PCR survey of vectorborne pathogens in dogs living in and around Barcelona, an area endemic for leishmaniosis

Blood samples from 153 dogs living in and around Barcelona were assayed for Leishmania infantum and Ehrlichia, Anaplasma, Rickettsia, Bartonella, Hepatozoon, Babesia and Theileria species by PCR amplification of DNA, and the amplicons obtained were sequenced. The prevalence of the infectious agents was L infantum (29.4 per cent), Ehrlichia and Anaplasma species (4.0 per cent), Hepatozoon canis (3.3 per cent), Babesia canis vogeli (2.0 per cent), Babesia gibsoni (2.0 per cent), Babesia canis canis (1.3 per cent) and Theileria annae (0.7 per cent). Coinfections were present in seven of the dogs and they were significantly associated with L infantum infection (P=0.024). There was a significant correlation between clinical signs of illness and the load of L infantum.

Summary: This study identified the inter-hospital dissemination of carbapenem-resistant high-risk clones of Klebsiella pneumoniae and Escherichia coli in Catalonia, Spain. The isolates were found to carry small transferable plasmids containing bla(NDM) genes, with most showing multidrug resistance and NDM production. The main clone belonged to ST147 and co-produced NDM-1 and CTX-M-15, with a few isolates also carrying bla(OXA-48).

Long-read metagenomics retrieves complete single-contig bacterial genomes from canine feces

Summary: Long-read sequencing in metagenomics allows for the assembly of complete genomes from complex microbial communities, providing important biological information that is often missed by short-read sequencing. In this study, long-read metagenomics with Nanopore sequencing successfully retrieved eight high-quality metagenome-assembled genomes (HQ MAGs) from a dog fecal sample, including crucial ribosomal genes and tRNAs. Additionally, high-molecular-weight DNA extraction improved assembly contiguity and recovery of rRNA operons, demonstrating the significance of long reads in metagenomics research.

Transmission of Similar Mcr-1 Carrying Plasmids among Different Escherichia coli Lineages Isolated from Livestock and the Farmer

Summary: Colistin, once discontinued in human medicine due to toxicity, is now being used as a last-resort antibiotic for treating hospital infections caused by multi-drug resistant Enterobacteriaceae. A study found that colistin-resistant E. coli isolates carrying mcr-1 were transmitted from livestock to a farmer through horizontal gene transfer. Additionally, virulence factors including Shiga toxin E. coli were detected, emphasizing the role of livestock as a reservoir for potential zoonotic pathogens.

The release of these genomes, including both single-contig complete genomes and genomes with circular main contigs using Nanopore sequencing, provides valuable resources for future research.

Concordance between Antimicrobial Resistance Phenotype and Genotype of Staphylococcus pseudintermedius from Healthy Dogs

Summary: In this study, the correlation between phenotype and genotype regarding antimicrobial resistance of Staphylococcus pseudintermedius from healthy dogs was assessed using Nanopore sequencing technology. Good correlation was observed for some antimicrobials, while correlation was low for others. Multiple antibiotic resistance genes were also found to be located on plasmids. This study provides important insights into the antimicrobial resistance of S. pseudintermedius from healthy dogs.

Real-time genomics for One Health

Summary: The degradation of natural systems and environmental changes have put our society at a crossroad with respect to our future relationship with the planet. The concept of One Health highlights the interdependence of human and environmental health, but many of these connections are not well-understood. This article discusses how real-time genomic analyses, particularly nanopore sequencing, can benefit One Health by enabling timely assessments of ecosystem health, and it showcases studies on zoonotic diseases, food security, environmental microbiome, emerging pathogens, and environmental health. The importance of equitable access to real-time genomics in the context of One Health is emphasized, along with practical, legal, and ethical considerations.

Sphingomyelin-Rich Lipid Extract Collar for Canine Atopic Dermatitis

Summary: Canine atopic dermatitis (CAD) is a chronic skin disease. A new collar called Atopivet (®) collar was developed for CAD, containing a lipid extract (Biosfeen(& REG;)) that has been shown to benefit skin health. Studies showed that this collar effectively released the lipid extract, had good efficacy and safety, and improved clinical symptoms in dogs with CAD. It could be a useful therapy to consider in a multimodal approach to CAD.

Summary: We de novo assembled and polished 61 Staphylococcus pseudintermedius genome sequences using Nanopore-only long reads. The completeness reached 99.25%, with an average genome size of 2.70 Mbp, containing various coding and noncoding sequences, as well as CRISPR arrays.
