Summary: This study aimed to explore the risk factors of anastomotic leakage (AL) after laparoscopic anterior resection (AR) of rectal cancer and established a prediction model to assess its occurrence probability. By analyzing the clinical and surgical data of patients, it was found that factors such as male gender, tumor distance from the anus verge, tumor growth pattern, operation time, and whether diverting stoma was performed were associated with the occurrence of AL. The established prediction model based on these factors can effectively predict the probability of AL.

Summary: The study aimed to assess the impact of staple height and rectal wall thickness on the development of an anastomotic leak following laparoscopic low anterior resection. A total of 199 patients treated between 2013 and 2021 were included and divided into two groups based on the presence of an anastomotic leak. Clinicopathological factors were compared between the groups. The results showed that the rate of anastomotic leaks was 4%. Lower staple height (1.5 mm or 1.8 mm) was used more frequently in the leak group, and rectal wall thickness and the rectal wall thickness to staple height ratio were significantly associated with anastomotic leaks. However, rectal wall thickness was significantly increased in patients who received neoadjuvant treatment and those with advanced T stage tumors. In conclusion, linear stapler staple height and rectal wall thickness are closely associated with the development of an anastomotic leak after laparoscopic low anterior resection. Larger staples should be selected for patients with thicker rectal walls.

The results showed that MDST had a significantly lower AL rate compared with DST, suggesting that this technique could be an alternative approach to maximize the benefit of laparoscopic anterior resection on patients with middle and high rectal cancer.

Summary: This study developed a method to handle human liver organoids (HLOs) by overexpressing essential factors in a balanced manner and using conditioned medium as a cost-effective alternative. The researchers controlled the proliferation and maturation of the cells and confirmed the physiological function of HLOs. Efficient gene transduction into HLOs was achieved through a transient 2-dimensional culture. This work provides a fundamental platform for handling HLOs in various research fields.
