Characterization of cytokines associated with Th17 cells in the eyes of horses with recurrent uveitis

Objective Equine recurrent uveitis (ERU) is a spontaneous disease that is the most common cause of blindness in horses, affecting up to 15% of the horse population. Th17 cells are a major cell population driving the pathogenesis in several mouse models of autoimmune inflammation, including experimental autoimmune uveitis. The purpose of this study is to investigate the role a Th17 cell-mediated response plays in the pathogenesis of ERU. Procedure Banked, Davidsons-fixed equine globes histopathologically diagnosed with ERU (n = 7) were compared immunohistochemically with healthy control globes (n = 7). Immunohistochemical staining was performed using a pan-Leptospira antibody and antibodies against IL-6, IL-17, and IL-23. Additionally, immunostaining was performed for T-cell (CD3) and B-cell (CD79a) markers. Specificity of immunoreactivity was confirmed by western blot analysis. Results Immunohistochemical staining was positive for IL-6, IL-17, and IL-23 within the cytoplasm of nonpigmented ciliary epithelial cells and mononuclear inflammatory cells infiltrating the iris, and ciliary body of ERU horses (n = 7) but negative in controls (n = 7). ERU-affected eyes were CD3 positive (n = 7) and CD79a negative (n = 7). Staining for Leptospira was negative in all ERU and control globes. Conclusions Strong immunoreactivity for IL-6, IL-17, and IL-23, in conjunction with the fact that T lymphocytes are the predominating inflammatory cells present in ERU, suggests that IL-17-secreting helper T-cells play a role in the pathogenesis of ERU. These findings suggest that horses with ERU may serve as a naturally occurring animal model for autoimmune uveitis.

Outcome and prognosis for canine appendicular osteosarcoma treated with stereotactic body radiation therapy in 123 dogs

Summary: Canine appendicular osteosarcoma can be effectively treated with stereotactic body radiation therapy (SBRT), improving lameness in most patients and achieving >80% tumor necrosis in 50% of cases for local disease control. While some patients may experience fractures and require amputation after treatment, dogs who undergo salvage amputation have a significantly longer median survival time compared to those who do not. Metastatic disease at the time of treatment does not significantly impact survival time.

Role of Periostin Expression in Canine Osteosarcoma Biology and Clinical Outcome

Summary: Periostin is an important matricellular protein involved in regulating bone, tooth, and cardiac development, but also plays a role in inflammatory diseases and certain cancers. In this study, periostin expression was found to be significantly upregulated in canine osteosarcoma tumors, but not associated with time to metastasis. Tumors with high periostin expression showed activation of pro-tumorigenic pathways, while tumors with low periostin expression showed enhanced anti-tumor immune responses.

Rotator cuff repair using a bioresorbable nanofiber interposition scaffold: a biomechanical and histologic analysis in sheep

Summary: This study evaluated the mechanical, structural, and histologic quality of rotator cuff repairs using an interposition nanofiber scaffold composed of polyglycolic acid-poly-L-lactide-co-epsilon-caprolactone in an acute sheep model. The results demonstrated that using the scaffold enhanced the repair strength and facilitated the formation of a tissue structure similar to the natural shoulder cuff attachment.

Characterizing the molecular and immune landscape of canine bladder cancer

Summary: Transitional cell carcinoma is the most common type of bladder cancer in humans and dogs, with high metastatic risk. Canine transitional cell carcinoma serves as a model for evaluating novel therapies for human muscle-invasive TCC. Molecular characterization revealed a high prevalence of BRAF variants in canine TCC, suggesting potential therapeutic targets for both human and canine cancers.

Summary: This study performed a histological assessment of a biphasic acellular interpositional cancellous allograft in a sheep model of rotator cuff repair (RCR) to evaluate its safety profile and effects on tendon healing after RCR.
