Summary: Inflammatory Bowel Disease (IBD) is a chronic autoimmune disease characterized by intestinal inflammation. The incidence of IBD is rapidly increasing in newly developed countries, making it a global concern. Two major types of IBD, ulcerative colitis (UC) and Crohn's disease (CD), have different manifestations and locations of inflammation in the digestive tract. Current FDA-approved treatments for IBD are only effective for 50% of patients and come with serious side effects, highlighting the need for safer and more effective therapies. Plant-based diets, such as the Mediterranean diet rich in anti-inflammatory phytochemicals, have shown benefits in preventing and improving symptoms of IBD. Terpenes, a class of phytochemicals abundant in Mediterranean herbs and citrus fruits, have been studied for their potential to modulate inflammatory signaling in IBD. However, there is still conflicting and inconclusive evidence regarding their effectiveness. This review evaluates preclinical studies on terpenes and their impact on basic inflammatory signaling in IBD.

It predominantly exhibits anti-inflammatory effects, especially in ulcerative colitis, arthritis, sepsis, and endotoxemia, but may promote and aggravate certain diseases such as periodontitis and gingivitis in oral inflammation. The molecular mechanism of nicotine in these inflammatory diseases involves regulating immune cells, immune factors, and the vagus and acetylcholinergic anti-inflammatory pathways, offering potential value for remedying acute and chronic inflammation in humans.

Summary: Two undescribed polyoxygenated seco-cyclohexene derivatives named macclureins A and B, and three undescribed polyoxygenated cyclohexene derivatives macclureins C-E, together with 15 known analogues were isolated from the twigs and leaves of Uvaria macclurei. (-)-Zeylenone showed the most potent anti-inflammatory effect against NO production and downregulated NF-kappa B signaling pathway. In vivo experiments using a mouse model of sepsis demonstrated that (-)-zeylenone significantly alleviated sepsis severity by measuring weight, murine sepsis score, survival rate and the serum levels of pro-inflammatory factors TNF-alpha and IL-6.

Summary: In this study, 26 derivatives of Coixol were designed and synthesized to discover new anti-inflammatory agents. The derivatives containing furan or nitrofuran moieties exhibited stronger anti-inflammatory activity than Coixol and celecoxib, as evidenced by the reduced expression of iNOS, TNF-α, IL-6, and IL-1β. Mechanistic investigations revealed that the inhibition of the NF-κB signaling pathway contributed to the anti-inflammatory effects of 9c and 9j. In vivo studies confirmed their anti-inflammatory activity in a xylene-induced mice auricles edema model.
