5%, 71.3% and 79.8% respectively. Pathway analysis revealed increased glycolysis, lipogenesis, and production of volatile organic metabolites indicating the metabolic alterations in breast cancer.

Metabolomics-based researches in autoimmune liver disease: A mini-review

Summary: Metabolomics is a promising approach for studying metabolites in the biological system. It offers new opportunities for assessing disease progression and biomarker selection, and provides a better understanding of the pathophysiology of diseases. Autoimmune liver disease, including autoimmune hepatitis, primary biliary cholangitis, and primary sclerosing cholangitis, is a major global public health concern, with poorly understood pathogenesis. This review provides a comprehensive overview of the role of metabolomics in these diseases and discusses the disturbed metabolic pathways.

Acidotic and hypoxic tumor microenvironment induces changes to histone acetylation and methylation in oral squamous cell carcinoma

Summary: Hypoxia and acidosis are common characteristics of the tumor microenvironment and affect the metabolism of cancer cells. These stresses change the post-translational modifications of histones, leading to tumorigenesis and drug resistance. However, the impact of these changes on oral squamous cell carcinoma (OSCC) is not well understood. This study investigated the effects of hypoxic and acidotic microenvironment on histone acetylation and methylation in OSCC using LC-MS-based proteomics. It identified specific histone marks associated with gene regulation and revealed the position-dependent changes in histone modifications. The findings provide insights into the adaptation of tumor cells to hypoxic and acidotic stresses and their connections with histone crosstalk.

PX-12 synergistically enhances the therapeutic efficacy of vorinostat under hypoxic tumor microenvironment in oral squamous cell carcinoma in vitro

Summary: Hypoxia is a characteristic feature of solid tumors, and the hypoxia-inducible factor 1-alpha (HIF-1 alpha) is a key regulator of the tumor microenvironment. Vorinostat and PX-12 are inhibitors of HIF-1 alpha with different mechanisms of action, and their combination shows synergistic effects against oral squamous cell carcinoma (OSCC) under hypoxic conditions.

Untargeted metabolomics analysis of gentamicin-induced tolerant colonies of Klebsiella pneumoniae

Summary: To understand the development of antibiotic resistance in Klebsiella pneumoniae, this study conducted metabolomics analysis on sensitive and gentamicin-resistant strains. The results showed that increasing gentamicin concentration led to morphological changes in K. pneumoniae colonies, and strains grown at the highest concentration were considered resistant. Metabolomic analysis identified several regulated metabolites associated with resistance development, which could potentially be used for rapid detection of gentamicin-resistant clinical strains.

Dysregulation of metalloproteins in ischemic heart disease patients with systolic dysfunction

Summary: This study aims to investigate the role of metalloproteins in the pericardial fluid of ischemic heart disease (IHD) patients. By analyzing the samples from patients with different left ventricular ejection fraction (LVEF) levels, a total of 154 metalloproteins were identified, among which the copper binding protein ceruloplasmin and the iron binding protein serotransferrin were found to be associated with the ferroptosis pathway in IHD patients.

Desmoarylcoumarin and episoyasapogenol B: two new compounds from Desmodium salicifolium (Poir.) DC (Fabaceae)

Summary: A new 3-arylcoumarin and a previously unreported triterpenoid, along with five known flavonoids and one carbohydrate, were isolated from the roots of Desmodium salicifolium. Their structures were determined through extensive spectroscopic and mass spectrometric analyses. The methanolic extract, EtOAc and n-BuOH fractions, as well as some isolated compounds, were evaluated for their antibacterial and antioxidant activities. The EtOAc fraction exhibited moderate activity against Enterococcus faecalis, and the methanolic extract and EtOAc fraction showed DPPH scavenging activity. Compound 1 showed a moderate antibacterial activity against Enterococcus faecalis and moderate DPPH scavenging activity.

Recent studies on advance spectroscopic techniques for the identification of microorganisms: A review

Summary: The continuous development of resistance to antibiotic drugs by microorganisms causes high mortality and morbidity. Pathogens with distinct features and biochemical abilities make them destructive to human health. Therefore, early identification of the pathogen is of substantial importance for quick ailments and healthcare outcomes. Several advanced spectroscopic techniques have been utilized for the identification of microorganisms, particularly bacteria, in research studies conducted in the last five years. Among these techniques, mass spectrometry (MS), infrared (IR) spectroscopy, Raman spectroscopy (RS), and nuclear magnetic resonance (NMR) spectroscopy have been widely used. MALDI-TOF/MS, a type of MS technique, has been particularly favored for microbial identification. The review discusses the identification of 44 bacteria using these techniques, with the most studied organisms being E. coli, S. aureus, and other microorganisms.

Targeted Analysis of Veterinary Drugs in Food Samples by Developing a High-Resolution Tandem Mass Spectral Library
