Papaverine hydrochloride containing nanostructured lyotropic liquid crystal formulation as a potential drug delivery system for the treatment of erectile dysfunction

Purpose: Papaverine hydrochloride (PaHCl) is an old, well-known drug with spasmolytic activity but it has therapeutic effect in erectile dysfunction, too. As an intracavernous injection, it is not used in urologic clinics today because the side effects of the injection are pain, scarring or priapism. Our aim was to develop and test a topical semi-solid preparation containing PaHCl that would provide an alternative administration option by eliminating the undesirable side effects of the injection. Materials and methods: Lyotropic liquid crystal (LLC) systems were formulated as a semi-solid preparation with different concentrations of PaHCl. The characterization of the LLC structure was performed by polarization microscopy using a Leica image analyzer and rheological measurements. The drug diffusion and penetration tests were performed with in vitro synthetic membrane and an ex vivo human epidermis, using Franz diffusion cell to test the skin penetration of PaHCl. Human skin was investigated by Raman microscope to visualize the Active Pharmaceutical Ingredient (API) in different skin layers. Results: The results of diffusion and penetration showed reverse concentration dependency. The in vitro and ex vivo studies correlated with each other and the results of Raman microscopy. The LLC structure influenced the penetration results, the lower viscosity and lamellar structure increased penetration through the skin. Conclusion: Based on our results, a PaHCl containing topically used LLC formulation may be a suitable and effective alternative to the injectable formulation.

Rheological effects of hypertonic saline and sodium bicarbonate solutions on cystic fibrosis sputum in vitro

Summary: This study investigated the effect of hypertonic salt solutions on CF sputum viscoelasticity in vitro, using a rheometer to measure the rheological properties of samples. The results showed significant changes in gel strength when different additives were used at varying ratios, with NaHCO3 having the most pronounced effect. The study highlights the importance of individual evaluation of sputum samples based on their rheological properties for effective treatment.

Comparison of Synthetic Membranes to Heat-Separated Human Epidermis in Skin Permeation Studies In Vitro

Summary: This study compared permeation across innovative synthetic membranes and human epidermis, with the Strat-M membrane showing the greatest similarity to HSE. Good correlations were found between the permeation data of different formulations across different membranes, allowing the prediction of permeation across HSE using synthetic membranes. Strat-M and Skin PAMPA membranes have the potential to select and differentiate dermal formulations containing diclofenac sodium as an early screening model.

The effects of glycols on molecular mobility, structure, and permeability in stratum corneum

Summary: The skin provides an attractive alternative to conventional drug administration routes, but the stratum corneum (SC) presents challenges as an efficient barrier to most compounds. This study investigates the molecular effects of three types of glycols in the SC, aiming to understand their influence on molecular mobility and structure, as well as their efficiency as permeation enhancers. The results show that all three glycols increase lipid mobility in the SC and have permeation enhancing effects. There is also a non-monotonic response in permeation enhancement with respect to glycol concentration.

The effect of non-invasive dermal electroporation on skin barrier function and skin permeation in combination with different dermal formulations

Summary: This study investigated the effect of non-invasive dermal electroporation (EP) on the barrier function of the skin and the permeation of a macromolecule. The results showed that EP can temporarily decrease the skin's barrier function and the structure of the skin is restored shortly after treatment. EP significantly increased the permeation rate of the macromolecule, but the composition of the formulations had a great influence on the permeation.

Analytical Quality by Design (AQbD) Approach to the Development of In Vitro Release Test for Topical Hydrogel

Summary: The aim of this study was to design an effective in vitro release test method using USP apparatus IV with a semi-solid adapter for diclofenac sodium hydrogel, by applying the analytical quality by design approach. The analytical target profile and critical method attributes were defined, and an initial risk assessment was conducted. High-risk parameters were identified and a test protocol was developed for the hydrogel system.

Development of Capsaicin-Containing Analgesic Silicone-Based Transdermal Patches

Summary: In this study, the release properties, skin penetration, and analgesic effect of a modified silicone-polymer-based transdermal therapeutic system (TTS) loaded with low-dose capsaicin were investigated. The TTS exhibited controlled, zero-order kinetic release of capsaicin and effectively penetrated into the epidermis and dermis of human skin. It was concluded that this modified TTS is suitable for relieving traumatic and inflammatory pain.

Lipid Nanoparticles: An Effective Tool to Improve the Bioavailability of Nutraceuticals

Summary: Nano-scale bioactive colloidal carrier systems, such as lipid nanoparticles (LNPs), have shown great potential in overcoming challenges associated with disease treatments. LNPs not only improve the pharmacokinetics and stability of encapsulated compounds, but also provide efficient targeting and reduced toxicity. However, the poor solubility, stability, and absorption of natural therapeutic molecules have hindered research in the nutraceutical sector. Loading these molecules into LNPs offers a promising solution by utilizing their ability to accommodate both hydrophilic and hydrophobic molecules.

Design and Optimization of In Situ Gelling Mucoadhesive Eye Drops Containing Dexamethasone

Summary: This study aims to develop in situ gelling mucoadhesive ophthalmic preparations that can improve the bioavailability of eye drops. The results show that formulations based on thermosensitive polymers and mucoadhesive polymers can increase the residence time and penetration of the active pharmaceutical ingredient, thereby enhancing the bioavailability of the eye drops.

Application of Xanthan Gum and Hyaluronic Acid as Dermal Foam Stabilizers

Summary: Foams are becoming increasingly popular in dermatology due to their advantages in terms of easy spreading, good skin sensation, and applicability in special skin conditions. This study aimed to improve the stability of dermal foams by using different hydrogel-forming polymers. The combination of xanthan gum and dexpanthenol was found to be ideal for both foam formation and stability.
