Leukocyte transendothelial migration is controlled by chemokine-induced signalling in leukocytes and integrin-ligand-induced bidirectional signalling in both leukocytes and endothelial cells. It is now generally accepted that endothelial signalling following leukocyte adhesion, serves to facilitate the crossing of the endothelium, be it via the paracellular or transcellular route. This brief overview discusses the main findings within this area and highlights some recent findings that shed new light on adhesion-induced signalling in the context of leukocyte transendothelial migration. ADAM10-Mediated Cleavage of ICAM-1 Is Involved in Neutrophil Transendothelial Migration

Summary: The study reveals that endothelial ADAM10 plays a role in facilitating neutrophil transendothelial migration by cleaving ICAM-1, supporting the efficient release of neutrophils from the endothelium. Silencing or inhibiting endothelial ADAM10 leads to a significantly longer diapedesis step for neutrophils due to impaired dissociation from ICAM-1. Increased ICAM-1 levels on transmigrating neutrophils and potential membrane transfer from endothelium to neutrophils are also observed.

Summary: The kinase Arg/Abl2 has been identified as a mediator of endothelial barrier disruption. Depletion of Arg in endothelial cells resulted in activation of RhoA and Rac1, increased cell spreading and elongation, and redistribution of integrin-dependent cell-matrix adhesions. In vivo, genetic deletion of Arg diminished vascular leak in the skin and lungs, suggesting therapeutic inhibition of Arg may be a feasible strategy for treating conditions characterized by vascular leak.

Factors like chemokine cues, adhesion molecules, membrane protrusions, and physical factors contribute to the formation of transmigration hotspots where leukocytes exit the vasculature.

The increased expression of nuclear factor (NF)-kappa B2 and epigenetic modifications of the vWF promoter play a critical role in driving vWF transcription and promoting a prothrombotic phenotype in CTEPH-pulmonary artery endothelial cells.

Bone Marrow Harbors a Unique Population of Dendritic Cells with the Potential to Boost Neutrophil Formation upon Exposure to Fungal Antigen

Summary: Apart from controlling hematopoiesis, the bone marrow also functions as a secondary lymphoid organ. Bone marrow-resident dendritic cells (DCs) are mainly localized around sinusoids and can present antigens. They belong to the cDC2 subtype and are associated with Th2/Th17 immunity. These DCs express several c-type lectins, including Dectin-1, which can bind fungal beta-glucans. Compared to spleen DCs, bone marrow DCs are more efficient in phagocytosing yeast particles and Aspergillus conidia, and this is dependent on Dectin-1. Additionally, zymosan-stimulated bone marrow DCs enhance neutrophil differentiation and maintenance of progenitor cells.

Inhibition of E1 ubiquitin ligases leads to a rapid, reversible loss of integrity in quiescent endothelial monolayers with increased F-actin stress fibers and intercellular gaps. Fast protein turnover of negative regulators of cell-cell contact is essential for maintaining monolayer integrity in quiescent human endothelial cells.

The results show that by precisely controlling the activation of Rho GTPases, the strength of the vascular barrier can be precisely regulated in a VE-cadherin-independent manner through cell-cell overlap-dependent mechanism. Furthermore, optogenetic control over morphological features of endothelial cells, such as cell size and shape, is also demonstrated.
