Summary: Malignant rhabdoid tumor of the kidney (MRTK) has a poor prognosis and is unresponsive to radiotherapy and chemotherapy. In this study, BKM120 was identified as a potential therapeutic agent for MRTK by inhibiting the PI3K/Akt signaling pathway to induce cell apoptosis and cycle arrest. Both in vitro and in vivo experiments confirmed the inhibitory effects of BKM120 on MRTK.

Summary: Acute kidney injury (AKI) can lead to chronic kidney disease (CKD) through maladaptive repair of proximal tubule cells (PTCs), involving dedifferentiation and profibrotic cytokine secretion. Cyclin G1 (CG1) and cyclin-dependent kinase 5 (CDK5) are key factors in maladaptive dedifferentiation and could be therapeutic targets for CKD.

Summary: This study focused on the atypical cadherin MUCDHL, which is down-regulated in colorectal tumors, especially in tumors with worse prognosis. Loss of MUCDHL enhanced tumor incidence and burden in intestinal tumor-prone mice, and targeted key oncogenic signaling pathways through its extracellular domain. The study illustrates how the loss of a gene critical for the morphological and functional features of mature cells contributes to tumorigenesis by dysregulating oncogenic pathways.

Summary: Killin has been found to play a role in p53-mediated G1 arrest by upregulating p21 protein expression independently of p53. Knockout of Killin significantly decreases p53-mediated G1 arrest, and double knockout of Killin and p21 completely abolishes this arrest. These findings suggest that Killin is involved in multiple cell cycle checkpoint controls, including p53-mediated G1 arrest.

Summary: Although many diterpenoid alkaloids have been evaluated for their antiproliferative activity, little is known about the effects of hetisine-type C20-diterpenoid alkaloids and their derivatives. In this study, a total of 43 novel derivatives were prepared and their antiproliferative effects against human cancer cell lines were evaluated. The results showed that some newly synthesized derivatives exhibited significant suppressive effects, suggesting that these compounds could be potential candidates for overcoming triple-negative breast cancer.

Summary: This study aimed to develop clinically tools for early detection of anastomotic leakage (AL) by analyzing inflammatory factors (IFs) in abdominal drainage. The research proposed an Anastomotic Score system (AScore-POD3) based on IFs with high diagnostic efficacy, which was validated in an independent cohort. A nomogram combining AScore-POD3 with other clinical risk factors showed high C-index, suggesting the potential for early diagnosis of AL after gastric cancer surgery.

Summary: PCID2 is amplified in colorectal cancer (CRC) and promotes cell proliferation, cell cycle progression, and migration/invasion while inhibiting apoptosis. It functions as an oncogene in CRC by enhancing canonical Wnt/beta-catenin signaling and suppressing the CTNNB1-ARF-p53 axis. PCID2 may serve as an independent predictive marker for CRC recurrence.

Summary: This study elucidated the functional significance and molecular mechanisms of DUSP5P1 lncRNA in gastric carcinogenesis. High expression of DUSP5P1 was associated with shortened survival in gastric cancer patients and promoted migration, invasion, and metastasis of gastric cancer cells. Mechanistically, DUSP5P1 activated the transcription of ARHGAP5 and dysregulated focal adhesion and MAPK signaling pathways. DUSP5P1 also played a role in platinum drug resistance.

Summary: This meta-analysis compared the short- and long-term outcomes of laparoscopic gastrectomy (LG) versus open gastrectomy (OG) for gastric cancer. The results showed that the intraoperative complication rate was similar between the two groups, but LG had fewer postoperative complications. The severe postoperative complication rate and perioperative mortality were comparable. LG had a lower number of retrieved lymph nodes and a shorter proximal resection margin distance compared to OG. However, the distal resection margin distance was similar. LG resulted in shorter recovery time, including time to first ambulation and time to first flatus. Additionally, LG had a shorter postoperative length of stay. The 5-year overall survival and disease-free survival were similar between the two groups.

Summary: Linc01503 is significantly upregulated in GC and closely associated with patient survival. The transcription factor EGR1 activates linc01503 transcription. Knockdown of linc01503 leads to apoptosis activation and G1/G0 phase arrest in GC. Linc01503 interacts with histone modification enzymes EZH2 and LSD1 to silence DUSP5/CDKN1A expression in GC, mediating cell cycle progression and tumorigenesis.
