Summary: The use of metformin as a single antitumor agent has been disappointing, but combining it with the NAD biosynthesis inhibitor FK866 has shown superior antitumor activity. This study reveals the mechanism of metformin's antitumor actions and suggests that targeting mitochondria and NAD biosynthesis can lead to effective antitumor therapies.

Summary: This study presents the synthesis of eight novel sulfonamide-based biguanides and evaluates their anti-cancer effects on breast cancer cells. The synthesized compounds showed stronger cytotoxic properties compared to metformin, with compound 2 exhibiting the highest activity. Compound 2 induces apoptosis and mitochondrial dysfunction, arrests cell cycle, and inhibits cancer cell migration. The study concludes that chemical modification of the biguanide scaffold can improve its anti-neoplastic potential.

Summary: The aim of this study was to examine the relationship between pyruvate carboxylase (PC) expression and metabolic reprogramming in pancreatic ductal adenocarcinoma (PDAC). PC protein expression was measured in PDAC and precancerous tissues. The effects of PC expression on PDAC tumorigenesis and metabolic reprogramming were evaluated in vivo and in vitro. The findings suggest that PC upregulation is associated with metabolic reprogramming in PDAC.

Summary: Pancreatic ductal adenocarcinoma is the most common cancer of the exocrine pancreas, but has benefited the least from clinical progress in the last three decades. Despite advances in understanding the molecular alterations and subtypes of the tumor, effective treatment options remain limited. More research is needed to identify new drugs and improve survival rates for PDA patients.

Summary: Evasion from drug-induced apoptosis is a crucial mechanism of cancer treatment resistance. In this study, it was found that inhibiting the transcription factor RUNX1 can unleash the death-inducing potential of NOXA, leading to inhibition of pancreatic ductal adenocarcinoma (PDAC) growth. This discovery provides a new approach to target therapy-resistant PDAC.

Summary: Cancer poses a significant challenge for effective treatment due to its complex mechanism, different stages of progression, and lack of screening and identification procedures. Pancreatic cancer, with a low survival rate of around 5 years, is ranked seventh globally among mortality-associated cancers. Therefore, it is crucial to develop diagnostic procedures for early detection, understand pancreatic cancer mechanisms, and explore therapeutic strategies. This review highlights recent developments in pancreatic cancer progression, mechanisms, and therapeutic approaches, including molecular techniques and biomedicines for effective cancer treatment.

Summary: The cases and deaths related to pancreatic cancer are on the rise globally, posing a significant global health concern. The heterogeneous nature of pancreatic cancer within and between tumors has been identified as a critical factor contributing to its poor prognosis. However, there is a lack of precise management strategies for this complex disease. Subtyping pancreatic tumors based on their molecular characteristics shows promise in improving the accuracy of treatment decisions, but its feasibility and potential flaws need further exploration.

Summary: The effect of metformin (MET) on PRODH/POX-dependent apoptosis was studied in wild-type MCF-7 cells and POX knockdown MCF-7 cells. MET was found to increase prolidase activity and decrease collagen biosynthesis, resulting in an increase in substrate concentration for PRODH/POX-dependent ROS formation and caspases activation.
