Quantitative evaluation of fungi of the genus Candida in the feces of adult patients with type 1 and 2 diabetes - a pilot study

Background: Gastrointestinal tract microbiota, particularly bacterial microflora, seem to have a different qualitative and quantitative composition in both type 1 (T1DM) and type 2 diabetes (T2DM) mellitus cases as compared to non-diabetic individuals. So far, there are no data from diabetes research concerning the prevalence of fungi, particularly the most common genus, i. e. Candida, which are important components of human colon microflora. We aimed to examine whether there are quantitative changes of Candida fungi in the feces of patients with T1DM and T2DM as compared to healthy controls. Findings: Overall, we included 44 diabetic patients (27 patients with T1DM and 17 with T2DM) as well as 17 healthy, non-diabetic controls. Feces and blood samples were collected from all study individuals. DNA was isolated from fecal samples and quantitative real time PCR (qPCR) was applied in order to determine the number of fungal cells. Statistical association with selected clinical and biochemical features was examined. There was a difference in the amount of Candida in the feces among the three examined groups (p = 0.007). Candida spp. populations in T1DM and T2DM subjects were larger as compared to controls (p = 0.017 and p = 0.037, respectively). However, no difference was found between T1DM and T2DM. No association was identified between the quantity of fungi and examined patients' characteristics, except for negative correlation with blood lipid parameters in T2DM group. Conclusions: Candida fungi appear to be more prevalent in the feces of patients with T1DM and T2DM. Their amount seems to be associated with serum lipids in T2DM patients. This initial finding requires further confirmation.

Summary: This study compared the accuracy and efficacy of two Illumina platforms (iSeq and MiSeq) in sequencing 16S rRNA and described some important similarities and differences in 20 fecal samples. The results showed that iSeq technology had lower species richness compared to MiSeq, and MiSeq had a higher number of taxa. Additionally, MiSeq performed better in the detailed analysis of microbiota composition.

Biocidal Activity of Tannic Acid-Prepared Silver Nanoparticles towards Pathogens Isolated from Patients with Exacerbations of Chronic Rhinosinusitis

Summary: This study assessed the biological activity of tannic acid-prepared silver nanoparticles (TA-AgNPs) on sinonasal pathogens and nasal epithelial cells. The results showed that TA-AgNPs were effective against most pathogens isolated from patients with acute exacerbations of chronic rhinosinusitis (AECRS) at non-toxic concentrations to human cells. However, prolonged exposure to TA-AgNPs increased toxicity to nasal epithelial cells and enabled some pathogens to resist their biocidal action. Sensitivity testing may be necessary before application.

Identifying Bacteria Species on Microscopic Polyculture Images Using Deep Learning

Summary: Preliminary microbiological diagnosis is time-consuming, but deep learning methods based on microscopic images have been introduced to replace bacteriological examination. In this study, we propose using multi-MIL, a multi-label classification method, to analyze polyculture images and further shorten the diagnosis time. We evaluate our approach using a dataset of microscopic images for four considered bacteria species, achieving an ROC AUC above 0.9 and proving the feasibility for future experiments.

Do NAAT-Based Methods Increase the Diagnostic Sensitivity of Streptococcus agalactiae Carriage Detection in Pregnant Women?

Summary: The aim of this study was to evaluate the sensitivity of group B Streptococcus (GBS) detection using polymerase chain reaction primers and a preincubation step. Vaginal and rectal swabs were collected from 97 pregnant women and subjected to enrichment broth culture-based diagnostics and amplification using specific primers. The preincubation step increased the sensitivity of GBS detection, and additional GBS cases were identified using nucleic acid amplification techniques. The atr gene primers showed the highest number of true positive results compared to other primers.

New insights into diversity of the upper respiratory tract microbiota and its relationship with SARS -CoV-2 viral load in the nasopharyngeal epithelial cells in patients with COVID-19

Summary: This study investigated the effects of SARS-CoV-2 infection on the composition of the upper respiratory tract microbiota and found a possible association between microbiota composition and viral load. The study analyzed samples from 60 patients and divided them into groups based on viral replication rate. The results showed significant differences in microbiota composition between groups with high and low replication rates, suggesting an association between viral load and microbiota composition.

Summary: This study investigated the quantity of Bifidobacterium and total bacteria in the colon of patients with T1DM and T2DM as well as in healthy subjects. The results showed that the number of Bifidobacterium was significantly lower in T1DM and T2DM groups compared to the control group, while the total bacteria showed no statistical difference. Furthermore, correlations were found between the number of Bifidobacterium and age, ALT, triglycerides, HBA1, and glucose level in the specific diabetes groups.
