The mechanism by which mice, exposed to the cold, mobilize endogenous or exogenous fuel sources for heat production is unknown. To address this issue we carried out experiments using 3 models of obesity in mice: C57BL/6J+/+ (wild-type B6) mice with variable susceptibility to obesity in response to being fed a high-fat diet (HFD), B6. Ucp1(-/-) mice with variable diet-induced obesity (DIO) and a deficiency in brown fat thermogenesis and B6. Lep(-/-) with defects in thermogenesis, fat mobilization and hyperphagia. Mice were exposed to the cold and monitored for changes in food intake and body composition to determine their energy balance phenotype. Upon cold exposure wild-type B6 and Ucp1(-/-) mice with diet-induced obesity burned endogenous fat in direct proportion to their fat reserves and changes in food intake were inversely related to fat mass, whereas leptin-deficient and lean wild-type B6 mice fed a chow diet depended on increased food intake to fuel thermogenesis. Analysis of gene expression in the hypothalamus to uncover a central regulatory mechanism revealed suppression of the Npvf gene in a manner that depends on the reduced ambient temperature and degree of exposure to the cold, but not on adiposity, leptin levels, food intake or functional brown fat.

Uncoupling protein-1 expression does not protect mice from diet-induced obesity

Hui Wang, Monja Willershaeuser, Yongguo Li, Tobias Fromme, Katharina Schnabl, Andrea Bast-Habersbrunner, Samira Ramisch, Sabine Mocek, Martin Klingenspor

Summary: While UCP1 is essential for cold-induced thermogenesis, its presence alone does not protect mice from diet-induced obesity. A new mouse model with low UCP1 levels demonstrates that impaired cold-induced thermogenic capacity does not increase susceptibility to diet-induced obesity. Pharmacological activation of brown fat is necessary to promote negative energy balance in diet-induced obese mice.

The Influence of Intravaginal Gestagens Treatment on the Morphological Features and Endometrial Steroid Hormone Receptors Content during Anestrus Type II in Dairy Cattle

Summary: This study evaluated the effectiveness of vaginal progesterone inserts therapy in cows with anestrus type II. The expression of progesterone and estrogen receptors in the endometrium was assessed, and blood levels of estradiol and progesterone were evaluated. The results showed a decrease in mRNA expression of certain receptors in both treated and untreated animals. However, treated animals showed an increase in the expression of specific receptors compared to untreated animals. In addition, the recurrence of ovarian cyclicity and pregnancy rates were higher in the treated group.

Summary: Negative energy balance induced through either lower ambient temperature or exercise resulted in substantial and similar improvements in markers of inflammation and mitochondrial damage in the hypothalamus of mice with diet-induced obesity, potentially by reducing oxidative stress.

miR-26a-5p and miR-125b-5p affect trophoblast genes and cell functions important during early pregnancy†

Joanna Szuszkiewicz, Anna Nitkiewicz, Klaudia Drzewiecka, Monika M. Kaczmarek

Summary: Embryo implantation is the most critical stage of pregnancy, dependent on the synchronized developmental capacity of the embryo and uterine receptivity. In early pregnancy, both the conceptus and uterus release various factors to facilitate successful implantation and placenta formation. MicroRNAs, which regulate gene expression, are considered important regulators of early pregnancy events.

When fat meets the gut-focus on intestinal lipid handling in metabolic health and disease

Magdalena Wit, Jonathan Trujillo-Viera, Akim Strohmeyer, Martin Klingenspor, Mohammed Hankir, Grzegorz Sumara

Summary: Overconsumption of energy-dense foods can lead to increased intestinal nutrient absorption and excessive lipid accumulation, resulting in obesity and obesity-associated diseases. Targeting the pathways that regulate intestinal nutrient absorption holds significant therapeutic potential.

Summary: Proteomics was used to study non-small cell lung cancer and investigate the differences in proteins between the tumor center and tumor margin. Several important proteins related to lung cancer development and molecular specificity were identified, which could potentially serve as biomarkers and targets for anticancer therapy.

EBI2 is a negative modulator of brown adipose tissue energy expenditure in mice and human brown adipocytes

Francesca Copperi, Inna Schleis, Martin Roumain, Giulio G. Muccioli, Stefano Casola, Martin Klingenspor, Alexander Pfeifer, Thorsten Gnad

Summary: The study identifies GPR183/EBI2 as the most highly expressed inhibitory G-protein-coupled receptor in brown adipose tissue, which decreases energy expenditure. Loss of Ebi2 in mice results in increased brown adipocyte energy expenditure in response to cold exposure, providing potential insights into modulating energy expenditure in humans.

Early steps of embryo implantation are regulated by exchange of extracellular vesicles between the embryo and the endometrium

Joanna Szuszkiewicz, Kamil Myszczynski, Zaneta P. Reliszko, Yael Heifetz, Monika M. Kaczmarek

Summary: Research has shown that during early pregnancy, communication between the embryo and uterus occurs through the exchange of extracellular vesicles carrying miRNAs. These vesicles impact embryonic development and trophoblast cell function and interactions, playing a crucial role in successful embryo implantation and pregnancy.

Extracellular vesicles: Focus on peri-implantation period of pregnancy in pigs

Summary: The establishment of cell-to-cell communication is crucial for successful mammalian pregnancy. Extracellular vesicles (EVs) carrying unique molecular cargo have been identified as mediators of this communication during early pregnancy. However, the mechanism of EV-mediated crosstalk between early embryos and the endometrium needs further investigation. Understanding the role of EVs in mammalian reproduction can greatly enhance our knowledge of successful reproductive performance.

WNT4 Gene and Protein Expression in Endometrial Cancer and Its Significance

Jolanta Kiewisz, Tomasz Wasniewski, Jacek Kiezun, Agnieszka Skowronska, Monika M. Kaczmarek, Blazej Szostak, Anna E. Kowalczyk, Zbigniew Kmiec

Summary: This study aimed to evaluate the alterations of WNT4 gene expression and protein immunoreactivity in endometrial cancer (EC) and their association with tumor characteristics, clinicopathological factors, and estrogen dependence. The results showed that WNT4 gene expression was decreased in EC samples, particularly in endometrioid EC, and was associated with tumor characteristics. However, there was no statistical difference in WNT4 protein immunoreactivity or estrogen receptor alpha immunoreactivity between EC and control group. The study also found that alterations in WNT4 gene expression tended to be mutually exclusive with alterations in ESR1 gene expression. Overall, this study highlights the importance of evaluating the molecular background of EC, and suggests that WNT4 gene may be a marker of functional estrogen signaling in endometrioid EC.
