This study developed an isothermal point-of-care diagnostic test using recombinase polymerase amplification (RPA) and targeting the species-specific apxIVA gene, which achieved rapid and sensitive screening for APP.

Summary: qPCR is a powerful tool for accurate nucleic acid detection and quantification, but current methods for multiplexing are limited to a small number of targets or require expensive instrumentation. This review discusses the use of machine learning algorithms and data-driven solutions to enhance multiplexing in qPCR. The review also explores the use of computer simulation to optimize the design of mPCR assays. The integration of knowledge-based and data-driven software solutions could streamline assay design and improve target detection and quantification in the multiplex setting.

Summary: Researchers summarized the different analytical approaches currently used to study the dissemination of SARS-CoV-2 variants in wastewater and discussed their advantages and disadvantages, while providing preliminary results of two sensitive and cost-effective research methods. Next-generation sequencing approaches allow for simultaneous detection of signature mutations of different variants of concern, whereas targeted PCR methods require continuous updating but are sensitive and cost-effective.
