Sna3 Is an Rsp5 Adaptor Protein that Relies on Ubiquitination for Its MVB Sorting

The process in which ubiquitin (Ub) conjugation is required for trafficking of integral membrane proteins into multivesicular bodies (MVBs) and eventual degradation in the lumen of lysosomes/vacuoles is well defined. However, Ub-independent pathways into MVBs are less understood. To better understand this process, we have further characterized the membrane protein Sna3, the prototypical Ub-independent cargo protein sorted through the MVB pathway in yeast. We show that Sna3 trafficking to the vacuole is critically dependent on Rsp5 ligase activity and ubiquitination. We find Sna3 undergoes Ub-dependent MVB sorting by either becoming ubiquitinated itself or associating with other ubiquitinated membrane protein substrates. In addition, our functional studies support a role for Sna3 as an adaptor protein that recruits Rsp5 to cargo such as the methionine transporter Mup1, resulting in efficient Mup1 delivery to the vacuole.

Summary: Among patients with acute hypoxemic respiratory failure due to COVID-19, an initial strategy of CPAP significantly reduced the risk of tracheal intubation or mortality compared with conventional oxygen therapy, but there was no significant difference between an initial strategy of HFNO compared with conventional oxygen therapy. The study may have been underpowered for the comparison of HFNO vs conventional oxygen therapy, and early study termination and crossover among the groups should be considered when interpreting the findings.

Fabry-Perot cavities and quantum dot formation at gate-defined interfaces in twisted double bilayer graphene

Summary: The rich and tunable phase diagram exhibited by moire materials has made them an ideal platform for hosting versatile devices. Understanding electronic transport and localization across electrically defined interfaces is of fundamental importance for engineering such devices. In this study, a cavity is electrostatically defined across a twisted double bilayer graphene sample, revealing Fabry-Perot oscillations and Coulomb blockade resonances indicative of strong electronic confinement. Quantum dots are observed at the interfaces of the cavity, providing a first step towards better understanding interfacial phenomena in single crystal moire devices.

The Effect of Lithium on the Budding Yeast Saccharomyces cerevisiae upon Stress Adaptation

Summary: Lithium has been shown to prolong life span in various species, but not yet in budding yeast. In this study, we investigate the influence of lithium on yeast cells' viability and find that it can extend long-term survival under glucose deprivation. We also observe that lithium affects the cellular adaptation to oxidative stress and acute glucose starvation. These findings provide novel insights into the effects of lithium on eukaryotic cells.
