Hepatic Stellate Cells Preferentially Expand Allogeneic CD4+CD25+FoxP3+ Regulatory T Cells in an IL-2-Dependent Manner

Background. Organ transplantation has been successfully practiced for decades, but the outcome of cell transplantation remains disappointing. This is the case in animal models; liver allografts in mice are spontaneously accepted without requirement of immunosuppression, whereas hepatocyte transplants in the same combination are acutely rejected, apparently resulting from immune attacks because syngeneic hepatocyte transplants survive indefinitely. This suggests that liver nonparenchymal cells play an important role in protecting parenchymal cell from rejection. We have shown that hepatic stellate cells (HpSC), well known to participate in liver repairing and fibrosis, mediate potent immunomodulatory functions through induction of activated T-cell death. Methods and Results. Here, we report that HpSC acquired antigen presenting capacity after activated by interferon-gamma. in contrast to professional antigen-presenting cells dendritic cells that predominantly stimulated CD4(+) T cells to generate CD25(+) forkhead box P3 (Foxp3)(-) effector cells, HpSC selectively expanded CD4(+)CD25(+)Foxp3(+) cells in an interleukin-2-dependent manner. These expanded CD4(+)CD25(+)Foxp3(+) cells showed T regulatory cell (Treg) activity in effectively inhibiting T-cell proliferation in responses to anti-CD3 monoclonal antibody or alloantigens in a major histocompatibility complex nonspecific fashion. The Treg cells were expanded from the CD4(+)CD25(+) population with the help of interleukin-2, independent of B7-H1 and transforming growth factor-P. Administration of HpSC into allogeneic recipients resulted in expansion of CD4(+)CD25(+)FoXP3(+) cells in vivo. Conclusion. Liver stromal HpSC acted as nonprofessional antigen -presenting cells, and preferentially expanded CD25(+)FoxP3(+) Treg cells, which may contribute to immune regulation in the liver.

Summary: This study investigated the safety and efficacy of stereotactic body radiation therapy (SBRT) with immunotherapy in patients with hepatocellular carcinoma (HCC). The results showed that the combination of SBRT with nivolumab plus ipilimumab had better survival and efficacy compared to immunotherapy alone in HCC patients, suggesting the need for further investigation.

An edge-based augmented system approach for second-order nodal consensus

Summary: In this paper, an edge-based augmented system approach is proposed for second-order nodal consensus, utilizing the edge loop transfer matrix of the closed-loop multi-agent system. This approach introduces additional poles and zeros into edges to construct a channel filter with previous data stored in the memories. The effectiveness of the proposed method is verified through illustrative simulations.

Edge-Preserving Consensus via Non-Recursive Filters: A Parallel System Design

Summary: This study focuses on the problem of edge-preserving consensus in discrete-time multi-agent systems. A method for designing channel filters, consisting of nominal elements and masking elements, is proposed using an edge-based parallel system design approach. Design criteria for edge-preserving protocols are provided based on the edge sensitivity matrix of the multi-agent system being studied. It is demonstrated that the design of masking elements is based on a single parameter of the nominal element and the H-infinity norm of a collapsed system. An algorithm for designing heterogeneous channel filters is also presented, using a cascade realization of several subsystems.

Fixed-Time Algorithms for Time-Varying Convex Optimization

Summary: In this paper, a novel type of fixed-time algorithms is investigated to solve time-varying convex optimization problems with time-dependent cost function, constraints or both. First, a general framework algorithm is developed for the unconstrained time-varying optimization problem to track its optimal trajectory within fixed time, which includes the gradient flow-based scheme and Newton-type method as special cases. Then, another algorithm with fixed-time convergence is designed for time-varying optimization problems involving equality constraint, including Newton-type scheme as a special case. The simulation result using first-order Euler discretization is provided to verify the fixed-time convergence achieved by the proposed approach.

Detecting faulty edges of complex dynamical networks based on compressive sensing

Retinoblastoma Expression and Targeting by CDK4/6 Inhibitors in Small Cell Lung Cancer

Summary: Genomic studies have found that some small cell lung cancer (SCLC) tumors do not have RB1 mutations, contrary to the canonical model. This study examined RB1 protein expression and its function in SCLC and found that 14% of SCLC tumors expressed abundant RB1 protein, which was associated with neuroendocrine gene expression and enriched in YAP1 expression. SCLC cells with RB1 protein expression were sensitive to growth inhibition by the CDK4/6 inhibitor palbociclib, and a patient with wild-type RB1 SCLC showed a significant decrease in mutant TP53 ctDNA allelic fraction and tumor mass after treatment with the CDK4/6 inhibitor abemaciclib.

Reactive-diffusion epidemic model on human mobility networks: Analysis and applications to COVID-19 in China

Summary: This study develops a reactive-diffusion epidemic model on human mobility networks to characterize the spatio-temporal propagation of COVID-19 in China. A novel time-dependent function is incorporated to describe the effects of human intervention. The results indicate that human intervention significantly inhibits the correlation between population mobility and infection cases, and spatial diffusion phenomenon of cases from high infection density cities to low infection density cities is revealed.

Model-Based Reinforcement Learning Control of Electrohydraulic Position Servo Systems

A Novel Coupled Code Shifted M-ary Differential Chaos Shift Keying Modulation

Summary: In this paper, a novel coupled code shifted M-ary differential chaos shift keying modulation scheme is proposed for reference removal. The scheme transmits only two information-bearing signals, modulated by selected Walsh code sequences and coupled by the same chaotic message bearer. Theoretical BER expressions are derived and simulations show better performance with lower complexity.

Surface engineering: Phosphorus doped Co3(PO4)2 generates P(6-)-Co bonds for photocatalytic hydrogen evolution

Summary: This paper studies the doping of phosphorus elements in Co3(PO4)2 during the high-temperature phosphorylation reaction. UV-Vis experiments show that the color of the phosphorylation catalyst changes from pink to black-purple, with significantly increased light absorption range and intensity. SEM images reveal the growth of many nanoparticles on the catalyst surface. XPS experiments demonstrate the growth of a stable P (6- )-Co bond on the catalyst surface. The P (6- )-Co bond can inhibit the complexation of photogenerated electron and hole pairs and enhance the transport efficiency of photogenerated electrons, thus improving the hydrogen production performance. Phosphorus-doped Co3(PO4)2 releases three times more hydrogen than CO3(PO4)2. This paper provides a novel design solution for the photocatalyst preparation method.

Slip-Line Solution for the Active Earth Pressure of Narrow and Layered Backfills against Inverted T-Type Retaining Walls Rotating about the Base

Summary: In this research, a slip-line method and adaptive finite-element software were used to investigate the active earth pressure of narrow and layered backfills against inverted T-type retaining walls rotating around the base mode. Slip-line field calculation models were established for the inverted T-type retaining wall with narrow and layered backfills, considering the effect of the heel bottom plate and the wall characteristics. The proposed method was verified by comparing its results with those from finite-element limit analysis and existing theoretical solutions, and parametric studies were conducted to study the effects of backfill width, heel length, and interface strength.

Observability of Discrete-Time Two-Time-Scale Multi-Agent Systems with Heterogeneous Features under Leader-Based Architecture

Summary: This paper investigates the observability of discrete-time two-time-scale multi-agent systems with heterogeneous features under leader-follower architecture. It establishes a singular perturbation difference model for the discussed system based on consensus agreement. To eliminate the numerical ill-posed problem, the order of the system model is reduced using the boundary layer theory of the singular perturbation system. Based on matrix theory, algebraic and graphical features that guarantee system observability are obtained. The theoretical results are verified by a numerical example.
