Enucleation-Induced Rat Adrenal Gland Regeneration: Expression Profile of Selected Genes Involved in Control of Adrenocortical Cell Proliferation

Enucleation-induced adrenal regeneration is a highly controlled process; however, only some elements involved in this process have been recognized. Therefore, we performed studies on regenerating rat adrenals. Microarray RNA analysis and QPCR revealed that enucleation resulted in a rapid elevation of expression of genes involved in response to wounding, defense response, and in immunological processes. Factors encoded by these genes obscure possible priming effects of various cytokines on initiation of regeneration. In regenerating adrenals we identified over 100 up- or downregulated genes involved in adrenocortical cell proliferation. The changes were most significant at days 2-3 after enucleation and their number decreased during regeneration. For example, expression analysis revealed a notable upregulation of the growth arrest gene, Gadd45, only 24 hours after surgery while expression of cyclin B1 and Cdk1 genes was notably elevated between days 1-8 of regeneration. These changes were accompanied by changes in expression levels of numerous growth factors and immediate-early transcription factors genes. Despite notable differences in mechanisms of adrenal and liver regeneration, in regenerating adrenals we identified genes, the expression of which is well recognized in regenerating liver. Thus, it seems legitimate to suggest that, in the rat, the general model of liver and adrenal regeneration demonstrate some degree of similarity.

Effects of Galp and alarin peptides on HPA axis gene expression and adrenal function: In vivo experiments

Summary: The expression changes of peptides and their receptors, as well as steroidogenesis enzymes, were investigated in the hypothalamus, pituitary, and adrenal glands of rats after intraperitoneal administration of peptides. The results showed significant expression changes in the hypothalamus and pituitary after 1-hour exposure to peptides, including a decrease in corticotropin-releasing hormone expression and a decrease in the expression of galanin receptors. The expression of steroidogenesis enzymes also changed, with an increase in cholesterol desmolase expression after peptide administration.

Summary: Wharton's jelly-derived mesenchymal stem cells (WJ-MSCs) possess multilineage differentiation potential and specific surface markers. In this study, RNA sequencing analysis revealed differentially expressed genes associated with apoptosis and identified upregulation of ZBTB16 and FOXO1, as well as downregulation of TGFA, in all differentiated groups. Additionally, several novel marker genes related to WJ-MSCs differentiation were identified. This study sheds light on the molecular mechanisms involved in long-term culture and differentiation of WJ-MSCs.

Summary: The homeostasis of the adrenal gland is crucial for its proper functioning in both non-stressful and stressful conditions, involving interactions between different types of cells. However, there is insufficient information regarding marker gene expression in the rat adrenal gland under non-stressful conditions. This study aimed to determine the expression of marker genes in different zones of the rat adrenal gland and revealed distinct expression patterns of interstitial cell markers in specific zones.

The Enhanced Expression of ZWILCH Predicts Poor Survival of Adrenocortical Carcinoma Patients

Summary: The study examines the association of ZWILCH gene with adrenocortical carcinoma (ACC). The higher expression of ZWILCH gene was found in ACC tissue compared to normal adrenal glands. Upregulation of ZWILCH is correlated with tumor mitotic rate and patient survival. The enhanced ZWILCH level is also associated with activation of genes involved in cell proliferation and inhibition of genes related to the immune system.

Summary: This study aimed to investigate the relationship between serum MOTS-c concentration and muscle strength parameters, as well as oxygen consumption, in physically active individuals. The results showed that higher serum MOTS-c concentration was associated with greater muscle mass, force, and power, but not with exercise capacity.

The Prognostic Value of Cancer Stem Cell Markers (CSCs) Expression-ALDH1A1, CD133, CD44-For Survival and Long-Term Follow-Up of Ovarian Cancer Patients

Summary: Recurrent disease and treatment-associated chemoresistance are major contributors to poor clinical outcomes in ovarian cancer patients. This study investigated the expression of CSC-associated markers ALDH1A, CD44, and CD133 to predict patient prognosis. ALDH1A1-positive patients had significantly shorter overall survival (OS) and progression-free survival (PFS). Multivariate analysis revealed that ALDH1A1 expression, FIGO stage III-IV, and residual disease were associated with shorter OS. Targeting ALDH1A1-positive cancer cells may be a promising therapeutic strategy.

Estrogen receptor β affects hypoxia response in colorectal cancer cells

Summary: The presence of estrogen receptor beta (ER beta) is inversely correlated with the occurrence of colorectal cancer (CRC), and low expression of ER beta is associated with poorer overall survival of CRC patients. ER beta can inhibit tumorigenesis through various molecular pathways and also has an impact on the hypoxic response in CRC cells.
