Gastric cancer (GC) is one of the leading causes of cancer death in the world. Numerous efforts are being made to find chemoprotective agents able to reduce its risk. Amongst these, green tea has been reported to have a protective effect against stomach cancer. This article aims to critically evaluate all epidemiological studies reporting an association between green tea consumption and GC risk. MEDLINE, EBSCOHOST and Google Scholar were used to search for clinical trials of green tea and its correlation to stomach cancer. Studies include cohort and case-control studies. Outcome of interests are inverse association, no association, and positive association. Seventeen epidemiologic studies were reviewed. Eleven studies were conducted in Japan, five in China, and one with Japanese descendent in Hawaii. Ten case-control studies and seven cohort studies were included. The relative risks or odds ratio of GC for the highest level of green tea consumption was compared. Seven studies suggested no association, eight an inverse association, and one a positive association. One study had shown a significantly lowered GC risk when tea was served warm to cold. Another study also showed a significantly risk with lukewarm tea. All studies that analyzed men and women separately have suggested a reduced risk in women than in men, albeit no significant difference. This review demonstrates that there is insufficient information to support green tea consumption reduces the risk of GC. More studies on the subject matter are warranted. (C) 2013 Baishideng. Novel Strategies for the Bioavailability Augmentation and Efficacy Improvement of Natural Products in Oral Cancer

Alisha Sachdeva, Dimple Dhawan, Gaurav K. Jain, Muekerrem Betuel Yerer, Taylor E. Collignon, Devesh Tewari, Anupam Bishayee

Summary: Oral cancer is a global cause of death, and traditional treatment methods face challenges of cancer recurrence and adverse reactions to chemotherapy drugs. Natural products play a significant role in drug discovery and have potential effects against various cancers, including oral cancer. The limited bioavailability of natural products is a major obstacle to their therapeutic application. Therefore, comprehensive utilization of various formulation strategies to improve bioavailability is important. This work presents recent advancements in novel strategies for natural product delivery that significantly enhance bioavailability and reduce adverse events in the prevention and treatment of oral cancer.

Summary: About one-fourth of the global population is overweight or obese, increasing the risk of insulin resistance, cardiovascular diseases, and infections. Obesity is connected to the severity of COVID-19 and can lead to metabolic disorders. Diabetes is associated with persistent viral infections and affects vaccine effectiveness. Monitoring inflammatory and energy homeostasis markers is important in preventing obesity-related diseases.

Bioactive phytocompounds for oral cancer prevention and treatment: A comprehensive and critical evaluation

Jack T. Burcher, Lindsay K. DeLiberto, Andrea M. Allen, Kaitlyn L. Kilpatrick, Anupam Bishayee

Summary: The high incidence of oral cancer and excessive treatment cost highlight the need for new oral cancer prevention and treatment options. The value of natural agents, including plant secondary metabolites, in preventing cancer and providing a wide range of anticancer drugs has been established. This study provides a comprehensive analysis of current preclinical and clinical results on the use of phytochemicals for oral cancer prevention and treatment. Phytochemicals exert their anticancer effects by modulating various signaling pathways and show promising effects in clinical trials. However, concerns about bioavailability may limit their clinical application. Future research should focus on in-depth mechanistic studies, novel formulations of phytochemicals, adjuvant use with conventional treatment, and randomized clinical trials.

Mechanism of epithelial-mesenchymal transition in cancer and its regulation by natural compounds

Hui Li Ang, Chakrabhavi Dhananjaya Mohan, Muthu K. Shanmugam, Hin Chong Leong, Pooyan Makvandi, Kanchugarakoppal S. Rangappa, Anupam Bishyaee, Alan Prem Kumar, Gautam Sethi

Summary: Epithelial-mesenchymal transition (EMT) is a crucial process in which epithelial cells transform into mesenchymal cells, playing a significant role in tumor progression and resistance to therapy. Various factors, such as signaling pathways, transcriptional regulators, noncoding RNAs, and epigenetic alterations, are involved in the regulation of EMT. However, despite preclinical data, there is a lack of clinical translation in targeting EMT therapeutically. This review discusses the role of these factors in EMT regulation, the contribution of EMT to drug resistance, and potential therapeutic interventions using natural products and nano-formulations as EMT blockers.

Potential of Synthetic and Natural Compounds as Novel Histone Deacetylase Inhibitors for the Treatment of Hematological Malignancies

Dilipkumar Pal, Khushboo Raj, Shyam Sundar Nandi, Surajit Sinha, Abhishek Mishra, Arijit Mondal, Ricardo Lagoa, Jack T. Burcher, Anupam Bishayee

Summary: Histone deacetylases (HDACs) and histone acetyltransferases (HATs) regulate DNA expression by removing or adding acetyl groups to histones, respectively. HDACis have shown promise in treating malignancies by boosting immune response and diminishing tumor immunity. Additional studies are needed to evaluate the efficacy of HDACis, such as anobinostat, vorinostat, and tricostatin A, as therapeutic agents in hematological malignancies.

Summary: It has been found that cancer stem cells exhibit altered metabolism compared to non-CSCs, which plays a crucial role in tumor initiation, progression, immunosuppression, and resistance to conventional therapy. Two natural flavonoids, apigenin and isovitexin, have been shown to enhance the efficacy of conventional chemotherapeutic drugs by sensitizing CSCs. This study aims to critically evaluate the potential of apigenin and isovitexin in targeting CSCs in different cancers as novel therapeutic agents.
