Signaling pathway alterations are important in the development of gastric cancer (GC). Deregulation of the PI3K/AKT/mTOR pathway plays a crucial role in the regulation of multiple cellular functions including cell growth, proliferation, metabolism, and angiogenesis. Our goal was to assess expression of proteins involved in the PI3K/AKT/mTOR pathway by immunohistochemistry (IHC) in tumor and nontumor gastric mucosa from patients with advanced GC. We evaluated 71 tumor and 71 nontumor gastric mucosa samples from advanced GC patients, selected from Hernan Henriquez Aravena Hospital (Temuco, Chile). The targets studied were PI3K, AKT, p-AKT, PTEN, mTOR, p-mTOR, P70S6K1, p-P70S6K1, 4E-BP1, p-4E-BP1, eIF4E, and p-eIF4E. Expression data were correlated with clinicomorphological data. Descriptive and analytical statistics were used (95 % confidence interval, p < 0.05). For survival analyses, the Kaplan-Meier method and the log-rank test were used. PI3K, AKT, p-AKT, p-mTOR, p-4E-BP1, P70S6K1, p-P70S6K1, eIF-4E, and p-eIF-4E proteins were significantly overexpressed in tumor tissue. Conversely, PTEN was underexpressed in tumor tissue, notably in pT3-pT4 tumors (p = 0.02) and tumors with lymph node metastases (p < 0.001). P70S6K1 expression was associated with pT3-pT4 tumors (p = 0.03). Moreover, PI3K (p = 0.004), AKT (p = 0.01), p-AKT (p = 0.01), P70S6K1 (p = 0.04), p-P70S6K1 (p = 0.001), and eIF-4E (p = 0.004) were overexpressed in tumors with lymph node metastases. Low expression of 4E-BP1 was associated with poor overall survival (p = 0.03). Our results suggest that the PI3K/AKT/mTOR pathway is activated in GC, with overexpression in tumor tissue of most of the studied proteins (total and phosphorylated). These might be considered as target for specific targeted therapy in GC.

Summary: The GB-RADS ultrasound risk stratification system categorizes gallbladder wall thickening into six categories based on features such as symmetry, layered appearance, intramural features, and interface with the liver. This collaborative effort aims to improve the accuracy of detecting malignant lesions and enhance management strategies for gallbladder conditions.

Environmental and Lifestyle Risk Factors in the Carcinogenesis of Gallbladder Cancer

Summary: Gallbladder cancer is an aggressive neoplasm that is often asymptomatic in early stages and is typically diagnosed at an advanced stage. The presence of gallstones is the main risk factor, while other factors such as inflammation can also contribute to the development of gallbladder cancer. Understanding the early carcinogenic mechanisms is crucial for prevention.

Long Non-Coding RNAs (lncRNAs) as Regulators of the PI3K/AKT/mTOR Pathway in Gastric Carcinoma

Ismael Riquelme, Pablo Perez-Moreno, Barbara Mora-Lagos, Carmen Ili, Priscilla Brebi, Juan Carlos Roa

Summary: Gastric cancer is a significant cause of cancer-related deaths, particularly among younger people. Late diagnosis, metastasis, and drug resistance contribute to the high mortality rate of gastric cancer. There is a need for new biomarkers and therapeutic targets. RNA-seq technology has identified long non-coding RNAs (lncRNAs) as potential regulators of cancer cells' signaling pathways, including the PI3K/AKT/mTOR pathway, and they may serve as diagnostic or prognostic biomarkers or therapeutic targets for gastric cancer.

Summary: This study investigated the molecular mechanisms of acquired resistance in gallbladder cancer by developing and characterizing gemcitabine-resistant cell lines. Transcriptome profiling and phosphotyrosine proteomic analyses revealed dysregulated biological processes and signaling pathways, as well as potential therapeutic targets in gallbladder cancer. The findings greatly enhance our understanding of acquired drug resistance in gallbladder cancer.

Summary: This study aimed to investigate the frequency and genetic diversity of EBV in gastric cancer (GC) patients in southern Chile. The study found that 22.2% of the GC samples were EBV-positive and significantly associated with diffuse-type histology. The results provide important information for epidemiological follow-up of different EBV subtypes that may cause GC in southern Chile.

Glioblastoma Microenvironment and Invasiveness: New Insights and Therapeutic Targets

Jose Ignacio Erices, Carolina Bizama, Ignacio Niechi, Daniel Uribe, Arnaldo Rosales, Karen Fabres, Giovanna Navarro-Martinez, Angelo Torres, Rody San Martin, Juan Carlos Roa, Claudia Quezada-Monras

Summary: Glioblastoma (GBM) is a common and malignant brain cancer with poor prognosis. Its low effectiveness is attributed to tumor infiltration into healthy brain tissue, which depends on the interaction between GBM cells and the tumor microenvironment (TME). This review highlights the use of 3D patient-derived glioblastoma organoid cultures as a new platform for studying TME and invasiveness. It discusses the mechanisms involved in GBM-TME interaction and proposes potential prognostic biomarkers and therapeutic targets.

l-Asparaginase as the gold standard in the treatment of acute lymphoblastic leukemia: a comprehensive review

Maria Tosta Perez, Lisandra Herrera Belen, Pablo Letelier, Yolanda Calle, Adalberto Pessoa, Jorge G. Farias

Summary: L-Asparaginase is an approved antileukemic drug for childhood acute lymphoblastic leukemia, but its efficacy and use have been debated due to adverse effects, rapid plasma elimination, multiple administrations, and allergic reactions. Finding less immunogenic variants has been a focus of study. This review presents the aspects of L-asparaginase enzyme from structural, pharmacological, and clinical perspectives in chemotherapy protocols with other drugs.
