Atlastin is an integral membrane GTPase localized to the endoplasmic reticulum (ER). In vitro and in vivo analyses indicate that atlastin is a membrane fusogen capable of driving membrane fusion, suggesting a role in ER structure and maintenance. Interestingly, mutations in the human atlastin-1 gene, SPG3A, cause a form of autosomal dominant hereditary spastic paraplegia (HSP). The etiology of HSP is unclear, but two predominant forms of the disorder are caused by mutant proteins that affect ER structure, formation and maintenance in motor neurons. In this review, we describe the current knowledge about the molecular mechanism of atlastin function and its potential role in HSP. Greater understanding of the function of atlastin and associated proteins should provide important insight into normal ER biogenesis and maintenance, as well as the pathology of disease.

Summary: The phi C31 integrase system is commonly used for transgene targeting in Drosophila melanogaster. AttP40, one of the popular docking sites, has been found to cause larval muscle nuclear clustering and decreased msp-300 transcript levels in third instar larvae. Furthermore, certain transgenic RNAi insertions into attP40 can lead to pupal or adult inviability, infertility, or dominant nuclear clustering effects in specific genetic backgrounds. These findings highlight the importance of considering the effects of attP40 and its insertion derivatives in data interpretation from attP40-bearing flies.

Summary: This study using a Drosophila model found that the absence of atlastin protein leads to muscle issues and affects the autophagy pathway and MTOR signaling. Therefore, the research suggests that mutations in the ATL1/SPG3A gene may cause pathological changes in patients with HSP.
