Effects of static magnetic field and cadmium on oxidative stress and DNA damage in rat cortex brain and hippocampus

The present study was undertaken to determine the effect of co-exposure to static magnetic field (SMF) and cadmium (Cd) on the antioxidant enzymes activity and DNA integrity in rat brain. Sub-chronic exposure to CdCl (CdCl2, 40 mg/L, per os) for 30 days resulted in a significant reduction in antioxidant enzyme activity such as the glutathione peroxidase (GPx), catalase (CAT) and superoxide dismutase (SOD) in frontal cortex and hippocampus. Total GSH were decreased in the frontal cortex of the Cd-exposed group. Cd exposure induced an increase in malondialdehyde (MDA) concentration in the frontal cortex and hippocampus. Moreover, the same exposure increased 8-oxo-7,8-dihydro-2'-desoxyguanosine (8-oxodGuo) level in rat brain. Interestingly, the combined effect of SMF (128 mT, 1 hour/day for 30 consecutive days) and CdCl (40 mg/L, per os) decreased the SOD activity and glutathione level in frontal cortex as compared with the Cd group. Moreover, the association between SMF and Cd increased MDA concentration in frontal cortex as compared with Cd-exposed rats. DNA analysis revealed that SMF exposure failed to alter 8-oxodGuo concentration in Cd-exposed rats. Our data showed that Cd exposure altered the antioxidant enzymes activity and induced oxidative DNA lesions in rat brain. The combined effect of SMF and Cd increased oxidative damage in rat brain as compared with Cd-exposed rats.

Summary: This study aimed to identify new biomarkers of exposure to sulfur mustard (SM) and validate their detectability in various models and body fluids. The results demonstrated that N7Gua-CEES, glutathione, and cysteine conjugates of 2-chloroethyl ethyl sulfide (CEES) were easily quantifiable in different models and plasma, showing promising potential for application in toxicological studies related to SM.

Summary: Sunlight can generate three types of DNA photolesions, with differences in efficiency between summer and winter attributed to seasonal variations in the UVA/UVB ratio. The conversion of 6-4PPs to DewarPPs is more efficient in winter than in late summer, indicating a significant influence of seasonal UVA/UVB ratio on photoisomerization.

Summary: Photobiological effects greatly depend on incident photon wavelength, and the effects of complex light sources cannot simply be summed up as monochromatic exposures. Similar to nonadditivity effects in chemical toxicology, photobiological processes can exhibit additive, antagonistic, and synergistic effects. The combination index can be applied to study the nonadditive effects in photobiological processes, but further research is needed to evaluate the error in its determination.

DNA photoproducts released by repair in biological fluids as biomarkers of the genotoxicity of UV radiation

Summary: UV-induced formation of photoproducts in DNA is a major initiating event of skin cancer, and many analytical tools have been developed for their quantification. In this study, a liquid chromatography-mass spectrometry method was extended to quantify short DNA fragments containing photoproducts released from cells. The method was validated with a robust protocol and showed high sensitivity and specificity. Proof of concept for the use of excreted DNA repair products as biomarkers of UV genotoxicity was obtained through in vitro and ex vivo studies.

Identification of 2-Methylthio-methylenethio-N6-(cis-4-hydroxyisopentenyl)adenosine (msms2io6A37) as a Novel Modification at Adenosine 37 of tRNAs from Salmonella typhimurium

Summary: Post-transcriptional modifications of tRNA nucleotide play a crucial role in determining their folding, structure, and function. We have discovered and characterized a novel modified base called 2-methylthio-methylenethio-N-6-(cis-4-hydroxyisopentenyl)adenosine, which is found at position 37 in specific tRNAs. Furthermore, we have identified the catalytic cycle of the tRNA-diiron monooxygenase, MiaE, in Salmonella typhimurium, which converts the known 2-methylthio-methylenethio-N-6-(isopentenyl)adenosine nucleoside into this newly identified modified adenosine.

Summary: In this study, it is experimentally demonstrated that the presence of furanose in a southern conformation in oligonucleotides is necessary for the formation of (6-4) photoproduct. The location of the southern sugar conformation, particularly at the 5'-end, is more favorable for the formation of the (6-4) adduct in thymine dinucleotide. The flexibility of the southern pucker at the 3'-end has little influence on the formation of the (6-4) adduct.

Summary: Silver nanoparticles are commonly used as biocides, but studies have shown that acute exposure to these nanoparticles can have a stronger impact on DNA integrity compared to repeated exposure. Both exposure regimens were found to alter cellular metabolism, induce intracellular ROS accumulation, and disrupt cell cycle progression, indicating detrimental effects on cells.
