Pathogenic Mechanisms in Chronic Obstructive Pulmonary Disease Due to Biomass Smoke Exposure

Chronic obstructive pulmonary disease (COPD) mortality and Morbidity have increased significantly worldwide in recent decades. Although cigarette smoke is still considered the main risk factor for the development of the disease, estimates suggest that between 25% and 33% of COPD patients are nonsmokers. Among the factors that may increase the risk of developing COPD, biomass smoke has been proposed as one of the most important, affecting especially women and children in developing countries. Despite the epidemiological evidence linking exposure to biomass smoke with adverse health effects, the specific cellular and molecular mechanisms by which this pollutant can be harmful for the respiratory and cardiovascular systems remain unclear. In this article we review the main pathogenic mechanisms proposed to date that make biomass smoke one of the major risk factors for COPD. (C) 2014 SEPAR. Published by Elsevier Espana, S.L.U. Summary: The study found causal effects of gallstone disease on GBC risk in Chileans and Europeans, and also a causal effect of BMI on GBC risk in Chileans, while higher CRP concentrations increased GBC risk in Europeans. Furthermore, BMI showed a causal effect on gallstone risk in Europeans, which was itself causally linked to GBC risk.

Is adherence to the Mediterranean diet associated with healthy habits and physical fitness? A systematic review and meta-analysis including 565 421 youths

Summary: A meta-analysis study on children and adolescents showed that adherence to the Mediterranean diet was positively associated with physical activity, cardiorespiratory fitness, and muscular fitness, while it was negatively associated with sedentary behavior and speed-agility. However, the associations remained significant only for physical activity when controlling for sex and age.

Summary: The onset of late onset Alzheimer's disease (LOAD) may be related to peripheral stress before entering the brain and causing neuroinflammatory brain disease. Preservation of dendritic spines and synaptic plasticity is crucial for preventative or disease modifying therapies, while systemic/environmental stressors impact pathways associated with spine morphogenesis and synaptic maintenance. Targeting these stressors with a combination of drugs may modify the course of LOAD by preventing dendritic spines and synapse loss.

Summary: This study investigates the relationship between genotype-based expression of circulating lncRNAs and the risk of gallbladder cancer (GBC). The results suggest that preselection of lncRNAs and utilization of cis-lncRNA-eQTLs can facilitate the identification of circulating noncoding RNAs linked to cancer risk.

Summary: This study induced chronic metabolic stress by feeding a high-fat diet and studied the effects of JNK1 and JNK3 loss of function on cognition. The results showed that JNK3 loss led to increased body weight and decreased cognitive capacity, while JNK1 loss promoted normal or heightened energetic metabolism and preserved cognitive function.

Decreased Expression of EC-SOD and Fibulin-5 in Alveolar Walls of Lungs From COPD Patients

Summary: This study analyzed the expression of the main oxidant scavenger superoxide dismutase (EC-SOD), its main binding protein Fibulin-5, and several oxidative and nitrosative-derived products in the lung of COPD patients and controls. The results showed that COPD patients have reduced expression of EC-SOD and fibulin-5 in the lung interstitium, supporting the hypothesis of an impaired pulmonary antioxidant response in COPD patients.

Summary: Peroxisome proliferator-activated receptor beta/delta (PPAR beta/delta) plays a key role in microglial homeostasis and metabolism in the central nervous system. However, its contribution to neuronal and memory processes is still not well-understood. This study aimed to evaluate the role of PPAR beta/delta in neuropathological pathways associated with memory impairment and the modulation of these pathways by obesity. The results showed that PPAR beta/delta deficiency affects neuronal and synaptic structure, leading to memory dysfunction.

Summary: PTP1B is a potential target for the treatment of neurological diseases, as its activation in brain cells can lead to various neuronal deficits. However, inhibition of PTP1B has shown promising therapeutic effects and potential to improve cognitive processes. Clinical trials evaluating the neurological benefits of PTP1B inhibition are still needed.

Pharmacogenetic Variation and Its Clinical Relevance in a Latin American Rural Population

Summary: Latin-American populations have been underrepresented in genomic studies of drug response and disease susceptibility. This study compared the frequency of gene variants involved in drug response among a Chilean population and others using the 1000 Genomes Project data. Several gene variants were found to have low prevalence in Chileans compared to other populations, and two markers showed significant differences between different Mapuche ancestry groups. These findings emphasize the importance of considering ancestry in pharmacogenetic studies and provide a pharmacogenetic landscape of an understudied Latin American population.

A novel rhein-huprine hybrid ameliorates disease-modifying properties in preclinical mice model of Alzheimer's disease exacerbated with high fat diet

Summary: This study evaluated the effects of a synthesized inhibitor called RHE-HUP on APP/PS1 mice, a familial AD mouse model challenged by high-fat diet to simulate a T2DM-like condition. Results showed that RHE-HUP reduced the main hallmarks of AD, including Tau hyperphosphorylation, A beta(42) peptide levels, and plaque formation. Additionally, it decreased inflammation, increased synaptic proteins and neurotrophic factors, leading to memory improvement. Importantly, these improvements were observed at a central level without peripheral modifications induced by HFD consumption.

Summary: There is a strong association between the proportion of Mapuche ancestry and the risk of gallbladder cancer (GBC). Mapuche ancestry has a putatively causal effect on GBC risk and gallstone disease, while showing a negative effect on BMI. These findings have significant implications for GBC prevention and future admixture mapping studies.

Summary: The increasing population ageing and growth have led to a rise in the number of people living with dementia, with Alzheimer's disease being the most common cause. Although a cure for Alzheimer's disease has not been found yet, there are proposed treatments that may alter disease progression and help manage symptoms. In addition to the traditional hypotheses of Alzheimer's disease etiopathogenesis involving amyloid beta peptide accumulation and tau hyperphosphorylation, there is a trend that emphasizes the role of other molecular mechanisms, prompting the study of different therapeutic targets.
