Genotyping of bovine Prototheca mastitis isolates from Poland

Bovine mastitis due to unicellular, achlorophyllous algae of the genus Prototheca is a serious and complex ailment that accounts for high economic losses in the dairy industry. Bovine protothecal mastitis has been almost exclusively associated with only one species Prototheca zopfii. Recently, based on the 18S rDNA sequence analysis, P. zopfii has been divided into three genotypes (1-3), of which P. zopfii genotype 3 has been given the status of a new species Prototheca blaschkeae sp. nov. The aim of this study was to investigate the genotypic composition of the population of P. zopfii bovine mastitis isolates from Poland, according to that threefold classification. Of the 44 isolates tested, 43(98%) were identified as P. zopfii genotype 2. Only one isolate was assigned to the newly established P. blaschkeae species. These findings clearly show the predominance of the P. zopfii genotype 2 in the etiology of bovine mammary protothecosis in Poland, but also underline the involvement of P. blaschkeae in the disease. The study is the first to provide molecular characterization of Polish P. zopfii mastitis isolates, collected from different regions of Poland. (C) 2010 Elsevier B.V. All rights reserved.

Summary: Invasive candidiasis caused by mixed yeast (MY) infections was studied in a multicenter prospective research, with Europe having the highest MY rate. The most frequent combinations were Candida albicans/C. glabrata and C. albicans/C. parapsilosis. All Candida isolates were susceptible to amphotericin B.

Summary: The study evaluated the use of MALDI-TOF MS for the rapid and accurate identification of three major Prototheca species in mastitis cases. With an extended algae database, MALDI-TOF MS demonstrated the ability to quickly and effectively identify Prototheca species from mastitis cases.

Molecular snapshot of drug-resistant Mycobacterium tuberculosis strains from the Plateau State, Nigeria

Summary: Nigeria has the highest burden of tuberculosis in Africa and ranks 6th globally. However, few studies have examined the molecular epidemiology of Mycobacterium tuberculosis in the country. This study analyzed the genetic structure of drug-resistant tuberculosis in central Nigeria and found that the Cameroon and T lineages were the most prevalent, with no association between the Cameroon lineage and drug resistance.

Summary: The study investigates the occurrence of very rare and poorly studied microalgae of the genus Prototheca, potentially pathogenic to humans and animals, in different water environments. Given the potential hazard to human and animal health from exposure to water-inhabiting protothecae, the prevalence of the algae in aquatic habitats deserves an insightful examination.

Summary: This study investigated the genetic diversity of Mycobacterium tuberculosis isolates in Poland, with a focus on multidrug-resistant strains. The results showed that the Beijing genotype was the most prevalent among MDR-TB cases, especially among foreign-born patients, indicating possible transmission from FSU countries. These findings are important for monitoring and controlling the spread of tuberculosis in Poland.

Intralesional amphotericin B in a cat with cutaneous protothecosis

Summary: A domestic cat was diagnosed with Prototheca wickerhamii infection through cytological and histopathological examination, and culture. Intralesional application of amphotericin B in conjunction with oral terbinafine resulted in a significant reduction of the nasal lesion and complete resolution of the pinnal lesion, without adverse effects.

Summary: The in vitro algaecide activity of quaternary ammonium (QA) against Prototheca isolated from bovine clinical mastitis was investigated. The study highlights the predominance of P. bovis as the causative agent of algal mastitis in bovines, and the strong algaecide activity of QA at low concentrations against Prototheca.

The Development of a Multiplex PCR Assay for Fast and Cost-Effective Identification of the Five Most Significant Pathogenic Prototheca Species

Summary: A multiplex PCR system (m-PCR) has been developed for accurate differentiation of five important pathogenic Prototheca species. The method is cost-effective and uses a simple heat-shock method in a TE buffer for DNA extraction. The method can identify the species within three hours and can detect mixed infections involving two or three Prototheca species.

Cutaneous protothecosis in a dog successfully treated with oral itraconazole in pulse dosing

Summary: This article reports a case of chronic cutaneous protothecosis in a dog in Brazil, successfully treated with long-term pulse therapy with itraconazole. The study suggests that oral itraconazole pulse dosing can effectively control this skin infection.

Prototheca-ID: a web-based application for molecular identification of Prototheca species

Summary: Prototheca is a genus of unicellular, achlorophyllous, yeast-like algae widely distributed in the environment, known to infect vertebrates including humans. Protothecosis can be clinically challenging and treatment-resistant, requiring accurate identification through DNA sequencing. Current limitations in sequence databases make Prototheca identification difficult and inconclusive, with a lack of bioinformatic tools for data processing and analysis. The introduction of Prototheca-ID aims to address these challenges by providing a user-friendly, web-based application for fast and reliable speciation of Prototheca isolates.

Protothecosis in Dogs and Cats-New Research Directions

Summary: Protothecosis is a disease of humans and animals caused by infection with fungus-like, colourless microalgae of the genus Prototheca. While still uncommon, there is an increasing number of diagnosed cases worldwide. This review summarizes new findings in basic science and clinical features, diagnosis and treatment of protothecosis in dogs and cats based on information presented at a medical phycology symposium.
