Summary: This study demonstrates that the H7N9 VLP vaccine provides complete clinical protection against HP H7N9 virus, significantly inhibits virus shedding, and abolishes viral replication in chickens. The VLP vaccine represents a promising alternative strategy for the development of novel H7N9 avian influenza vaccines for chickens.

Avian Influenza H7N9 Virus Adaptation to Human Hosts

Summary: Avian influenza virus H7N9, which has been circulating in avian hosts for decades, was identified as a human pathogen in 2013. Through comparing sequences from avian and human H7N9 datasets, it was found that the virus has undergone significant amino acid substitutions possibly essential for human adaptation. The rapid evolution of the avian H7N9 virus has resulted in almost identical protein sequences in avian and human viruses, leading to increased risk of human infection and potential human-to-human transmission.

Summary: Research shows a strong association between rare variants in the MX1 gene and H7N9 infection in humans. The identified MxA protein variants lost the ability to inhibit avian IAVs, exerting a dominant-negative effect on wild-type MxA's antiviral function, possibly leading to an MxA null phenotype in heterozygous carriers. This study provides genetic evidence for the crucial role of MX1-based antiviral defense in controlling zoonotic IAV infections in humans.

Risk assessment of the newly emerged H7N9 avian influenza viruses

Summary: Since 2013, the H7N9 avian influenza viruses (AIVs) have caused over 1500 human infections and a mortality rate of approximately 40%. Despite vaccination efforts, the viruses continue to evolve rapidly in poultry and some strains have the ability to escape vaccine-induced immunity. Recent analysis showed that the H7N9 AIVs from the early epidemic waves had dual receptor binding characteristics, but more recent strains lost or retained weak human receptor binding. The viruses also demonstrated increased acid stability but reduced thermal stability compared to earlier strains, and showed poor cross-reactivity with human monoclonal antibodies and ferret antisera.

Summary: This study systematically analyzed sex hormone and cytokine levels in male and female H7N9 influenza patients, and found that H7N9-infected men had significantly reduced testosterone levels associated with poor outcomes. A mouse model confirmed the causal link between viral infection and testosterone depletion.

Summary: This study examined the B cell response to H7 HA in individuals who had recovered from natural H7N9 virus infection. Circulating H7-reactive B cells were found to recognize a diverse antigenic landscape on the HA molecule and exhibit broad recognition of unrelated H7 strains. Functional testing of the antibodies revealed diverse patterns of inhibition, including neutralizing and HA trimer-disrupting activities.

Spatio-temporal spread and evolution of influenza A (H7N9) viruses

Summary: This study conducted a spatial and temporal Bayesian evolutionary analysis to understand the spread and evolution process of the H7N9 influenza A virus. The findings showed that the virus likely emerged in August 2012 in a border area of Anhui Province, 6 months earlier than the first reported human case. By the end of 2013, two major epicenters had formed in the Yangtze River Delta and Pearl River Delta regions, and the virus spread to other regions at an average speed of 6.57 km/day. The study also developed a model based on the random forest algorithm to predict the occurrence risk of the H7N9 virus.

Summary: After the introduction of the H7N9 inactivated vaccine in 2017, low-pathogenic H7N9 strains decreased while highly-pathogenic H7N9 strains dominated. The 2019 H7N9 strains formed a separate genetic branch, distinct from strains isolated in previous years. The analysis suggested accelerated evolution of the H7N9 influenza virus subtype under vaccination pressure.

Summary: The H7N9 avian influenza virus has caused five waves of human infection in China, but further cases have been prevented since September 2017 through the use of a vaccine in poultry. However, the virus has not been eradicated from poultry, with genetic analysis revealing new genotypes and the potential for escape from vaccine-induced immunity. The antigenic differences identified between the 2019 H7N9 viruses and the vaccine strain used for control in poultry highlight ongoing challenges in evolution and control of the virus.

Phylogenetic analysis suggested two distinct trade routes as potential sources of the virus introduction into Gansu.

Summary: Avian influenza viruses have the ability to infect mammals, posing a significant threat to public health. The H7N9 subtype of avian influenza virus that emerged in China in 2013 resulted in a large number of human infections, the reasons for which are still unclear. The presence of a mammal-adapted H7N9 virus in poultry in 2013 could be a significant factor contributing to the unusually high number of human infections during that period.

Summary: Avian influenza A (H7N9) virus, closely associated with live poultry markets, has caused infections in humans in China since 2013. Five waves of the epidemic occurred between 2013 and 2017, with the emergence of highly pathogenic strains causing concern. Rapid response and control by specialists and officials utilized new technologies and surveillance tools. Characteristics of the virus were reviewed from multiple perspectives to provide tools for reducing future threats from H7N9 and other viruses.

Influenza H7N9 Virus Hemagglutinin with T169A Mutation Possesses Enhanced Thermostability and Provides Effective Immune Protection against Lethal H7N9 Virus Challenge in Chickens

Summary: In this study, a T169A mutation was introduced into the HA protein of H7N9 avian influenza virus to generate a mutant with increased thermostability. Immunization with the mutant combined with ISA 71VG induced higher levels of cross-reactive antibodies and cytokine secretion, and provided effective protection against heterologous H7N9 AIV infection. The results suggest that the mutant represents a promising subunit vaccine candidate for the prevention and control of H7N9 avian influenza.

Assessing different interventions against Avian Influenza A (H7N9) infection by an epidemiological model

Summary: This study evaluates the effectiveness of various intervention measures against avian influenza A(H7N9) infection using an epidemiological model. Results show that reducing poultry-human/poultry interaction and culling domestic poultry are highly effective in diminishing infections, while vaccination of a certain proportion of poultry is essential for disease prevention. Cleaning and disinfecting the environment have limited impact on reducing infection risk. Importantly, screening imported poultry is crucial for stopping disease diffusion.

Digital PCR applications for the diagnosis and management of infection in critical care medicine

Summary: Infection is a major cause of morbidity and mortality in critical care medicine. Digital PCR (dPCR) offers technical advantages for the management of infection, such as higher sensitivity, reproducibility, and absolute quantification. This review examines the applications of dPCR in critical care medicine, including microbial gene amplification, quantification of microbial burden, and analysis of host responses.
