Summary: Acute kidney injury (AKI) is a common and devastating condition with no effective pharmacological treatment. Toll-like receptor 4 (TLR4) and its ligands play a crucial role in the pathogenesis of AKI by mediating inflammatory responses. Excessive activation of TLR4 can lead to chronic inflammation. This review discusses the role of TLR4 in ischemic/reperfusion-induced AKI and sepsis-associated AKI, as well as its potential signaling patterns in kidney injury.

Summary: This study investigated the effects of MAGL inhibitor KML29 and FAAH inhibitor URB597 on kidney ischemia-reperfusion injury. The results showed that administration of KML29 and URB597 can reduce kidney damage and inflammation caused by IR. The study suggests that inhibition of MAGL and FAAH may be a new therapeutic strategy for preventing kidney IR injury.

Summary: This study aimed to investigate whether a novel Toll-like receptor 4 agonist could attenuate acute kidney injury induced by ischemia-reperfusion injury. The results showed that pretreatment with the agonist improved kidney function, reduced renal tissue injury and apoptosis. The findings suggest that the agonist can provide protection against ischemia-reperfusion injury-induced acute kidney injury.

Summary: This study aimed to evaluate the effect of Eprosartan, an AT-1 antagonist, on kidney I/R injury in rats. Results showed that Eprosartan significantly improved kidney function and increased antioxidant markers. Eprosartan exhibited anti-inflammatory activity by downregulating NF-kappa B and pro-inflammatory factors. It also prevented I/R-induced changes in apoptotic parameters. Additionally, Eprosartan + I/R rats showed higher levels of Sirtuin 1. In conclusion, Eprosartan has nephroprotective effects against I/R-induced kidney damage in rats by reducing oxidative stress, inflammation, and apoptosis, as well as increasing Sirtuin 1 levels.

They are able to target multiple pathways, acting as antioxidants, anti-inflammatory agents, and immune response and cell death modulators. This study investigated the effect of a nutraceutical solution on liver ischemia-reperfusion injury (IRI), and found that it decreased apoptosis and histologic injury in liver tissue.

Summary: A novel drug delivery system has been developed to precisely deliver drugs to pathological renal tissue for attenuating renal ischemia-reperfusion injury. The system exhibits responsive properties to reactive oxygen species, biocompatibility, and cell penetration. The intracellular concentration of hydrogen sulfide released by the system is threefold higher than that in the control group, and it demonstrates anti-inflammatory effects by blocking the NF-KB signaling pathway. This research provides a promising strategy for pharmaceutical treatment of renal ischemia-reperfusion injury.
