Summary: The gold standard for treating peripheral nerve injuries with large nerve gaps is autologous nerve grafts. However, incorporating mineral coated microparticles (MCMs) to release growth factors like NGF and GDNF can significantly enhance axon growth and improve limb function in rats with nerve grafts.

Summary: This study reveals that axon regeneration after conditioning lesion is more pronounced in young mice than in old mice. The mechanism behind this difference is not clearly explained. By analyzing gene expression and constructing networks, AGTR1a is identified as a potential therapeutic target for spinal cord injury treatment.

Summary: The study investigates the role of NT3-chitosan in the treatment of chronic spinal cord injury (SCI). Results show that NT3-chitosan can induce robust neural regeneration after clearing the lesion core and trimming scar tissues. Magnetic resonance-diffusion tensor imaging (MR-DTI) can be used to monitor spatial and temporal changes of the lesion area, which correlates well with anatomical analyses. This research suggests that NT3-chitosan may be a potential treatment for chronic SCI.

Summary: "The study suggests that CCL2 is not essential for macrophage accumulation, myelin clearance, and axon regeneration in the peripheral nervous system. Other CCR2 chemokines, resident macrophage proliferation, and CCR2-independent monocyte recruitment can compensate for the absence of CCL2."

Summary: Presynapses efficiently communicate information by locally recycling synaptic vesicles. This study investigates the mechanisms of trafficking older synaptic vesicles back to the soma for recycling, and shows that the choice of microtubule or actin cytoskeleton networks depends on the speed of vesicle trafficking and the direction of transport. The loss of synaptic vesicle with bound myosin V is identified as the differentiating mechanism. These findings provide insights into the maintenance and plasticity of presynaptic function.

Summary: The study identified that upregulation of SARM1 in neurons and astrocytes at early stage after SCI. Conditional deletion of SARM1 in neurons and astrocytes promoted functional recovery and neuronal regeneration after SCI, while reducing neuroinflammation.

Summary: Melatonin has long-term benefits on cerebral damage after ischemia and reperfusion injury, as it reduces infarct volume, neuronal cell death, reactive astrogliosis, and glial scar formation, while promoting axonal regeneration and neurobehavioral recovery. By inhibiting the activities of GSK-3 beta and RIP1K, melatonin effectively counteracts the deleterious effects due to astrocyte responses and improves functional recovery during the chronic phase of cerebral ischemia and reperfusion injury.

Summary: This study reveals the underlying principles of macropinocytic cup formation and closure in Dictyostelium amoebae using light-sheet microscopy. Cups are formed by recruiting PIP3 domains and a specialized F-actin scaffold, and they close through inward or outward actin polymerization and membrane stretching and delamination when cup expansion stalls. This research is important for understanding intracellular fluid engulfment processes.

Summary: Proliferative astrocytes play a critical role in the pathogenesis of CCM by being a major source of VEGF; non-cell-autonomous crosstalk between CCM endothelium and astrocytes propels vascular lesion development; components of the hypoxic program represent potential therapeutic targets for CCMs.

Summary: This study investigated the dynamic expression of cytokines in the distal nerve stumps after peripheral nerve injury using protein microarray analysis, revealing distinct patterns between proximal and distal nerve stumps. Distal stumps showed increased expression of interleukins and proteins related to inflammatory and apoptotic responses, while proximal stumps mainly upregulated growth factors. The findings suggest potential therapeutic targets and may advance clinical treatments for peripheral nerve injury.

Summary: Taxifolin reduces neuroinflammation and microglial pyroptosis after spinal cord injury by inhibiting oxidative stress and regulating the PI3K/AKT signaling pathway. It also promotes axonal regeneration and improves functional recovery. Taxifolin may be a potential therapeutic agent for spinal cord injury.

Summary: A homolog of actin in planarian Dugesia japonica was identified, and it was found that RNAi targeting actin can affect planarian regeneration, resulting in the formation of protrusions on the dorsal side, increased division of phospho-H3 mitotic cells, and decreased differentiation in regenerating tissues, indicating that actin plays an important role in proliferation and differentiation control in planarian regeneration.

Summary: Neonatal hypoxic-ischemic encephalopathy (HIE) is a major neurological disorder in newborns with unclear mechanisms. This study demonstrated that hypoxia and ischemia (HI) caused significant neurological damage in neonatal rats, with brain-derived neurotrophic factor (BDNF) being down-regulated and potentially playing a role in promoting neurite regrowth after injury. These findings suggest a novel strategy for treating neurological defects induced by cerebral ischemia and hypoxia.

Summary: This review discusses the role and mechanism of mussel-inspired PDA-based multilayered coatings in enhancing bone formation. These coatings have multifunctionality, which can synergistically enhance immunomodulation, angiogenesis, antibiosis, and antitumor properties, promoting bone defect repair and regeneration. The research inspires the design and fabrication of multifunctional PDA-based multilayered coatings to meet the demands of different application scenarios, but further clinical translation is needed.
