Characterization of the NES2Y cell line and its use in the production of human glucose-responsive insulin producing (hGRIP) cell lines by cell-cell fusion

Diabetes mellitus is a debilitating disease and alternative methods of treatment are a priority if the short-term and long term sequelae are to be avoided. Here the authors manipulate NES2Y cells, which have the potential to be used as 'fusion partners' to produce human insulin-producing glucose-responsive hybrids. The fusion experiments were carried out using polyethylene glycol (PEG) and electroporation. Human insulin production of the resulting hybrids (in response to glucose) was measured using ELISA. Our results showed that it is possible to engineer human glucose-responsive insulin-producing (hGRIPs) hybrid cells by the manipulation of two different cell types. The resulting hybrids continuously grow in culture and are insulin-secreting and glucose-responsive for a period of time. Immortalised cells with the characteristics of human beta cells could provide an important resource for experimental studies in Type 1 diabetes, such as an improved understanding of the fundamental mechanisms of glucose-responsive insulin processing and secretion, transplantation and drug screening programs.

Recovery of serum testosterone levels is an accurate predictor of survival from COVID-19 in male patients

Summary: This study aimed to assess the feasibility of a validated assay of T-cell responses as a complement to antibody tests for evaluating the levels of protection generated by natural infection or SARS-CoV-2 vaccines. The results showed that the IGRA test detected T-cell responses in naturally exposed and vaccinated individuals, and there was a high correlation with specific antibody levels. These findings suggest that the IGRA test could be a promising tool for measuring the state of the immune response to SARS-CoV-2.

Summary: Plastic pollution is a global problem, with potential health consequences for animals and humans who ingest and inhale plastic particles. Nanoplastics (NPs), ranging from 1 nm to 1000 nm, are particles formed from the erosion or breakage of larger plastic debris and can vary in physical properties and composition. The exposure to NPs may lead to alterations in xenobiotic and energy metabolism, nutrients absorption, cytotoxicity, and behavior. Our study prospectively investigated the presence of NPs in human peripheral blood using fluorescence techniques and nanocytometry, demonstrating the accurate detection of NPs with flow cytometry.

Immunoregulatory Biomarkers of the Remission Phase in Type 1 Diabetes: miR-30d-5p Modulates PD-1 Expression and Regulatory T Cell Expansion

Summary: The partial remission phase of type 1 diabetes is characterized by endogenous insulin production and downregulated autoimmunity. This study identifies and validates a unique miRNA signature during this phase and demonstrates the immunomodulatory role of miR-30d-5p. Inhibition of miR-30d-5p results in increased regulatory T cells and changes in immune cell populations in vivo. This miRNA signature provides potential biomarkers and enhances understanding of the mechanisms driving the partial remission phase.
