Involvement of type VI secretion system in secretion of iron chelator pyoverdine in Pseudomonas taiwanensis

Rice bacterial blight caused by Xanthomonas oryzae pv. oryzae (Xoo) is one of the most destructive rice diseases worldwide. Therefore, in addition to breeding disease-resistant rice cultivars, it is desirable to develop effective biocontrol agents against Xoo. Here, we report that a soil bacterium Pseudomonas taiwanensis displayed strong antagonistic activity against Xoo. Using matrix-assisted laser desorption/ionization imaging mass spectrometry, we identified an iron chelator, pyoverdine, secreted by P. taiwanensis that could inhibit the growth of Xoo. Through Tn5 mutagenesis of P. taiwanensis, we showed that mutations in genes that encode components of the type VI secretion system (T6SS) as well as biosynthesis and maturation of pyoverdine resulted in reduced toxicity against Xoo. Our results indicated that T6SS is involved in the secretion of endogenous pyoverdine. Mutations in T6SS component genes affected the secretion of mature pyoverdine from the periplasmic space into the extracellular medium after pyoverdine precursor is transferred to the periplasm by the inner membrane transporter PvdE. In addition, we also showed that other export systems, i.e., the PvdRT-OpmQ and MexAB-OprM efflux systems (for which there have been previous suggestions of involvement) and the type II secretion system (T2SS), are not involved in pyoverdine secretion.

Harnessing Fluorescent Moenomycin A Antibiotics for Bacterial Cell Wall Imaging Studies

Evaluation of the Antifungal Activities of Photorhabdus akhurstii and Its Secondary Metabolites against Phytopathogenic Colletotrichum gloeosporioides

Summary: This study demonstrated that Photorhabdus akhurstii sp. nov. 0813-124 has the potential to control the devastating fungus Colletotrichum gloeosporioides in strawberries. The bioactive ingredients, glidobactin A and cepafungin I, were identified and their expression in different phases was analyzed. These findings suggest that P. akhurstii sp. nov. 0813-124 could be used as a biocontrol agent or as part of a natural product repertoire for combating C. gloeosporioides.

Untargeted LC-MS/MS-Based Multi-Informative Molecular Networking for Targeting the Antiproliferative Ingredients in Tetradium ruticarpum Fruit

Summary: This study examined the antiproliferative activity of different extracts of Tetradium ruticarpum (TR) fruit and highlighted potentially antiproliferative compounds using an untargeted LC-MS/MS-based multi-informative molecular networking strategy. The EtOAc fraction F2-3 exhibited the most potent antiproliferative activity against human cell lines. Through computational tool-aided structure prediction and integration of various data, 11 indole alkaloids and 47 types of quinolone alkaloids were successfully annotated and visualized into three targeted bioactive molecular families. Several previously unreported quinolone alkaloids in TR fruit were discovered. Four indole alkaloids and five types of quinolone alkaloids were identified as potentially antiproliferative compounds in the EtOAc fraction F2-3. Evodiamine was confirmed as a major antiproliferative compound, and its structural analogues found in the molecular network showed promise as antitumor agents. This study exemplifies the application of an LC-MS/MS-based multi-informative molecular networking strategy in the discovery and annotation of bioactive compounds from complex mixtures of potential functional food ingredients.

Transcriptome of Nosema ceranae and Upregulated Microsporidia Genes during Its Infection of Western Honey Bee (Apis mellifera)

Summary: This study investigated the gene expression profile of Nosema ceranae, a fungal parasite of honey bees, at different time points post-infection. The results revealed that the gene expression of N. ceranae during infection is closely related to gene transcription, protein synthesis, and structural proteins. The findings have important implications for the control of nosemosis in apiculture.

Surfactin secreted by Bacillus amyloliquefaciens Ba01 is required to combat Streptomyces scabies causing potato common scab

Summary: This study aimed to determine whether surfactin is the main compound active against Streptomyces scabies in potato common scab. The results showed that the srf gene cluster of Bacillus amyloliquefaciens Ba01 is responsible for the secretion of surfactin and inhibition of S. scabies. Knocking out the srf gene cluster significantly decreased the swarming ability, biofilm formation, enzyme activity, and inhibitory effect of Ba01 on S. scabies.

Structure of the heterotrimeric membrane protein complex FtsB-FtsL-FtsQ of the bacterial divisome

Summary: The FtsB-FtsL-FtsQ complex plays a crucial role in regulating cell wall synthesis during bacterial cell division by mediating the synthesis of the cell-wall peptidoglycan. The full-length structure of the heterotrimeric FtsBLQ complex is reported, showing a tilted V-shaped conformation. This structure may facilitate interactions with other divisome proteins in an allosteric manner, leading to a proposed mechanism for the regulation of peptidoglycan synthases by the FtsBLQ complex.

Metabolomic Analysis Demonstrates the Impacts of Polyketide Synthases PKS14 and PKS15 on the Production of Beauvericins, Bassianolide, Enniatin A, and Ferricrocin in Entomopathogen Beauveria bassiana

Summary: This study investigated the metabolite profiles of pks14 and pks15 gene overexpression strains in B. bassiana and compared them to their knockout strains using LC-MS/MS with GNPS. The results showed that pks14 and pks15 clusters interacted with biosynthetic clusters encoding insect-virulent metabolites and upregulated certain secondary metabolites. This suggests that pks14 and pks15 may be directly or indirectly associated with key pathways in insect pathogenesis of B. bassiana.
