Novel roles for Nogo receptor in inflammation and disease

The Nogo receptor (NgR), which was identified as a common receptor for three axon growth inhibitors associated with myelin, has been extensively characterized for its role in triggering growth cone collapse and arresting neurite/axon growth. Recent studies indicate that NgR is also expressed in nonneuronal cells and modulates macrophage responses during inflammation after peripheral nerve injury. In this article, we discuss the possibility that NgR might have wider effects on inflammation in a variety of neurological conditions ranging from central nervous system trauma to diseases such as multiple sclerosis or Alzheimer's disease.

Further experiments demonstrated that the therapeutic effects of IL-37 protein rely on the presence of the extracellular receptor IL-1R8, emphasizing the importance of IL-37 extracellular signaling in mediating neuroprotective actions after SCI.

These beneficial effects are attributed to a paracrine protective mechanism from the muscle to the motoneurons mediated by GDNF, with no adverse secondary effects detected, highlighting the potential for ALS therapy.

Summary: This study reveals that there is an imbalance in the production of specialized pro-resolving mediators (SPMs) in patients with multiple sclerosis (MS) and in mice with experimental autoimmune encephalomyelitis (EAE), and increasing the bioavailability of SPMs, such as Maresin-1 (MaR1), can reduce inflammation and mediate therapeutic effects. These findings suggest that immunoresolvent therapies could be a novel approach for the treatment of MS.

Summary: Spinal cord injuries permanently disrupt spinal connectivity and cause neurological disabilities. Current treatments are limited, so inhibiting the LPA(2) receptor has emerged as a potential therapeutic strategy for reducing SCI-associated damage. A new series of LPA(2) antagonists, including compound 54 (UCM-14216), showed efficacy in an in vivo mouse model, confirming the potential of LPA(2) inhibition for treating SCI.

This metabolic shift is important for minimizing cytotoxic responses of microglia and macrophages.

Administration of recombinant human IL-37 exerts therapeutic actions in EAE mice, but IL-37 transcripts are not up-regulated in peripheral blood mononuclear cells and brain lesions of MS patients.
