Retrotransposon-based molecular markers for grapevine species and cultivars identification

Insertional polymorphisms of two copia-like (Vine-1, Tvv1) and one gypsy-like (Gret1) retrotransposon found in the grapevine genome were studied in 29 Vitis genotypes (Vitis arizonica, Vitis cinerea, Vitis labrusca, Vitis rupestis, Vitis rotundifolia, Vitis vinifera subsp. sylvestris and 23 V. vinifera subsp. sativa) using inter-retrotransposon amplified polymorphism (IRAP), retrotransposon-microsatellite amplified polymorphism (REMAP) and sequence-specific amplified polymorphism (SSAP) techniques. IRAP, REMAP and SSAP polymorphisms were compared with amplified fragment length polymorphism (AFLP), Inter-single sequence repeats (ISSR) and SSR polymorphisms by evaluating the information content, the number of loci simultaneously analysed per experiment, the effectiveness of the analyses in assessing the relationship between accessions and the number of loci needed to obtain a coefficient of variation of 10%. The UPGMA dendrograms of each molecular marker system were compared and the Mantel matrix correspondence test was applied. Furthermore, the corresponding insertion ages of the transposable elements were estimated for each retrotransposon subfamily analysed. The presence of Gret1, Tvv1 and Vine-1 retrotransposons in all analysed genotypes suggests that copia-like and gypsy-like retrotransposons are widespread in Vitis genus. The results indicate that these retrotransposons were active before Vitis speciation and contributed to Vitis genus evolution. IRAP, REMAP and SSAP markers allow the discrimination of Vitis species and V. vinifera subsp. sativa cultivars with certainty as has been shown with AFLP, ISSR and SSR analyses, but phylogenetic trees obtained by retrotransposon-based molecular markers polymorphisms show some significant differences in the allocation of the analysed accessions compare to those obtained by ISSR, AFLP and SSR molecular markers. The phylogenetic tree resulting from REMAP polymorphism appeared the most representative of the effective relationship between all analysed accessions.

Management of high-quality dehydrated grape in vinification to produce dry red wines

The effect of regulated deficit irrigation on growth, yield, and berry quality of grapevines (cv. Sangiovese) grafted on rootstocks with different resistance to water deficit

Summary: The aim of this study was to investigate the effect of different irrigation regimes on the growth, yield, and berry quality of container-grown Sangiovese grapes grafted onto different rootstocks. The results showed that deficit irrigation significantly influenced vine water status and had a marked effect on berry and juice quality. The timing of water restriction also played a role, with early water limitation affecting trunk diameter while post-veraison water deficit had no effect. Yield was not affected by irrigation or rootstock. The study concluded that deficit irrigation could be used to manipulate berry quality in grapevines.

Deficit irrigation differently affects aroma composition in berries of Vitis vinifera L. (cvs Sangiovese and Merlot) grafted on two rootstocks

Physiological and Transcriptomic Evaluation of Drought Effect on Own-Rooted and Grafted Grapevine Rootstock (1103P and 101-14MGt)

Summary: The response of grapevines to drought is influenced by the interaction between the scion variety and the rootstock genotype. Under severe water deficit conditions, the rootstock genotype has a greater impact on gas exchange and stem water potential. The 1103P shows avoidance behavior, while the 101-14MGt exhibits a tolerance strategy. Transcriptomic analysis reveals that most differentially expressed genes are detected in roots, with a core set of genes related to the root response to drought being unaffected by genotype or grafting.

Genetic Diversity and Population Structure of Common Bean (Phaseolus vulgaris L.) Landraces in the Lazio Region of Italy

Summary: A study was conducted to characterize 114 accessions of 66 landraces of common beans in the Lazio region. The genetic analysis showed that the P. vulgaris germplasm in the region has a high level of genetic diversity, with the Andean gene pool prevailing and 12% of landraces representing putative hybrids. The combination of morphological, biochemical, and molecular data allowed for the differentiation of all landraces and the resolution of certain cases of homonymy and synonymy.

Combined effects of rootstock and irrigation on gas exchange, fruit quality and yield in 'Merlot' and 'Sangiovese' grapevines

Summary: The combination of rootstock and scion is important for grapevine tolerance to water stress. In an experiment on potted grapevines, different irrigation regimes and rootstocks were evaluated for their effects on leaf gas exchange, fruit quality, and yield. The results showed significant differences in stomatal conductance, net photosynthetic rate, and yield among the different combinations.

Tolerance and physiological response of young Ficus carica L. plants irrigated with saline water

Summary: This experiment evaluated the effect of saline water on 2-year-old potted fig plants. The results showed that saline water significantly affected plant water status, leaf gas exchange, and fruit growth. Differences between treatments were evident at different time points, especially in stem water potential and intrinsic water use efficiency. Additionally, there were differences between the two fig cultivars in some parameters.

Rootstock and irrigation modify berry aroma characteristics in 'Merlot' and 'Sangiovese' grapevines
