Organisms containing an increase in DNA content by whole number multiples of the entire set of chromosomes are defined as polyploid. Cells that contain more than two sets of chromosomes were first observed in plants about a century ago, and it is now recognized that polyploid cells form in many eukaryotes under a wide variety of circumstances. Although it is less common in mammals, some tissues, including the liver, show a high percentage of polyploid cells. Thus, during post-natal growth, the liver parenchyma undergoes dramatic changes characterized by gradual polyploidization during which hepatocytes of several ploidy classes emerge as a result of modified cell-division cycles. Liver cell polyploidy is generally considered to indicate terminal differentiation and senescence and to both lead to a progressive loss of cell pluripotency and to a markedly decreased replication capacity. In adults, liver polyploidization is differentially regulated upon loss of liver mass and liver damage. Here we review the current state of understanding about how polyploidization is regulated during normal and pathological liver growth, and detail by which mechanisms hepatocytes become polyploid. Summary: The expression of NKG2D ligands is associated with aggressive liver tumorigenesis, and downregulation of these ligands by beta-catenin signaling may contribute to the less aggressive phenotype of CTNNB1-mutated HCC tumors.

Tumor Cells and Cancer-Associated Fibroblasts: An Updated Metabolic Perspective

Summary: Tumors exhibit metabolic heterogeneity with utilization of carbon sources other than glucose, such as amino acids, fatty acids, and lactate. This diversity in metabolic pathways influences cancer cell invasion capabilities and is also present in Cancer-Associated Fibroblasts (CAF).

Hepatocyte Polyploidy: Driver or Gatekeeper of Chronic Liver Diseases

Romain Donne, Flora Sangouard, Severine Celton-Morizur, Chantal Desdouets

Summary: Polyploidy, or whole-genome amplification, is a condition where an organism has more than two basic sets of chromosomes, with diverse effects in different systems. The liver has the ability to adjust its ploidy, with about 30% of hepatocytes being polyploid. Research has explored mechanisms of polyploid cell development, regulation, consequences for liver function, and potential roles in tumor formation.

Glycolysis Dependency as a Hallmark of SF3B1-Mutated Cells

Summary: Cancer-associated SF3B1 mutations result in aberrant transcripts that alter protein translation in metabolic genes, leading to a switch towards increased glucose uptake and glycolysis dependency. This study provides evidence of the oncogenic involvement of mutant SF3B1 in uveal melanoma and suggests targeting glycolysis inhibitors as a potential therapeutic strategy.

Summary: Advanced gastric adenocarcinoma is a disease with poor prognosis. In recent years, new drugs, such as trastuzumab for HER2-positive tumors, ramucirumab alone or in combination with paclitaxel, and trifluridine-tipiracil, have improved patient survival. Other drugs, including monoclonal antibodies and targeted therapies, are also being evaluated.
