Synthesis, characterization, DNA interaction studies and anticancer activity of platinum-clotrimazole complexes

New trans-platinum complexes of clotrimazole (CTZ) have been prepared and characterized. Their interaction with DNA and activity against tumour cell lines were evaluated. [Pt (CTZ)(2)I-2] (1) was synthesized by the reaction of K2PtCl4, KI and CTZ, and [Pt(CTZ)(2)Cl-2] (2) by direct reaction of K2PtCl4 with CTZ. These complexes were characterized by a combination of elemental analysis, molar conductivity, UV-Vis, IR, and NMR spectroscopy in one and two dimensions. DNA-platinum complex interactions were studied by spectroscopic, thermal denaturation and viscosity titration measurements. Covalent interaction studies were also performed. From these results we suggest that complexes 1 and 2 interact with the minor groove of DNA. Both complexes showed growth inhibitory effects on four out of the six tumour cells lines with GI(50) (50% growth inhibition) values in the 5-25 mu M range, but there was no indication of cytotoxicity over the range of concentrations tested.

Zinc(II)-Sterol Hydrazone Complex as a Potent Anti-Leishmania Agent: Synthesis, Characterization, and Insight into Its Mechanism of Antiparasitic Action

Summary: In this study, a new complex called ZnCl2(H3)(2) was synthesized and evaluated for its efficacy against Leishmania amazonensis. The results showed that this complex exhibited stronger inhibitory effects on the parasite compared to the free ligand H3. Furthermore, it demonstrated higher selectivity. Additionally, the study revealed that H3 could induce depletion of endogenous parasite sterols and their replacement by 24-desalkyl sterols, and the zinc derivative of H3 resulted in a loss of cell viability.

Synthesis, characterization and biological evaluation of zinc and copper azasterol complexes against Sporothrix brasiliensis

Summary: New copper(I), copper(II), and zinc(II) azasterol derivatives were synthesized using 20-hydrazone-imidazolin-2-yl-5 alpha-pregnan-3 beta-ol (H1) as a ligand. The metal complexes were fully characterized and their molecular structures were investigated using computational methods. In vitro antifungal tests showed that the metal-azasterol complexes were more effective against S. brasiliensis than the ligand H1, with [Zn(H1)(2)(OH)(2)] demonstrating the highest efficiency. These results suggest the potential of metal-azasterol complexes in developing new antifungal compounds.
