Cyprinid herpesvirus 2 infection emerged in cultured gibel carp, Carassius auratus gibelio in China

An epizootic with severe mortality has emerged in cultured gibel carp, Carassius auratus gibelio, in China since 2009, and caused huge economic loss. The signs and epidemiology background of the disease were investigated. Parasite examination, bacteria and virus isolation were carried out for pathogen isolation. The causative pathogen was obtained and identified as Cyprinid herpesvirus 2 (CyHV-2) by experimental infection, electron microscopy, cell culture, PCR assay and sequence alignment, designated as CyHV-2-JSSY. Experimental infection proved the high virulence of CyHV-2-JSSY to healthy gibel carp. Electron microscopy revealed that the viral nucleocapsid was hexagonal in shape measuring 110-120 nm in diameter with a 170-200 nm envelope. The virus caused significant CPE in Koi-Fin cells at the early passages, but not beyond the fifth passages. Sequence alignment of the partial viral helicase gene (JX566884) showed that it shared 99-100% identity to the published sequences of other CyHV-2 isolates. This study represented the first isolation and identification of CyHV-2 in cultured gibel carp in China and laid a foundation for the further studies of the disease. (C) 2013 Elsevier B.V. The quantitative proteomic analysis of rare minnow, Gobiocypris rarus, infected with virulent and attenuated isolates of grass carp reovirus genotype II

Summary: This study investigated the protein levels in the spleen of rare minnow after infection with different genotypes of GCRV II using iTRAQ-based quantitative proteomics. The results showed that both virulent and attenuated isolates induced similar upregulated proteins related to ATP synthesis, while the virulent genotype induced more immunoproteins and increased levels of ubiquitin-proteasome system members. The DEPs in the attenuated infection group changed more quickly, indicating their sensitivity during virus infection.

Non-pathogenic grass carp reovirus infection leads to both apoptosis and autophagy in a grass carp cell line

Summary: This study investigated the cell death mechanisms involved in grass carp reovirus - HH196 (GCRV-HH196) infection using a grass carp ovary cell line (GCO cells). Results showed that GCRV-HH196 infection caused DNA damage, cell volume reduction, and cytoplasm shrinkage. The expression levels of pro-apoptotic genes were up-regulated and caspase proteins involved in apoptosis were activated. Flow cytometry analysis revealed a higher number of apoptotic cells in GCRV-HH196-infected cells compared to control cells. Additionally, autophagy was also involved in the regulation of GCRV-HH196 infection.

Rag1 and Rag2 Gene Expressions Identify Lymphopoietic Tissues in Larvae of Rice-Field Eel (Monopterus albus)

Summary: In this study, we cloned and sequenced the rag1 and rag2 genes of rice-field eel and investigated their expressions in different tissues. The results showed that multiple tissues of rice-field eel contain lymphocyte lineage cells and undergo lymphopoiesis, with the thymus being the primary lymphopoietic tissue and the liver and kidney also involved in lymphocyte differentiation.

Proteomic and Phosphoproteomic Analyses Reveal Gibel Carp Responses to Cyprinid Herpesvirus 2 Infection

Summary: Cyprinid herpesvirus 2 (CyHV-2) is a double-stranded DNA virus that causes severe herpesviral hematopoietic necrosis disease (HVHND) in gibel carp. This study used proteomic and phosphoproteomic analysis methods to uncover the host response to CyHV-2 infection in the kidneys of symptomatic and diseased gibel carp. The results showed that proteins involved in energy generation and ion balance were downregulated in the host after CyHV-2 infection, and phosphorylated proteins mainly participated in the regulation of RNA processing, translation, cytoskeleton organization, and immunoreaction. Furthermore, potential host kinases that interacted with viral proteins were identified, which could be candidate targets for disease treatment in the future.

Gut mucosal immune responses and protective efficacy of oral yeast Cyprinid herpesvirus 2 (CyHV-2) vaccine in Carassius auratus gibelio

Summary: This study developed an oral vaccine against CyHV-2 using yeast cell surface display technology and demonstrated its protective efficacy in gibel carp. The vaccine group showed higher activities of immune-related enzymes and gene expression, and strong innate and adaptive immune responses were elicited in mucosal and systemic tissues after oral vaccination with the yeast vaccine.

Isolation and identification of Chryseobacterium indologenes and its pathological changes in Pelodiscus sinensis

Summary: In this study, the pathogen causing the death of farmed Pelodiscus sinensis was isolated and identified as Chryseobacterium indologenes. Histopathological analysis revealed the pathological changes in tissues, and antibiotic susceptibility tests showed that the bacteria could be treated with certain antibiotics. These findings contribute to the future diagnosis and treatment of the disease.

Variations in the Intestinal Microbiota of the Chinese Soft-Shelled Turtle (Trionyx sinensis) between Greenhouse and Pond Aquaculture

Summary: In this study, the bacterial community structure of Chinese soft-shelled turtles (SST) and its aquaculture water system were analyzed using Illumina MiSeq sequencing. The results showed that the bacterial diversity and richness compositions in the intestinal tract and aquaculture water were the same, but the relative abundances of bacterial communities varied. The intestinal microorganisms in the SSTs experienced significant changes after transfer to a different environment.
