Influence of CLOCK on cytotoxicity induced by diethylnitrosamine in mouse primary hepatocytes

The Clock gene is a core clock factor that plays an essential role in generating circadian rhythms. In the present study, it was investigated whether the Clock gene affects the response to diethylnitrosamine (DEN)-induced cytotoxicity using mouse primary hepatocytes. DEN-induced cytotoxicity, after 24 h exposure, was caused by apoptosis in hepatocytes isolated from wild-type mouse. On the other hand, Clock mutant mouse (Clk/Clk) hepatocytes showed resistance to apoptosis. Because apoptosis is an important pathway for suppressing carcinogenesis after genomic DNA damage, the mechanisms that underlie resistance to DEN-induced apoptosis were examined in Clk/Clk mouse hepatocytes. The mRNA levels of metabolic enzymes bioactivating DEN and apoptosis-inducing factors before DEN exposure were lower in Clk/Clk cells than in wild-type cells. The accumulation of p53 and Ser15 phosphorylated p53 after 8 h DEN exposure was seen in wild-type cells but not in Clk/Clk cells. Caspase-3/7 activity was elevated during 24 h DEN exposure in wild-type cells but not in Clk/Clk cells. In addition, resistance to DEN-induced apoptosis in Clk/Clk cells affected the cell viability. These studies suggested that the lower expression levels of metabolic enzymes bioactivating DEN and apoptosis inducing factors affected the resistance to DEN-induced apoptosis in Clk/Clk cells, and the Clock gene plays an important role in cytotoxicity induced by DEN. Sulfasalazine alleviates neuropathic pain hypersensitivity in mice through inhibition of SGK-1 in the spinal cord

Summary: Diurnal variations in pain hypersensitivity are common in chronic pain disorders, with exacerbation of neuropathic pain hypersensitivity depending on glucocorticoid secretion. The study found that the drug SSZ has inhibitory effects on SGK-1 in the spinal cord, and concomitant administration with FBX can enhance its anti-neuropathic pain effects.

Summary: The study revealed that ADAR1-mediated regulation of alternative splicing of the ABCB1 gene is a key mechanism for the circadian expression of P-glycoprotein (P-gp) in human renal proximal tubular epithelial cells (RPTECs). This regulatory mechanism may explain the dosing time-dependent variations in the renal elimination of P-gp substrates.

The monocytic expression of GPR68, induced by altered CLOCK activation, exacerbates heart inflammation and fibrosis under CKD conditions.

Summary: Cancer cells have developed ways to evade the host immune system, including through the PD-1/PD-L1 immune checkpoint pathway. This study found that the expression of PD-1 on tumor-associated macrophages (TAMs) fluctuates in a diurnal manner and is regulated by a circadian regulatory component called DEC2. Furthermore, administering PD-1/PD-L1 inhibitors at the time of day when PD-1 expression on TAMs is increased enhances their anti-tumor efficacy.

Senescence-induced alteration of circadian phagocytic activity of retinal pigment epithelium cell line ARPE-19

Summary: Renewal of retinal photoreceptor outer segments involves shedding and phagocytosis, but senescence can modulate the circadian phagocytic activity of RPE cells. The phagocytic activity of senescent ARPE-19 cells increased constantly and exhibited altered circadian oscillation, accompanied by changes in the expression of circadian clock genes. Additionally, activation of the circadian clock component REV-ERBa enhanced the phagocytic activity of normal ARPE-19 cells and increased the expression of phagocytosis-related genes.

Oral administration of vancomycin alleviates heart failure triggered by chronic kidney disease

Summary: Chronic kidney disease (CKD) disrupts the intestinal microbiota, leading to cardiovascular inflammation and fibrosis. In this study, a mouse model was used to explore the effects of administering vancomycin (VCM) on the microbiota and cardiac pathology associated with CKD. Despite advanced renal dysfunction, VCM-induced changes in the intestinal microbiota composition reduced cardiotoxic substances, alleviated heart failure, and improved survival rates. These findings highlight the potential of targeting the intestinal microbiota to prevent and treat cardiovascular conditions in CKD.
