Drug repurposing of quinine as antiviral against dengue virus infection

Dengue virus (DENV) disease outbreaks continue to develop across the globe with significant associated mortality and economic burden, yet no treatment has been approved to combat this virus. In an attempt to identify novel drug candidates as therapeutics for DENV infection, we evaluated four US Food and Drug Administration (FDA) approved drugs including aminolevullic acid, azelaic acid, mitoxantrone hydrochloride, and quinine sulfate, and tested their ability to inhibit DENV replication using focus-forming unit assay to quantify virus production. Of the four investigated compounds, quinine was found to have the most pronounced anti-DENV activity. Quinine inhibited DENV production of DENV by about 80% compared to untreated controls, while the other three drugs decreased virus production by only about 50%. Moreover, quinine inhibited DENV production of all four serotypes of DENV. Reduction in virus production was documented in three different cell lines of human origin. Quinine significantly inhibited DENV replication by reducing DENV RNA and viral protein synthesis in a dose-dependent manner. In addition, quinine ameliorated expression of genes related to innate immune response. These findings suggest the efficacy of quinine for stimulating antiviral genes to reduce DENV replication. The antiviral activity of quinine observed in this study may have applicability in the development of new drug therapies against DENV.

Discovery and development of labdane-oxindole hybrids as small-molecule inhibitors against chikungunya virus infection

Summary: This study reports a new class of small-molecule CHIKV inhibitors, oxindole-labdanes, which can effectively block the replication of CHIKV with good selectivity. Compound (E)-42 shows high antiviral activity and minimal cytotoxicity to host cells, demonstrating both prophylactic and therapeutic effects. This finding opens up a new avenue for the development of antiviral drugs against CHIKV infection.

MicroRNA 573 Rescues Endothelial Dysfunction during Dengue Virus Infection under PPARγ Regulation

Summary: We need to identify molecular factors that can predict the onset of endothelial dysfunction in dengue patients. Increase in endothelial permeability during severe dengue infections is poorly understood. This study shows that miR-573 regulates endothelial function by reducing endothelial permeability and suppressing the inflammatory response in DENV2-infected endothelial cells, suggesting its potential as a diagnostic and therapeutic agent.

Platelets in the pathogenesis of flavivirus disease

Viruses can interact with platelet receptors and modulate immune responses, and understanding the role of platelets in viral pathogenesis could lead to innovative therapeutic approaches. This review presents the molecular aspects of platelet-flavivirus interactions and their impact on platelet activation, thrombocytopenia, and vascular endothelial leakage. Additionally, it examines the role of platelet-derived factors as biomarkers for predicting disease outcomes, using dengue infection as an example.

Discovery of Novel Andrographolide Derivatives as Antiviral Inhibitors against Human Enterovirus A71

Antiviral Activity of Catechin against Dengue Virus Infection

Summary: Catechin, a natural polyphenol compound, has been evaluated as a potential inhibitor of dengue virus (DENV) infection. It was shown to inhibit a specific stage of the DENV replication cycle and affect viral protein translation. Catechin demonstrated inhibitory effects on all four DENV serotypes and chikungunya virus (CHIKV), suggesting its potential as a starting scaffold for the development of antivirals against DENV infection.

A drug screening process using four compound libraries has identified 121 potential anti-SARS-CoV-2 compounds, with seven shortlisted for further validation. Calcitriol, the active form of vitamin D, exhibits strong antiviral potency against SARS-CoV-2 through modulation of the vitamin D receptor pathway. However, the effects of calcitriol on SARS-CoV-2 infection in K18-hACE2 mice were negligible, indicating the need for further investigation using other animal models.

Chelerythrine chloride inhibits Zika virus infection by targeting the viral NS4B protein

This mutation also leads to increased replication fidelity and reduced pathogenicity. These findings provide a foundation for the development of a safe and effective live attenuated CVA6 vaccine.

Antiviral effects of micafungin against pteropine orthoreovirus, an emerging zoonotic virus carried by bats

Global transcriptomic analysis in avocado nursery trees reveals differential gene expression during asymptomatic infection by avocado sunblotch viroid (ASBVd)

Summary: This study investigated the host gene expression in asymptomatic avocado nursery trees infected with Avocado sunblotch viroid (ASBVd) using RNA sequencing. The results showed that 631 genes were differentially expressed during infection, with 63% upregulated. ASBVd infection significantly affected plant defence responses, phytohormone networks, gene expression pathways, secondary metabolism, cellular transport, as well as protein modification and degradation. This study provides insights into the molecular interactions underlying avsunviroid-host interactions.

Rescue of a Cilevirus from infectious cDNA clones

Summary: This study reports the construction and validation of an infectious cDNA clone of Citrus leprosis virus C (CiLV-C) based on an agroinfection system. Agro-injected Nicotiana benthamiana plants showed localized lesions similar to the natural infection process. The virus recovered from the infected plant tissue could be mechanically transmitted between plants. The CiLV-C reverse genetic system provides a powerful molecular tool for unraveling the peculiarities of the citrus leprosis pathosystem.

Detection of a cluster of Omicron's BA.4 sublineage in Northern Senegal and identification of the first XAS recombinant variant in Senegal

Summary: Genomic surveillance in Senegal since March 2020 has detected the emergence of Omicron BA.4 and BA.5 sublineages in June 2022. Next-generation sequencing and phylogeny analysis of isolates from a cluster of cases in Northern Senegal in July 2022 revealed that the BA.4 cases originated from a XAS recombinant, marking the first reported sequence of this variant from Senegal. Continuous genomic surveillance of positive SARS-CoV-2 samples is crucial.
