Poly (I:C), an agonist of toll-like receptor-3, inhibits replication of the Chikungunya virus in BEAS-2B cells

Background: Double-stranded RNA (dsRNA) and its mimic, polyinosinic acid: polycytidylic acid [Poly (I:C)], are recognized by toll-like receptor 3 (TLR3) and induce interferon (IFN)-beta in many cell types. Poly (I:C) is the most potent IFN inducer. In in vivo mouse studies, intraperitoneal injection of Poly (I:C) elicited IFN-alpha/beta production and natural killer (NK) cells activation. The TLR3 pathway is suggested to contribute to innate immune responses against many viruses, including influenza virus, respiratory syncytial virus, herpes simplex virus 2, and murine cytomegalovirus. In Chikungunya virus (CHIKV) infection, the viruses are cleared within 7-10 days postinfection before adaptive immune responses emerge. The innate immune response is important for CHIKV clearance. Results: The effects of Poly (I:C) on the replication of CHIKV in human bronchial epithelial cells, BEAS-2B, were studied. Poly (I:C) suppressed cytopathic effects (CPE) induced by CHIKV infection in BEAS-2B cells in the presence of Poly (I:C) and inhibited the replication of CHIKV in the cells. The virus titers of Poly (I:C)-treated cells were much lower compared with those of untreated cells. CHIKV infection and Poly (I:C) treatment of BEAS-2B cells induced the production of IFN-beta and increased the expression of anti-viral genes, including IFN-alpha, IFN-beta, MxA, and OAS. Both Poly (I:C) and CHIKV infection upregulate the expression of TLR3 in BEAS-2B cells. Conclusions: CHIKV is sensitive to innate immune response induced by Poly (I:C). The inhibition of CHIKV replication by Poly (I:C) may be through the induction of TLR3, which triggers the production of IFNs and other anti-viral genes. The innate immune response is important to clear CHIKV in infected cells.

Inactivation of SARS-CoV-2 and influenza A virus by dry fogging hypochlorous acid solution and hydrogen peroxide solution

Summary: This study attempted to inactivate SARS-CoV-2 and influenza A virus adhering to environmental surfaces using dry fogging with hypochlorous acid solution and hydrogen peroxide solution. The results showed that SARS-CoV-2 was more resistant to the virucidal effects of these disinfectants than influenza A virus, requiring higher concentrations or longer contact times for inactivation.

Schwarzengrund was frequently detected as multi-antimicrobial resistant and had a high prevalence of virulence genes.

Construction and evaluation of a self-replicative RNA vaccine against SARS-CoV-2 using yellow fever virus replicon

Summary: Replicon RNA (repRNA) vaccine is a promising vaccine platform that can self-replicate and act as an adjuvant, generating robust immunity. This study successfully constructed and evaluated a repRNA vaccine that induced high levels of T-cell response but had minimal impact on antibody response. Future work should focus on optimizing vaccine administration methods and improving S protein expression to enhance the immune-inducing ability of the repRNA vaccine.

Summary: This study focused on developing a Japanese encephalitis (JE) vaccine that does not induce antibody-dependent enhancement (ADE) of dengue virus (DENV) infection. By introducing a mutation at the 106th amino acid residue of the JEV E protein, the modified JE vaccine was able to significantly reduce the production of cross-reactive ADE-prone antibodies against DENV.

Subtype Distribution and Drug Resistance Patterns Among HIV-1 Strains Prevalent in Makassar, Indonesia

Summary: This study investigated the distribution of HIV-1 subtypes and the emergence of drug resistance mutations (DRMs) in Makassar, South Sulawesi, Indonesia. CRF01_AE was identified as the predominant circulating recombinant form (CRF) in this region, along with subtype B and recombinant viruses containing CRF02_AG and/or subtype B gene fragments. DRMs against non-nucleoside reverse transcriptase inhibitors were found among ART-experienced individuals, while no transmitted drug resistance was observed in ART-naive individuals.

HTRA1 Regulates Subclinical Inflammation and Activates Proangiogenic Response in the Retina and Choroid

Summary: In this study, we characterized the phenotypic changes in transgenic mice expressing the HtrA1 gene and found the presence of subretinal white material and scattered white-yellowish retinal foci. Histopathological analysis revealed increased macrophages and RPE cell migration, while indocyanine green angiography showed punctate hyperfluorescent spots in the retina of the CAG-HtrA1 transgenic mice, and increased monocyte subpopulations in peripheral blood. Laser induced CNV demonstrated increased leakage and angiogenesis in the CAG-HtrA1 transgenic mice.
