Summary: The deterioration of inhibitory synapses may be a neurological basis for abnormal social behaviors. Manipulating GABAergic transmission has been linked to improved sociability. Ephrin-B2 (EB2), a synaptic protein, plays a key role in the maintenance and rearrangement of inhibitory synapses in the medial prefrontal cortex (mPFC). However, the specific role of EB2 in inhibitory cell types in the pathophysiology and treatment of social deficits is still unknown. This study found that depletion of EB2 specifically occurred in GABAergic neurons, leading to social deficits. Overexpression of mPFC EB2 restored the defective social behavior. The findings suggest that the effect of EB2 on social deficits is specific to certain anatomical and cell types. Furthermore, an upregulation of HDAC4 expression was observed in EB2-deficient mice, with nuclear HDAC4 playing a role in mediating the mechanism underlying the impairment. Treatment with LMK235 significantly rescued social deficits, indicating a potential use for HDAC targeting agents in treating social deficits.

Summary: Tolerogenic dendritic cell (tolDC) therapies aim to restore self-tolerance in patients with autoimmune diseases. This study tested the suitability of using nanoparticles containing F-19 for labeling therapeutic human tolDC for detection through F-19-MRI. The results showed that F-19-NP is a suitable imaging agent for tolDC and does not affect their function.

Summary: Viruses, including morbilliviruses, can impair immunity through various strategies. In this study, it was found that the highly contagious morbillivirus peste des petits ruminants virus (PPRV) can infect monocytes and dendritic cells (DC), leading to immunosuppression. PPRV infection compromised the differentiation and phagocytic ability of monocyte-derived DC, as well as their capacity to activate T cell responses. Furthermore, PPRV-infected DC exhibited an immunosuppressive profile. These findings contribute to our understanding of how morbilliviruses suppress the immune response.
