Silica nanoparticles induced the pre-thrombotic state in rats via activation of coagulation factor XII and the JNK-NF-κB/AP-1 pathway

Silica nanoparticles (SiNPs) play a vital role in medical applications such as drug delivery and cancer therapy. SiNPs can translocate into the bloodstream through all the possible routes of entry. However, there have been scarce studies on the pre-thrombotic effect of SiNPs and the mechanism of the prethrombotic state in vivo. We specifically focused on the changes of platelet function and blood coagulation in Wistar rats after 7 consecutive days of intravenous injection of SiNPs (52 nm). The platelet aggregation assay, structural changes of platelet membrane glucoproteins, coagulation test, coagulant/anticoagulant and fibrinolytic factors and the possible molecular mechanism of pre-thrombotic state formation were performed. Our results demonstrated a significant increase in platelet aggregation rate and platelet activation after SiNP exposure. The clotting time was significantly shortened while fibrinogen (FIB) contents were increased. There were sustained increases in coagulation factors and thrombin-antithrombin complex (TAT) expression induced by SiNPs. Antithrombin III (AT-III) of the SiNP-treated groups was significantly decreased while the concentrations of the tissue factor pathway inhibitor (TFPI), tissue plasminogen activator (t-PA) and D-dimer were elevated. The phosphorylation of nuclear factor-.B/p65 (NF-.B/p65) and activator protein-1/c-Jun (AP-1/c-Jun) and the protein levels of JNK were increased after SiNP exposure. In summary, our results revealed that SiNPs induced the hypercoagulable and prethrombotic state in rats through the interaction between platelet activation, coagulation system hyperfunction, anti-coagulation and fibrinolytic resistance. Direct interactions between SiNPs and coagulation factor XII (F XII) and the JNK-NF-kappa B/AP-1 pathway might be involved in the regulation of pre-thrombotic state formation.

Clinical implications of TPO and AOX1 in pediatric papillary thyroid carcinoma

Summary: This study demonstrates that TPO and AOX1 are significantly downregulated in pediatric PTC, with decreased levels of TPO and AOX1 proteins in blood plasma. These proteins show satisfactory predictive performance in plasma and low levels are associated with poor survival in patients with PTC. The findings suggest that TPO and AOX1 could serve as novel biomarkers for the diagnosis and prognosis of pediatric PTC.

Summary: This study describes four cases of sclerosing epithelioid fibrosarcoma (SEF) in children with unique genetic characteristics. The tumors were located in the deep soft tissue and exhibited prominent sclerotic hyalinized stroma and monomorphic epithelioid cells. Genetic analysis revealed translocation between EWSR1 and CREB3L1, as well as amplification of fusion parts, indicating the presence of a unique molecular variant of SEF.

Epidemiology of extrapulmonary tuberculosis among pediatric inpatients in mainland China: a descriptive, multicenter study

Summary: Pediatric extrapulmonary tuberculosis (EPTB) in China has not been well studied in terms of clinical characteristics and economic burden. This multicenter study aimed to investigate and summarize the situation of EPTB in pediatric patients. The results showed differences between EPTB, pulmonary TB (PTB), and combined TB in terms of hospitalization frequency, length of stay, discharge against medical advice, mortality, low birth weight, diagnosis at the first visit, and hospitalization plan situation. EPTB and combined TB had higher financial burdens compared to PTB.

Reduction of LPAR1 Expression in Neuroblastoma Promotes Tumor Cell Migration

Summary: The tumor metastasis in high-risk neuroblastoma patients is a significant problem that affects patient survival. This study utilized comprehensive bioinformatics analysis to identify LPAR1 as a potential target for the treatment of neuroblastoma. The decreased expression of LPAR1 in neuroblastoma cells and the migration-inhibitory effects of LPA-LPAR1 axis suggest that LPAR1 may be a promising target for future neuroblastoma treatment.

Clinical relevance of BCOR internal tandem duplication and TP53 aberration in clear cell sarcoma of the kidney

Summary: Clear cell sarcoma of the kidney (CCSK) is often associated with TP53 deletion, which is related to adverse clinical outcomes. A study analyzed 39 CCSK patients and found that 87.18% of patients had BCOR internal tandem duplication (ITD), 2.56% had YWHAE rearrangement, and 2.56% had BCOR-CCNB3 gene fusion. Furthermore, 25.64% of patients had TP53 aberration, and patients with both BCOR ITD and TP53 deletion had the poorest prognosis.

Summary: This study evaluated the important role of BPTF in bone marrow (BM) in predicting the progression of neuroblastoma (NB). It was found that BPTF is an independent risk factor for predicting NB progression. The BPTF inhibitor AU1 is expected to become a new targeted drug for NB therapy, revealing previously unknown mechanisms of BPTF in NB cell proliferation and metastasis through TFAP4 and PI3K/AKT pathways.

Translational practice of fluorescence in situ hybridisation to identify neuroblastic tumours with TERT rearrangements

Summary: Recently, TERT gene rearrangements have been found in neuroblastoma (NB), but their prevalence in other types of neuroblastic tumours (NTs) is unknown. This study developed a practical method for detecting TERT defects and evaluated their clinical relevance as biomarkers for NT prognosis. The study used fluorescence in situ hybridisation (FISH) to assess TERT genomic status in Chinese children with NTs. TERT rearrangements were identified in 6.0% of NTs overall, and in 12.4% of high-risk patients. TERT rearrangements were associated with aggressive NT subtypes and decreased survival chances.

Arsenic toxicity: sources, pathophysiology and mechanism

Summary: Arsenic toxicity is a complex and pervasive public health problem that affects millions of people worldwide. This review aims to consolidate current knowledge on arsenic toxicity, including its sources, chemical forms, and the pathways through which it affects human health. The study found that arsenic is associated with chronic diseases such as cancer, cardiovascular disorders, and neurotoxicity, and oxidative stress, genotoxicity, and epigenetic modifications play important roles in arsenic-induced cellular damage. Further research is needed to fill gaps in our understanding of arsenic toxicity and inform public health policies aimed at minimizing exposure.

Developmental neurotoxicity (DNT) QSAR combination prediction model establishment and structural characteristics interpretation

Summary: With the increasing incidence of neurodevelopmental disorders, it is important to screen and evaluate developmental neurotoxicity (DNT) compounds from a large number of environmental chemicals and understand their mechanisms. This study conducted a DNT qualitative structure-activity relationship (QSAR) study for the first time, and preliminarily illustrated the structural characterization of DNT compounds. Multiple models and methods were used to construct prediction models, resulting in the best model with good predictive ability. By combining different models, both MCC values and application domain values were improved. Additionally, through modeling descriptors analysis and structure alerts (SAs) identification, electronical properties, van der Waals volume-related properties, and S, Cl or P containing substructure were found to be associated with DNT. This study lays a foundation for further DNT prediction of environmental exposures in human and contributes to the understanding of DNT mechanism.

Impact of untreated and microbially treated equalization tank effluent of textile industry on freshwater fish Channa punctata using haematological, biochemical, histopathological and ultrastructural analysis

Summary: The unregulated discharge of untreated or partially treated industrial wastewater poses a serious threat to aquatic ecosystems. This study exposed Channa punctata fish to untreated and microbially treated textile industry effluent to analyze their toxicity effects. The results showed that the untreated effluent group had more severe hematological, biochemical, histopathological, and ultrastructural alterations compared to the microbially treated group. This highlights the importance of finding more effective microbial treatments to protect aquatic life and human health.

Mechanism of simulated lunar dust-induced lung injury in rats based on transcriptomics

Summary: This study investigated the mechanism of lunar dust simulant (CLDS-i) in inducing inflammatory lung injury. The results showed that CLDS-i regulated immune inflammatory cells and activated signaling pathways related to inflammatory diseases, promoting the occurrence and development of inflammatory injury. The study also found that amino acid metabolism imbalance might be related to the activation of the NF-kappa B signaling pathway.
