TIAR and TIA-1 mRNA-Binding Proteins Co-aggregate under Conditions of Rapid Oxygen Decline and Extreme Hypoxia and Suppress the HIF-1α Pathway

T-cell intracellular antigen (TIA)-1 and TIA-1-related protein (TIAR) are mRNA-binding proteins that can aggregate within granules under specific stress conditions. In this study, we analyzed TIAR/TIA-1 aggregation under different hypoxic conditions, and studied the effects on the hypoxia-inducible factor (HIF)-1 alpha in different cancer cell lines. Under acute and pronounced hypoxic conditions TIAR/TIA-1 co-aggregated to granules and positive co-staining with eIF3 eta marker suggested these to represent stress granules. In parallel, HIF-1 alpha expression was blocked in cells displaying TIAR/TIA-1 granules. Silencing of TIAR and TIA-1 caused upregulation of HIF-1 alpha expression, as demonstrated by western blot, immunocytochemistry and HIF-1-dependent reporter gene expression. Additionally, a critical region of the 3' end of the untranslated HIF-1 alpha mRNA with possible adenosine-uridine-rich elements (AREs) was coupled to the luciferase reporter gene, causing downregulation of expression. Employing this reporter construct, inhibition of TIAR by siRNA attenuated the inhibitory cis-effect of this ARE-sequence. Furthermore, immunohistochemical analysis of A549 cell tumor xenografts revealed a nearly complementary expression of HIF-1 alpha and TIAR reflecting the control of HIF-1 alpha expression by TIAR as revealed in the cell culture studies. In sum, rapid and severe hypoxia caused co-aggregation of TIAR/ TIA-1 and these proteins suppressed HIF-1 alpha expression.

RA area, RA indexed area, RA peak longitudinal strain, and RA total emptying fraction were significantly associated with combined end-points including death, clinical worsening, and/or lung transplantation. RA volume and volume index showed marginal significant associations.

Pitfalls of using sequence databases for heterologous expression studies - a technical review

Summary: Synthesis of DNA fragments based on gene sequences from public resources has replaced traditional cloning methods. However, errors in database entries can lead to false sequence information and impact protein characterization in expression systems. Five common problems in public resources: incomplete gene annotation, incorrect annotations, automated corrections in transcripts, sequence mismatches, and lack of experimental validation for splicing patterns. This review presents a strategy to bypass these issues and avoid critical mistakes in future studies.

Value of Right and Left Ventricular T1 and T2 Blood Pool Mapping in Patients with Chronic Thromboembolic Hypertension before and after Balloon Pulmonary Angioplasty

Summary: The aim of this study was to assess the RV and LV blood pool T1 and T2 values in CTEPH patients and their correlation to pulmonary hemodynamic. Results showed that compared to control subjects, CTEPH patients had significantly reduced RV T2 values, which increased significantly after balloon pulmonary angioplasty. RV T2 values were positively correlated with hemodynamic measures. Therefore, RV T2 mapping can be a novel non-invasive CMR imaging marker for assessing disease severity, prognosis, follow-up, and therapy monitoring in PH.

Summary: In patients with idiopathic pulmonary arterial hypertension or chronic thromboembolic pulmonary hypertension, estimated plasma volume status is associated with central venous congestion and prognosis. Patients with high estimated plasma volume status and without edema have a poorer transplant-free survival rate, which may represent an under-recognized subgroup.

Summary: Acute respiratory distress syndrome (ARDS) is a common complication of influenza virus (IV) infection, which leads to the accumulation and impairment of alveolar protein. It has been found that IV infection inhibits the clearance of alveolar protein, possibly through upregulation of a proteinase called MMP. In addition, inhibition of MMP proteinase may improve the clearance of alveolar protein and thus provide a potential therapeutic approach for ARDS.

Further validation studies are needed to assess the clinical usefulness and reliability of these biomarkers in diagnosing aspiration-related conditions.
