Summary: The study reveals that alterations in serine metabolism affect the sensitivity of colorectal cancer cells to 5-FU. Resistant cells become dependent on serine by increasing its synthesis or uptake. Mitochondrial compartmentalization of one-carbon metabolism supports purine biosynthesis and DNA damage response in resistant cells.

Summary: Five aromatic ergosterols with unique ring systems, named spectasterols A-E (1-5), were isolated from Aspergillus spectabilis. Compound 3 demonstrated cytotoxic activity and anti-inflammatory effects, inducing cell cycle arrest and apoptosis in HL60 cells, and inhibiting COX-2 levels and NF-kappa B p65 nuclear translocation.

Summary: Cisplatin, a key player in chemotherapy, has been improved by developing new platinum-based prodrugs with active ligands. Platin-C, a platinum(iv)-based prodrug tethered to curcumin, showed enhanced chemotherapeutic efficacy and targeted mitochondria of cancer cells. It also demonstrated the ability to attenuate cellular inflammation.

Summary: This study investigated the effects of dietary curcumin (CUR) as an antioxidant on hepatic cellular senescence in aged mice. The results showed that CUR supplementation downregulated senescence-associated hepatic gene expressions, enhanced antioxidant properties, suppressed MAPK signaling cascades, and inhibited proinflammatory cytokines and SASPs expression, suggesting that CUR may be a nutritional strategy to prevent hepatic cellular senescence.

Summary: This study elucidated the role of MLPH in GBM radiation resistance, showing that MLPH promotes radiotherapy resistance by regulating the NF-kappa B pathway. Additionally, O-GlcNAcylation of MLPH protects it from degradation and stabilizes its presence in cells. These findings provide insights into a potential mechanism of GBM radiation resistance and suggest a therapeutic strategy for GBM treatment.

Summary: In p53-deficient colorectal cancer organoids, 5-fluorouracil induces pyrimidine imbalance, which leads to DNA damage and cell death. Rewiring glucose metabolism through PDK inhibition by DCA enhances 5-FU toxicity in glycolytic p53-deficient organoids. This has important implications for improving chemotherapy strategies for colorectal cancer, and p53 emerges as a key factor in determining the response to 5-FU.

Summary: In this study, the researchers investigated the mechanism of curcumin in alleviating lung damage induced by lipopolysaccharide (LPS) in ducks. The results showed that dietary supplementation of curcumin significantly relieved oxidation stress and inflammation parameters in the lungs of ducks. The remission may be attributed to the activation of the nuclear erythroid 2-related factor 2 (Nrf2)-antioxidant reaction element (ARE) and nuclear factor kappa B (NF-kappa B) signaling pathways. This study provides insights into the potential application of curcumin in waterfowl feed.

Summary: This study investigated the effects of a novel chalcone derivative MPP in combination with curcumin on the inflammatory pathways in breast and ovarian cancer cells. The results showed that MPP inhibited NF-κB activity, reduced the expression of inflammatory genes, and decreased the levels of proinflammatory cytokines and nitric oxide. These effects were enhanced when MPP was combined with curcumin.

Summary: Keap1-Nrf2 is a crucial signaling cascade for promoting or preventing carcinogenesis. Nutraceuticals, as dietary agents with health benefits, show great potential as chemoprevention agents for cancer. This review systematically analyzes the role of 12 different nutraceuticals in modulating the Nrf2/Keap1 signaling mechanism. Although there is considerable preclinical evidence, further high-quality research is needed to validate the potential role of these nutraceuticals in Keap1-Nrf2 modulation.
