Mammalian cell growth on gold nanoparticle-decorated substrates is influenced by the nanoparticle coating

In this work, we study epithelial cell growth on substrates decorated with gold nanorods that are functionalized either with a positively charged cytotoxic surfactant or with a biocompatible polymer exhibiting one of two different end groups, resulting in a neutral or negative surface charge of the particle. Upon observation of cell growth for three days by live cell imaging using optical dark field microscopy, it was found that all particles supported cell adhesion while no directed cell migration and no significant particle internalization occurred. Concerning cell adhesion and spreading as compared to cell growth on bare substrates after 3 days of incubation, a reduction by 45% and 95%, respectively, for the surfactant particle coating was observed, whereas the amino-terminated polymer induced a reduction by 30% and 40%, respectively, which is absent for the carboxy-terminated polymer. Furthermore, interface-sensitive impedance spectroscopy (electric cell-substrate impedance sensing, ECIS) was employed in order to investigate the micromotility of cells added to substrates decorated with various amounts of surfactant-coated particles. A surface density of 65 particles/mu m(2) (which corresponds to 0.5% of surface coverage with nanoparticles) diminishes micromotion by 25% as compared to bare substrates after 35 hours of incubation. We conclude that the surface coating of the gold nanorods, which were applied to the basolateral side of the cells, has a recognizable influence on the growth behavior and thus the coating should be carefully selected for biomedical applications of nanoparticles.

Summary: This study evaluated the nationwide trends in care and postoperative outcomes for patients with distal esophageal and gastro-esophageal junction cancer. The results showed that there was a transition towards more neo-adjuvant treatment, transthoracic esophagectomy and minimally invasive surgery over a 10-year period, accompanied by improved postoperative outcomes.

Summary: The aim of this study was to determine the effectiveness, safety, and cost-effectiveness of prophylactic mesh placement in preventing parastomal hernia formation in the long term. The results showed that while the procedure did not reduce the incidence of parastomal hernia after 5 years of follow-up, it did delay its onset without significant differences in morbidity, mortality, or quality of life. Additionally, it was found to be a cost-effective option.

Summary: It is unclear whether Ivor Lewis (IL) or McKeown (McK) esophagectomy is preferred in patients with potentially curable esophageal or gastro-esophageal junction (GEJ) cancer. A study in the Netherlands found similar survival rates and complication rates between patients who underwent IL or McK esophagectomy. Therefore, both IL and McK esophagectomy can be considered for patients with mid to distal esophageal and GEJ cancer.

Bioinspired Membrane Interfaces: Controlling Actomyosin Architecture and Contractility

Summary: The creation of artificial lipid bilayers on planar supports allows for a controlled study of membrane-confined processes. In this study, actomyosin networks were bound to supported planar lipid bilayers doped with phosphatidylinositol-4,5-bisphosphate (PtdIns[4,5]P2) via the membrane-actin linker ezrin. By analyzing this membrane system using high-resolution fluorescence microscopy, the researchers were able to understand the connectivity and contractility of the actomyosin network. They found that the network's architecture and dynamics depend not only on the PtdIns[4,5]P2 concentration, but also on the presence of negatively charged phosphatidylserine (PS).

HAV-Peptides Attached to Colloidal Probes Faithfully Detect E-Cadherins Displayed on Living Cells

Summary: Cell adhesion molecules, such as E-cadherin, play a crucial role in various biological processes. Researchers synthesized E-cadherin mimetic peptides and attached them to solid-supported membranes on colloidal probes to identify this molecule on cell surfaces. Two different functionalization strategies were established, and binding studies showed successful attachment of the peptides to the membrane surface. The presence of E-cadherin on living cells was confirmed using colloidal probes functionalized with HAV-peptides.

Performance of a consensus-based algorithm for diagnosing anastomotic leak after minimally invasive esophagectomy for esophageal cancer

Summary: Oesophageal adenocarcinomas may exhibit different histopathological patterns, such as excessive acellular mucin pools, signet-ring cells, and poorly cohesive cells. These components have been suggested to correlate with poor outcomes after neoadjuvant chemoradiotherapy. However, their independent effects and potential confounders have not been thoroughly studied. In this study, we evaluated the presence of extracellular mucin, signet-ring cells, and poorly cohesive cells before and after treatment, and their association with pathological response and prognosis. The findings suggest that the presence of these factors is not related to pathological response, but a pre-treatment presence of ≥10% poorly cohesive cells and a post-treatment presence of ≥1% signet-ring cells may indicate inferior prognosis.

Summary: This study aimed to gain consensus on healthcare professionals' perspectives on the self-management support information needs of esophageal cancer patients during the preoperative phase. A survey was created based on literature, clinical consultations, and patient information leaflets. A total of 33 important topics were identified, and consensus was reached on the preferred sources of support, which were face-to-face contact and a case manager.
