Serum levels and tissue expression of matrix metalloproteinase 2 (MMP-2) and tissue inhibitor of metalloproteinases 2 (TIMP-2) in colorectal cancer patients

The objective of the study was the assessment of serum levels and tissue expression of matrix metalloproteinase 2 (MMP-2) and tissue inhibitor of matrix metalloproteinases 2 (TIMP-2) in patients with colorectal cancer (CRC). The study included 72 CRC patients and 68 healthy subjects. The serum levels of MMP-2 and TIMP-2 were measured using enzyme-linked immunosorbent assay (ELISA) method, whereas tissue expression of MMP-2 and TIMP-2 in cancer cells, interstitial inflammatory cells, and adjacent normal colorectal mucosa were examined by immunohistochemical staining of tumor samples. The serum levels of MMP-2 and TIMP-2 in cancer patients were significantly lower than those in control group, but the percentage of positive immunoreactivity of these proteins were higher in malignant and inflammatory cells as compared to normal tissue. There was a significant correlation between MMP-2 immunoreactivity in inflammatory cells and the presence of distant metastases and between TIMP-2 expression in inflammatory cells and tumor size, nodal involvement, and distant metastases. Area under receiver operating characteristic (ROC) curve (AUC) for serum MMP-2 was higher than for serum TIMP-2. Moreover, positive tissue expression of MMP-2 was a significant prognostic factor for CRC patients' survival. Our findings suggest that MMP-2 and TIMP-2 might play a role in the process of colorectal cancer invasion and metastasis, but the significance of their interactions with tumor stroma and interstitial inflammatory infiltration in colorectal neoplasia require further elucidation.

Alzheimer's Disease-Biochemical and Psychological Background for Diagnosis and Treatment

Beneficial Proapoptotic Effect of Heterobasidion Annosum Extract in Colorectal Cancer Xenograft Mouse Model

Summary: Fungal extract from Heterobasidion annosum (HA) has been evaluated for its potential anticancer activity in colorectal cancer. In an in vivo model, mice with implanted DLD-1 human cancer cells were treated with HA extract, the drug 5-fluorouracil (5FU), or a combination of both. Results showed that the combination treatment led to the smallest tumor volumes, with increased concentrations of apoptotic proteins caspase-8 and p53, and decreased levels of survivin and Bcl-2. The Ki-67 protein was found to be expressed in all tested groups. These findings provide innovative insights into the antitumor activity of HA extract and its interaction with chemotherapeutic agents.

The caspase-8 and procaspase-3 expression in gastric cancer and non-cancer mucosa in relation to clinico-morphological factors and some apoptosis-associated proteins

Summary: The aim of this study was to assess the expression of caspase-8 and procaspase-3 proteins in gastric cancer cells and non-cancerous mucosa, and its relationship with clinical and morphological characteristics of the tumor, postoperative survival, and other apoptosis-related proteins. The results showed that procaspase-3 was expressed in all gastric cancer cells, with a significant difference in expression compared to non-cancerous mucosa. Caspase-8 expression was more common in certain types of gastric cancer and was associated with longer survival and better prognosis.

Summary: This review examines the impact of COVID-19 on patients with autoimmunological disorder, specifically those with multiple sclerosis (MS). It found that MS patients have a similar risk, symptoms, and mortality rate compared to the general population when infected with COVID-19. Factors such as comorbidities, male sex, higher disability level, and older age contribute to a more severe course of COVID-19 in MS patients. Additionally, the use of disease-modifying therapies may affect the immune response to SARS-CoV-2 infection and vaccination.

The Clinical Significance of Serum Free Light Chains in Bladder Cancer

Summary: This study aimed to assess the clinical usefulness of serum kappa (?) and lambda (?) free light chains (FLCs) in patients with bladder cancer. The concentrations of FLCs, CEA, and CA19-9 were significantly higher in bladder cancer patients compared to the control group. There was also a correlation between FLC and other biomarkers. The findings suggest that FLC measurements may be useful in the diagnosis and prognosis of bladder cancer.

E-Cadherin Expression Varies Depending on the Location within the Primary Tumor and Is Higher in Colorectal Cancer with Lymphoid Follicles

Summary: Different distributions of E-cadherin expression were observed within colorectal cancer, with the highest percentage of positive expression in the invasive front and tumor center. The expression pattern varied depending on cancer progression and other clinical and histopathological features. Low E-cadherin expression appears to be a reliable indicator of higher cancer staging and progression.

Diagnostic significance and utility of circulating redox biomarkers in patients with gastric cancer - preliminary study

Molecular Aspects of a Diet as a New Pathway in the Prevention and Treatment of Alzheimer's Disease

Summary: Alzheimer's disease is the most common cause of dementia worldwide. The lack of established pathology makes it challenging to find suitable approaches for treatment. In addition to the known hallmarks of the disease, such as amyloid β peptides and hyperphosphorylated tau, inflammation and diet also play important roles. Adhering to a beneficial diet high in neuroprotective foods and BDNF-boosting substances may improve memory, cognition, and biomarkers, and even reduce the risk of developing Alzheimer's disease.

Higher Content but No Specific Activity in Gelatinase B (MMP-9) Compared with Gelatinase A (MMP-2) in Human Renal Carcinoma

Summary: Gelatinases A and B, also known as matrix metalloproteinase-2 and matrix metalloproteinase-9 respectively, play a crucial role in breaking down collagen in the extracellular matrix. This study examined the presence, activity, and expression of these gelatinases in human renal cancer. The results showed that MMP-9 had higher content but lower specific activity compared to MMP-2 in renal cancer. The study also revealed changes in the gelatinases in healthy and cancerous tissues of renal cell carcinoma, suggesting the potential of MMPs in the diagnosis and treatment of renal carcinoma.

Summary: Subfertility is a global health issue, and a significant proportion of cases remain unexplained. This study found that unexplained subfertility may be associated with dysregulation of gut microbiota, metabolic abnormalities, and immune dysfunction. One-time laboratory diagnostics including lipid profile, insulin resistance, anti-thyroid peroxidase, and total IgE levels may provide a comprehensive assessment for individuals and couples with unexplained subfertility, allowing for targeted medical and nutritional interventions to optimize patients' health.
