Summary: The cytotoxic potential of lichens from the Western Ghats in India was evaluated in this study. It was found that the acetone extract of Usnea cornuta (UC2A) showed significant cytotoxicity against multiple human cancer cell lines. Furthermore, the dominant cytotoxic compounds in the UC2A extract were identified as usnic acid (UA), atraric acid, and barbatic acid. Inhibition of autophagy was found to enhance the cytotoxicity of UA.

Summary: Pyrazole derivatives, a class of heterocyclic compounds, possess unique chemical structures and exhibit a broad spectrum of pharmacological activities. Recent studies have focused on synthesizing and evaluating pyrazole derivatives for their anticancer potential. Structure-activity relationship studies have shown that appropriate substitution on the pyrazole ring can enhance anticancer efficacy and selectivity. Furthermore, these derivatives have demonstrated multiple mechanisms of anticancer action, making them promising candidates for developing anticancer drugs.

Summary: Polyphenols extracted from pomegranate peel were purified and evaluated for their cytotoxic effects on Hela and NIH-3T3 cell lines. Punicalagin and ellagic acid (EA) isolated from pomegranate peel were found to regulate apoptosis in Hela and NIH-3T3 cells, with concentration and exposure time playing a role in determining the cytotoxic effect. Results from a commercial assay kit suggest that punicalin and EA may regulate apoptosis in these cell lines by altering protein expression levels.

Summary: Ruthenium polypyridine complexes containing 5-fluorouracil derivatives as ligands were synthesized and their anticancer activity was studied. Results showed that one of the complexes, Ru3, exhibited the best anticancer activity among the compounds tested, with selective activity. Mechanism studies revealed that Ru3 could specifically target lysosomes, induce cell apoptosis, arrest cell cycle, increase ROS level, and damage DNA, leading to cell death through the synergy of inducing apoptosis and autophagy.

Summary: Phosphodiesterases are important regulators of cyclic nucleotide levels, exerting control over cAMP and cGMP hydrolysis. They play a critical role in regulating signaling pathways and downstream biological effects, including gene expression, cell proliferation, and inflammation. Mutations in PDE genes have been linked to genetic diseases and PDEs may be involved in tumor development, particularly in cAMP-sensitive tissues. This review summarizes current knowledge on PDE expression and regulation in the testis, with a focus on their role in testicular cancer development.

Summary: Traumatic spinal cord injury (SCI) is characterized by severe neuroinflammation and hampered neuroregeneration. Current therapies have limited effectiveness, so new strategies targeting the initial inflammatory reactions and promoting endogenous repair are crucial. Adenosine monophosphate (cAMP) regulates these processes and inhibiting its hydrolyzing enzyme phosphodiesterase (PDE) has shown promise in modulating inflammation and promoting regeneration. This review focuses on the immunomodulatory and neuroregenerative role of cAMP-specific PDE inhibition in SCI.
