Identification of a blood-based biological signature in subjects with psychiatric disorders prior to clinical manifestation

Objectives. To determine whether a molecular signature is present in blood of patients with psychiatric disorders before manifestation of symptoms. Methods. Multiplex immunoassay analyses were carried out using serum obtained from two case-control studies of schizophrenia (n = 75) and bipolar disorder (n = 110) patients and their matched controls. The samples were drawn within 1 month before estimated onset of illness. Results. This led to identification of 20 molecules which were altered in pre-schizophrenia and 14 molecules in pre-bipolar disorder subjects compared to controls. Only two of these molecular changes were identical in both data sets and predictive testing confirmed that the biomarker signatures for pre-schizophrenia and pre-bipolar disorder were dissimilar. Conclusion. The present results suggest that there are distinct serum alterations that occur before clinical manifestation of schizophrenia and bipolar disorder. These findings could lead to development of diagnostic tests to help clinical psychiatrists identify and classify vulnerable patients early in the disease process, allowing for earlier and more effective therapeutic intervention.

Plasma proteomic signature of major depressive episode in the elderly

Induced pluripotent stem cell-derived astrocytes from patients with schizophrenia exhibit an inflammatory phenotype that affects vascularization

Summary: This study examined the proteome, inflammatory responses, and secretome effects of astrocytes derived from schizophrenia patients. The findings suggest that these astrocytes exhibit molecular and functional abnormalities, affecting immune function and vascularization. Reduced expression of NF-kappa B p65 subunit was observed, and the secretion of interleukin 8 (IL-8) was particularly elevated. The conditioned medium from these astrocytes could reduce the diameter of newly grown vessels.

Summary: Visceral adiposity is a risk factor for severe COVID-19, and the degree of adipose tissue infection and cellular response to SARS-CoV-2 depend on the adipose tissue depot and viral lineage. Visceral fat cells are more susceptible to SARS-CoV-2 infection and show higher expression of pro-inflammatory cytokines. Cell death occurs to a similar degree regardless of cell origin or viral lineage.

Protein Succinylation and Malonylation as Potential Biomarkers in Schizophrenia

Summary: Lysine succinylation and malonylation, two protein post-translational modifications, play important roles in protein regulation, glycolysis, and energy metabolism. The precursors of these modifications, succinyl-CoA and malonyl-CoA, are key players in central metabolic processes and are responsive to metabolic stimuli. As mitochondrial dysfunction and metabolic dysregulation are involved in psychiatric illnesses like schizophrenia, these modifications can provide insights into protein regulation and serve as potential biomarkers for disease diagnosis, progression, and treatment. The study analyzed data from shotgun mass spectrometry-based quantitative proteomics to investigate the succinylome and malonylome of postmortem brain tissue from schizophrenia patients, controls, and a schizophrenia chemical model, as well as the human oligodendrocyte precursor cell line MO3.13 with antipsychotics and co-treatments. The results revealed significant changes in the succinylome and malonylome, highlighting the importance of these understudied protein modifications.

Mitochondrial, cell cycle control and neuritogenesis alterations in an iPSC-based neurodevelopmental model for schizophrenia

Summary: Schizophrenia is a severe neurodevelopmental psychiatric disorder affecting around 1% of the global population. Proteomic studies on neural stem cells and neurons derived from induced pluripotent stem cells from schizophrenia patients have revealed differentially expressed proteins related to mitochondrial function, cell cycle control, DNA repair, and neuritogenesis. Functional analysis of neural stem cells also showed alterations in mitochondrial oxygen consumption and higher levels of intracellular reactive oxygen species in schizophrenia-derived cells.
