Summary: Our study identifies differentially expressed microRNAs in retinoblastoma and pediatric retina as well as advanced and non-advanced tumors. We provide evidence of the epithelial-mesenchymal transition (EMT) and chemoresistance programs in advanced tumors, potentially related to miR-181a-5p. Our findings highlight the importance of understanding the EMT and chemoresistance mechanisms in tumor progression and suggest miR-181a-5p as a potential therapeutic target.

Summary: The study identified a molecular network in T2E-positive prostate cancer driving NOTCH signaling, involving direct activation of NOTCH1 and HES1 by ERG, downregulation of miR-449a, and subsequent oncogenic processes. Inhibition of the NOTCH pathway and knockdown of HES1 reduced the oncogenic properties of PCa cells, suggesting a potential therapeutic target for T2E-positive prostate cancer.

Summary: Elevated TSH levels reduce the distribution of certain miRNAs in hepatic mitochondria, leading to the suppression of key genes involved in lipid metabolism and consequent impairment of fatty acid oxidation and triglyceride accumulation in hepatocytes. This study highlights the functional significance of altered miRNA mitochondrial-cytoplasmic distribution in regulating hepatic lipid metabolism.

Summary: The study reveals that CDK13 is significantly upregulated in human PCa tissues. Depletion or overexpression of CDK13 in PCa cells affects cell proliferation, and the interaction between CDK13 and E2F5 plays a role in promoting proliferation. Mechanistically, transcriptional activation of endogenous CDK13 promotes E2F5 expression by facilitating circCDK13 formation.

Summary: The study revealed that miR-4319 is downregulated in RB tissues and cell lines. Overexpression of miR-4319 inhibits cell proliferation, migration, invasion and promotes apoptosis in RB cells. CD147 is identified as a direct target gene for miR-4319, and miR-4319 suppresses cell functions through inhibiting the CD147-mediated MMPs expression.

Summary: This study revealed the role of the EZH2/miR-139-5p/TOP2A axis in regulating cellular senescence and cell proliferation in HCC, enriching the molecular mechanisms of EZH2-mediated epigenetic regulation in HCC. Therapeutically targeting EZH2 to induce cellular senescence and destroy senescent cells could be a potential treatment strategy for HCC.

Summary: Prostate cancer is a common male neoplasm and is the second leading cause of cancer death in American men. Traditional diagnosis of prostate cancer relies on prostate secreted antigen (PSA) evaluation, but false positives are common, making additional tests necessary. This study explores the role of miR-153 in controlling cell growth and invasion in high Gleason Score prostate cancer, and suggests that miR-153 could serve as a circulating biomarker for diagnosis.

Summary: This study investigates the role of vanin-1 (VNN1) in the progression of acute kidney injury (AKI) to chronic kidney disease (CKD). The results show that VNN1 expression is upregulated after ischemia/reperfusion (I/R) injury and high levels of VNN1 are associated with poor renal repair. Knocking out VNN1 accelerates recovery and inhibits renal fibrosis after I/R injury. The study also reveals that VNN1 plays a role in senescence of renal tubular cells by activating the expression and phosphorylation of retinoblastoma 1 (RB1). These findings suggest that targeting VNN1 could be a potential strategy to obstruct the AKI-CKD transition.

Summary: The origin and evolution of cancer cells are mainly driven by DNA mutations, but the role of translation errors in cancer biology remains unclear. Research indicates that cellular senescence induced by oncogenes, tumor suppressors, or chemotherapy is associated with a reduction in translational readthrough, controlled by the RB tumor suppressor pathway. Cells that escape from senescence exhibit increased translational readthrough, which may be linked to cancer progression.

Summary: This study explored the potential of using small activating RNAs and growth factors to induce the differentiation of human adipose-derived mesenchymal stem cells into hair cell-like cells. The results showed that selected small activating RNAs effectively activated the expression of the ATOH1 gene and induced the differentiation of hair cell progenitor cells into hair cell-like cells. This RNA activation technology could offer a new strategy for hair cell regeneration.
