MicroRNA expression profiles in the progression of prostate cancer-from high-grade prostate intraepithelial neoplasia to metastasis

Introduction: Models of the multistep process related to cancer progression have been designed for many cancers including prostate. The aim of this study is to propose a new model including a possible role for recently described micro RNAs in prostate cancer (CaP) progression. Methods: Sixty-three patients underwent radical prostatectomy to treat localized prostate carcinoma. The specimens of 15 patients were representative of high grade prostate intraepithelial neoplasia (HGPIN). Fourteen specimens represented localized favorable CaP, and 34 unfavorable, mostly non-organ-confined disease. Representing the advanced disease we studied 4 metastatic androgen-independent CaP and 2 cell lines. Micro RNAs were isolated using the mirVana miRNA Isolation kit and cDNA was obtained using the TaqMan miRNA Reverse Transcription kit to the miRNAs: hsa-miR-let7c, hsa-miR-15a, hsa-miR-16, hsa-miR-21, hsa-miR-25, hsa-miR-32, hsa-miR-100, hsa-miR-143, hsa-miR-145, hsa-miR-146a, hsa-miR-191, hsa-miR-199a, hsa-miR-206, and hsa-miR-218. Quantitative RT-PCR was carried out using the ABI 7500 Fast Real-Time PCR System and the TaqMan Universal PCR Master Mix. miRNA expression levels were measured by relative quantification, and fold expression changes were determined by the 2(-Delta Delta CT) method. The small nucleolar RNA RNU43 was used as an endogenous control. Results: Except for miR-21 and miR-206, the expression levels of all miRNAs significantly changed during the progression of CaP. Interestingly, there was a significant global loss of miRNA expression between HGPIN and metastasis at 2 important steps. The first was related to the transition from HGPIN to invasive adenocarcinoma, and the second was related to the transition from localized to metastatic adenocarcinomas. Conclusion: Through the analysis of 14 miRNAs in 4 groups of prostate lesions, which reproduced the progression of CaP, we showed that there is a global loss of miRNA expression at 2 distinct steps. The first related to the transition between HGPIN and localized invasive carcinoma, and the second associated with the transition from localized to metastatic CaP. The importance of our study is in the identification of possible miRNAs and miRNA-targeted genes involved in the progression of prostate carcinogenesis that may help the development of potential diagnostic or prognostic markers as well as the design of new target therapies. Summary: The study found that the AR gene was downregulated in the PC3 cell line, and X chromosome polysomy was present in both PC3 and LNCaP cell lines. HUMARA-Q assay revealed that both X chromosomes in PC3 cells exhibited AR activity, while only one AR gene was methylated in LNCaP cells.

Feeding difficulties in children fed a cows' milk elimination diet

Summary: A cross-sectional study compared feeding difficulties in children aged 2-5 years on a cows' milk elimination diet due to food allergy with a control group on an unrestricted diet. The study found that children on the elimination diet had higher frequency of picky eating and feeding problems. Picky eating was associated with lower weight-for-age z-scores. Food refusal and/or inappetence as manifestations of food allergy were linked to feeding difficulties, while current constipation and anticipatory gagging were associated with picky eating, avoidant eating, and feeding problems in both groups.

Ultrasound evaluation of uterine scar thickness after open fetal surgery for myelomeningocele

Summary: This study aimed to analyze the evolution of uterine scar thickness after open fetal surgery for myelomeningocele and establish a cut-off point for uterine scar thickness associated with high risk of uterine rupture. The results showed that thinner uterine scars were significantly associated with increased likelihood of thinning during pregnancy follow-up. A cut-off point of ≤3.0 mm in scar thickness may be useful for decision-making regarding the risk of uterine rupture and indication of caesarean section.

Summary: There is evidence that cribriform morphology is associated with a worse prognosis for prostatic adenocarcinoma. This study aimed to investigate the reproducibility of cribriform cancer diagnosis in prostate needle biopsies. A panel of nine prostate pathology experts independently reviewed 304 digitised biopsies and achieved a 2/3 consensus diagnosis of cribriform and noncribriform cancer in 90% of the cases. Strict diagnostic criteria were used to identify cribriform patterns and generate a set of consensus cases for standardization.

Factors associated with unrecovered falls among older adults

Summary: This cross-sectional exploratory study investigated factors associated with unrecovered falls among older patients with a history of falls in the previous year. Depressive symptoms, ADL/IADL limitation, mobility limitation, under-nutrition, and outdoor falls were found to be associated with unrecovered falls. Professionals should consider preventive strategies and preparedness procedures for those who are more likely to experience unrecovered falls.

Summary: This study aimed to evaluate the suppressive effect of miR-137 in prostate cancer, regardless of diet-induced hypercholesterolemia. The results showed that miR-137 could inhibit the transcription and translation of the p160 family, SRC-1, SRC-2, and SRC-3, and indirectly reduce the expression of AR. It also inhibited cell migration and invasion, reduced proliferation, and increased apoptosis rates. In vivo experiments demonstrated that tumor growth was arrested after intratumoral restoration of miR-137, with a more significant response in the hypercholesterolemic group.

LncRNA JHDM1D-AS1 Is a Key Biomarker for Progression and Modulation of Gemcitabine Sensitivity in Bladder Cancer Cells

Summary: Long non-coding RNAs are frequently dysregulated and have been associated with carcinogenesis, aggressiveness, and chemoresistance in tumors. In this study, we investigated the combined expression of JHDM1D and JHDM1D-AS1 in bladder tumors, and found that it can distinguish between low-grade and high-grade tumors. We also discovered that silencing JHDM1D-AS1 can reduce the growth and proliferation of high-grade bladder tumor cells and increase their sensitivity to gemcitabine treatment.

Initial Validation of the Brazilian Mindful Eating Scale Among Overweight and Low-Income Women

Summary: The objective of this study was to validate the Brazilian Mindful Eating Scale (MES-B), which assesses mindfulness in eating behavior. A total of 236 overweight or obese Brazilian women participated in the study. Through confirmatory factor analysis and reliability analysis, the MES-B was found to have 24 items and six domains. The scale was initially validated but further evaluation in different genders is necessary.

Underreporting of unfavorable outcomes of congenital syphilis on the Notifiable Health Conditions Information System in the state of Sao Paulo, Brazil, 2007-2018

Summary: The objective of this study was to describe the frequency of underreporting of unfavorable outcomes of congenital syphilis in the state of Sao Paulo, Brazil, from 2007 to 2018. Through probabilistic and deterministic linkage methods, the study analyzed cases of abortion, fetal and non-fetal deaths due to congenital syphilis reported on the Notifiable Health Conditions Information System (SINAN) and on the Mortality Information System (SIM). The results showed an increase in unfavorable outcomes, with a 11.4% increase in infant deaths due to congenital syphilis and a 3.0% increase in fetal deaths.
